IgA Nephropathy GWAS (IGANGWAS)

ABSTRACT:

In this second study version, we report a genome-wide association study of IgA nephropathy performed in individuals of European ancestry (US and Italian cohorts), followed by a meta-analysis with the existing GWAS cohorts (Han Chinese [study version 1] and French cohorts). Genomic DNA was extracted from whole blood and genotyped with the HumanHap550-2v3, Human610-Quadv1and HumanOmni1-Quad_v1 platforms. This GWAS also utilized pre-genotyped population controls from other studies.

Fabry nephropathy transcriptome

Project description:Fabry nephropathy (FN) is a rare disorder caused by mutations in the alpha-galactosidase A gene. In this study we aim at providing a framework allowing selection of biomarkers and drug-targets. Two independent Fabry Nephropathy cohorts (FA.NO and CH.RO) were subjected to RNAseq from archival kidney biopsies taken prior and up to 10 years of Enzyme Replacement Therapy. Four compartments were laser capture microdissected (glomeruli, proximal tubuli, distal tubuli and arteries). We found several pathways that were consistently altered and that these kidney compartments’ transcriptional landscapes can be leveraged in the search for drug-targets and biomarkers.

Genome-wide association study identifies three novel susceptibility loci for systemic mastocytosis

Project description:To identify SNPs predisposing to systemic mastocytosis (SM) we performed a two stage GWAs consisting of two cohorts from the UK and Germany at stage 1 and three cohorts from Spain, Italy and Denmark at stage 2. At both stages, the SM cases were compared with publicly available controls. The control cohorts used were WTCCC and KORA at stage 1 and the Spanish National DNA bank, InChianti and Inter99 at stage 2. The data available are raw intensity files for 480 SM cases that were genotyped at stage 1. Please see our publication (Galata et al 2020) for details regarding QC of the stage 1 samples.
