ABSTRACT: Chromosome segregation is mediated by kinetochores, multi-protein structures that connect centromeric chromatin to spindle microtubules. Comparative genomics revealed a complex ancestral kinetochore and recurrent loss or replacement of subcomplexes across eukaryotic diversity. Understanding kinetochore composition and organization in diverse lineages reveals trajectories of kinetochore evolution and aids in dissecting subcomplex functions. The ciliate Tetrahymena thermophila has an elusive kinetochore composition. Here, we leverage proximity proteomics coupled to deep homology analysis to identify 16 kinetochore proteins in Tetrahymena, dubbed KiTTs (Kinetochore-of-Tetrahymena-thermophila 1-16). In addition to orthologs of NDC80, NUF2 and STU2 (KiTT1-2+14), we find that nine KiTTs (3-9 + 1
