Spatial ECM mapping defines cellular matrix environments associated with allergic airway pathology

ABSTRACT:

The extracellular matrix (ECM) forms the scaffold in which cells reside and interact guiding tissue function. Cellular organisation can be investigated with multiplexed imaging. However, the spatial relationship between cells and ECM has been broadly overlooked. Using imaging mass cytometry, we investigated cellular niches and their surrounding matrix environment during murine allergic airway pathology. We developed a novel pipeline Deepthresh, to characterise cellular ECM environments alongside cellular classifications. Applying this methodology, we revealed disparate and restricted responses during allergic lung pathology. Recruited neutrophils localised within the allergic alveolar parenchyma alongside altered alveolar type I versus type II cell ratios associated with increased proxi
