Project description:Perceptual learning refers to experience-induced improvements in the pick-up of information. Perceptual constancy describes the fact that, despite variable sensory input, perceptual representations typically correspond to stable properties of objects. Here, we show evidence of a strong link between perceptual learning and perceptual constancy: Perceptual learning depends on constancy-based perceptual representations. Perceptual learning may involve changes in early sensory analyzers, but such changes may in general be constrained by categorical distinctions among the high-level perceptual representations to which they contribute. Using established relations of perceptual constancy and sensory inputs, we tested the ability to discover regularities in tasks that dissociated perceptual and sensory invariants. We found that human subjects could learn to classify based on a perceptual invariant that depended on an underlying sensory invariant but could not learn the identical sensory invariant when it did not correlate with a perceptual invariant. These results suggest that constancy-based representations, known to be important for thought and action, also guide learning and plasticity.

Project description:Extracellular vesicles (EVs) are membrane-bound particles released by cells in various (patho)physiological conditions. EVs can transfer effector molecules and elicit potent responses in recipient cells, making them attractive therapeutic agents and drug delivery platforms. In contrast to their tremendous potential, only a few EV-based therapies and drug delivery have been approved for clinical use, which is largely attributed to limited therapeutic loading technologies and efficiency. As EV cargo has major influence on their functionality, understanding and translating the biology underlying the packaging and transferring of biomolecule cargos (e.g. miRNAs, pathogen antigens, small molecule drugs) into EVs is key in harnessing their therapeutic potential. In this review, through recent insights into EVs' content packaging, we discuss different mechanisms utilized by EVs during cargo packaging, and how one might therapeutically exploit this process. Apart from the well-characterized EVs like exosomes and microvesicles, we also cover the less-studied and other EV subtypes like apoptotic bodies, large oncosomes, bacterial outer membrane vesicles, and migrasomes to highlight therapeutically-diverse opportunities of EV armoury.

Project description:Life in a multisensory world requires the rapid and accurate integration of stimuli across the different senses. In this process, the temporal relationship between stimuli is critical in determining which stimuli share a common origin. Numerous studies have described a multisensory temporal binding window-the time window within which audiovisual stimuli are likely to be perceptually bound. In addition to characterizing this window's size, recent work has shown it to be malleable, with the capacity for substantial narrowing following perceptual training. However, the generalization of these effects to other measures of perception is not known. This question was examined by characterizing the ability of training on a simultaneity judgment task to influence perception of the temporally-dependent sound-induced flash illusion (SIFI). Results do not demonstrate a change in performance on the SIFI itself following training. However, data do show an improved ability to discriminate rapidly-presented two-flash control conditions following training. Effects were specific to training and scaled with the degree of temporal window narrowing exhibited. Results do not support generalization of multisensory perceptual learning to other multisensory tasks. However, results do show that training results in improvements in visual temporal acuity, suggesting a generalization effect of multisensory training on unisensory abilities.

Project description:Developing expertise in any field usually requires acquisition of a wide range of skills. Most current studies on perceptual learning have focused on a single task and concluded that learning is quite specific to the trained task, and the ubiquitous individual differences reflect random fluctuations across subjects. Whether there exists a general learning ability that determines individual learning performance across multiple tasks remains largely unknown. In a large-scale perceptual learning study with a wide range of training tasks, we found that initial performance, task, and individual differences all contributed significantly to the learning rates across the tasks. Most importantly, we were able to extract both a task-specific but subject-invariant component of learning, that accounted for 38.6% of the variance, and a subject-specific but task-invariant perceptual learning ability, that accounted for 36.8% of the variance. The existence of a general perceptual learning ability across multiple tasks suggests that individual differences in perceptual learning are not "noise"; rather, they reflect the variability of learning ability across individuals. These results could have important implications for selecting potential trainees in occupations that require perceptual expertise and designing better training protocols to improve the efficiency of clinical rehabilitation.

Project description:Perceptual learning operates on distinct timescales. How different neuromodulatory systems impact on learning across these different timescales is poorly understood.Here, we test the causal impact of a novel influence on perceptual learning, the androgen hormone testosterone, across distinct timescales.In a double-blind, placebo- controlled, cross-over study with testosterone, subjects undertook a simple contrast detection task during training sessions on two separate days.On placebo, there was no learning either within training sessions or between days, except for a fast, rapidly saturating, improvement early on each testing day. However, testosterone caused "off-line" learning, with no learning seen within training sessions, but a marked performance improvement over the days between sessions. This testosterone-induced learning occurred in the absence of changes in subjective confidence or introspective accuracy.Our findings show that testosterone influences perceptual learning on a timescale consistent with an influence on "off-line" consolidation processes.

