Phase I Study to Evaluate Safety and Anti-tumor Activity of PB101, an Anti-angiogenic Immunomodulating Agent

ABSTRACT: This clinical trial is designed as a multi-center, open-label, dose-escalation, dose-expansion, phase 1 clinical trial and will be evaluating the safety and efficacy of PB101 in patients with advanced solid tumors who have progressed after standard of care. PB101 may stop the growth of tumor cells by blocking blood flow to the tumor and modulating the tumor microenvironment.

Interleukin-17 mediated paracrine network promote tumor resistance to anti-angiogenic therapy

Project description:While angiogenesis inhibitors have provided significant clinical benefit as cancer therapeutics, the mechanisms of anti-VEGF resistance remain incompletely understood. We uncovered an interleukin-17 mediated paracrine network of signaling between the adaptive and innate immune system associated with resistance to anti-VEGF treatment in multiple tumor models. The expression of tumor-associated fibroblasts and tumor-associated granulocytes (defined as CD11b+Gr1+) and tumor-associated monocytic cells (defined as CD11b+Gr1-) were compared between wildtype and IL-17RC knockout tumor bearing mice.

Tenascin-C promotes tumor angiogenesis through pro-angiogenic and anti-angiogenic effects involving YAP, Ephrin-B2 and CXCL12 signaling

Project description:In the tumor microenvironment the extracellular matrix molecule tenascin-C is highly expressed which correlates with worsened survival prognosis. Tenascin-C promotes multiple steps in cancer progression. In particular, tenascin-C promotes the tumor angiogenic switch and the formation of more but poorly functional blood vessels by ill defined mechanisms. Here, we studied tenascin-C angio-modulatory functions by tumor and stromal cells. Unexpectedly, we observed that direct contact of endothelial cells with tenascin-C impairs angiogenesis through disruption of actin polymerization. This resulted in cytoplasmic retention of the F-actin sensor and co-transcription factor YAP and led to downregulation of YAP pro-angiogenic target genes. Conversely, tumor cells and carcinoma associated fibroblasts exposed to tenascin-C secreted pro-angiogenic factors that promoted endothelial cell survival and tubulogenesis. We identified and functionally validated CXCL12 and Ephrin- B2 as important pro-angiogenic effectors of tenascin-C. Proteomic analysis of the secretome of tumor cells exposed to tenascin-C revealed a signature that predicts shorter survival of patients with low grade glioma and glioblastoma with a particular significance for a combined expression of tenascin-C and Ephrin-B2. Altogether, we demonstrated a dual mechanism of action of tenascin-C in tumor angiogenesis where direct contact of endothelial cells with tenascin-C impairs angiogenesis, and paracrine signals derived from contact of tumor cells and carcinoma associated fibroblasts with tenascin-C promotes angiogenesis. These opposing effects provide for the first time an explanation of divergent mechanisms controlled by tenascin-C which can result in a denser but less functional tumor blood vasculature and might unveil new targeting and prediction opportunities.

Tasquinimod, a Quinoline-3-Carboxamide Anti-Angiogenic Agent, treated Prostate Cancer Cell Line in-vivo and in-vitro

Project description:The quinoline-3-carboxamide ABR-215050 (Tasquinimod (INN)), exhibits anti-tumor activity via inhibition of tumor angiogenesis in human and rodent tumors. To further explore the mode of action of ABR-215050, in vivo experiments with gene microarray analysis were performed using samples of LNCaP prostate tumors excised from nude mice exposed to ABR-215050 for 24h at 10 mg/kg(ad.lib.). The array data were validated by real-time PCR (sqRT-PCR), as well as by protein expression experiments. Among several significant differentially expressed gene after exposure to ABR-215050, were cytokine receptor CXCR4, cytochrome P450 1A1 (CYP1A1), thrombospondin-1 (THBS1) and Lysyloxidase preprotein (LOX). To further explore the mode of action of ABR-215050, in vitro experiments with gene microarray analysis were performed using LNCaP prostate tumor cells. The array data were validated by real-time PCR (sqRT-PCR), as well as by protein expression experiments. One of the most significant differentially expressed genes after exposure to ABR-215050, was thrombospondin-1 (THBS1). 2-color treatment vs. control microarray experiment comprising four biological replicates. Each biological replicate was hybridized with dye-swap; resulting in eight hybridizations for each dataset (in vitro and in vivo), for a total of 16 hybridizations.

DCBLD2 promotes perivascular anti-tumor immune deprivation by educating SPP1+ pro-angiogenic macrophages

Project description:The crosstalk between tumor vasculature and immune microenvironment underpins the anti-tumor effect of anti-angiogenic therapy (AAT) combined with immunotherapy. However, the efficacy improved by AAT in the above combination therapy is limited and its adverse effects are concerning. Integrated transcriptome and surfaceome data identified DCBLD2 as a bipotent molecule on tumor cells as a trigger for abnormal tumor vascular-immune interactions. Dcbld2 overexpression accelerated tumor growth through tumor-associated macrophages (TAMs) in preclinical mouse models. Mechanically, DCBLD2 domesticates SPP1+ pro-angiogenic TAMs through ALDH1A3-RXRG-PLK1 axis, constructing TAMs-coated vascular niches that sequester CD8+ T cell infiltration. DCBLD2 expression levels predicted the efficacy of traditional AAT in our lung cancer patient cohorts. Targeting DCBLD2 or ALDH1A3 combined with PD-1 antibody outperformed traditional AAT and immunotherapy combination. Our study identifies DCBLD2 on tumor cells as a checkpoint in abnormal vascular-immune crosstalk, offering a promising target to amplify the potency of vascular normalization and immune activation.
