Impact of anti-FAP CAR-T cells on fibroblasts in a mouse model of Duchenne Muscular Dystrophy evaluated by single cell RNA-sequencing


ABSTRACT: Cardiac fibrosis is a known complication of Duchenne Muscular Dystrophy (DMD). In this project, we aimed to analyze the impact of Chimeric Antigen Receptor (CAR)-T cells directed against Fibroblast Activation Protein (FAP), a protein expressed by activated fibroblasts, on cardiac fibroblasts in a mouse model of DMD (D2.mdx mice). To this end, we performed single-cell RNA-sequencing analysis of total viable non-cardiomyocyte cardiac cells in DBA2/J mice (healthy controls, n=2), dystrophic D2.mdx mice treated with GFP-transduced T lymphocytes (n=5) and dystrophic D2.mdx mice treated with GFP anti-FAP CAR-T cells (n=5). Samples were pooled using cell hashing and single cells were captured using the 10x Genomics V4 Chromium technology.