Project description:UnlabelledIn this work we present an approach to understand neuronal mechanisms underlying perceptual learning. Experimental results achieved with stimulus patterns of coherently moving dots are considered to build a simple neuronal model. The design of the model is made transparent and underlying behavioral assumptions made explicit. The key aspect of the suggested neuronal model is the learning algorithm used: We evaluated an implementation of Hebbian learning and are thus able to provide a straight-forward model capable to explain the neuronal dynamics underlying perceptual learning. Moreover, the simulation results suggest a very simple explanation for the aspect of "sub-threshold" learning (Watanabe et al. in Nature 413:844-884, 2001) as well as the relearning of motion discrimination after damage to primary visual cortex as recently reported (Huxlin et al. in J Neurosci 29:3981-3991, 2009) and at least indicate that perceptual learning might only occur when accompanied by conscious percepts.Electronic supplementary materialThe online version of this article (doi:10.1007/s11571-010-9134-9) contains supplementary material, which is available to authorized users.

Project description:Glioblastoma multiforme, one of the most common and aggressive brain tumors in adults, is highly resistant to currently available therapies and often recurs. Due to its poor prognosis and difficult management, there is an urgent need for the development and translation of new anti-glioma therapeutic approaches into the clinic. In this context, oncolytic virotherapy arises as an exciting treatment option for glioma patients. These natural or genetically engineered viruses are able to effectively infect cancer cells, inducing a specific anti-tumor cytotoxic effect. In addition, some viruses have been redesigned to modulate glioma microenvironment, to express cytokines to boost a systemic anti-glioma immune response and to incorporate angiostatic genes to decrease glioma vasculature. Although recent clinical trials have confirmed the safety of oncolytic virotherapies in the brain, their moderate clinical efficacy has not yet matched the encouraging preclinical laboratory results. In this review, we will discuss the leading anti-glioma virotherapy approaches that are presently under preclinical and clinical evaluation. We will also review different delivery methods, in vivo virus behavior, fate, replication, intratumoral spread, activation of anti-tumor immune response, and targeting of glioma stem cells. We will focus on the advantages and limitations of each therapeutic approach and how to overcome these hurdles to effectively translate exciting laboratory results into promising clinical trials.

Project description:What determines how much an organism can learn? One possibility is that the neural factors that limit sensory performance prior to learning, place an upper limit on the amount of learning that can take place. We tested this idea by comparing learning on a sensory task where performance is limited by cortical mechanisms, at two retinal eccentricities. Prior to learning, visual performance at the two eccentricities was either unmatched or equated in two different ways (through spatial scaling or visual crowding). The magnitude of learning was equivalent when initial levels of performance were matched regardless of how performance was equated. The magnitude of learning was a constant proportion of initial performance. This Weber-like law for perceptual learning demonstrates that it should be possible to predict the degree of perceptual improvement and the final level of performance that can be achieved via sensory training, regardless of what cortical constraint limits performance.

Project description:Traditional perceptual learning protocols rely almost exclusively on long periods of uninterrupted fixation. Taking a first step towards understanding perceptual learning in natural vision, we had observers report the orientation of a briefly flashed stimulus (clockwise or counterclockwise from a reference orientation) presented strictly during saccade preparation at a location offset from the saccade target. For each observer, the saccade direction, stimulus location, and orientation remained the same throughout training. Subsequently, we assessed performance during fixation in three transfer sessions, either at the trained or at an untrained location, and either using an untrained (Experiment 1) or the trained (Experiment 2) stimulus orientation. We modeled the evolution of contrast thresholds (i.e., the stimulus contrast necessary to discriminate its orientation correctly 75% of the time) as an exponential learning curve, and quantified departures from this curve in transfer sessions using two new, complementary measures of transfer costs (i.e., performance decrements after the transition into the Transfer phase). We observed robust perceptual learning and associated transfer costs for untrained locations and orientations. We also assessed if spatial transfer costs were reduced for the remapped location of the pre-saccadic stimulus-the location the stimulus would have had (but never had) after the saccade. Although the pattern of results at that location differed somewhat from that at the control location, we found no clear evidence for perceptual learning at remapped locations. Using novel, model-based ways to assess learning and transfer costs, our results show that location and feature specificity, hallmarks of perceptual learning, subsist if the target stimulus is presented strictly during saccade preparation throughout training.

Project description:Studies of perceptual learning have revealed a great deal of plasticity in adult humans. In this study, we systematically investigated the effects and mechanisms of several forms (trial-by-trial, block, and session rewards) and levels (no, low, high, subliminal) of monetary reward on the rate, magnitude, and generalizability of perceptual learning. We found that high monetary reward can greatly promote the rate and boost the magnitude of learning and enhance performance in untrained spatial frequencies and eye without changing interocular, interlocation, and interdirection transfer indices. High reward per se made unique contributions to the enhanced learning through improved internal noise reduction. Furthermore, the effects of high reward on perceptual learning occurred in a range of perceptual tasks. The results may have major implications for the understanding of the nature of the learning rule in perceptual learning and for the use of reward to enhance perceptual learning in practical applications.
