ABSTRACT: Colorectal cancer is one of the most common causes of cancer-related death. Early diagnosis is extremely important in terms of treatment and mortality. In this study, we investigated the diagnostic value of serum autotaxin levels in colorectal cancer.

Project description:We used microarrays to perform systematic profiling of human microRNAs in serum from glioblastoma multiforme (GBM) patients to find potential biomarkers. By comparing the serum microRNA profiles of the GBM patients and healthy volunteers, potential biomarkers for GBM were investigated. Using a custom microarray platforms containing 1843 species of miRNAs, we previously identified 19 serum miRNAs with differential expression in GBM patients.

Real-time quantitative PCR analysis of pre-treatment serum microRNAs from patients with prostate cancer

Project description:The overall goal of the study was to determine if serum or serum EV microRNAs added prognostic value to current clinical nomograms for prostate cancer. Serum was collected from 203 patients with biopsy-proven prostate cancer. EVs were isolated from the 132 serum samples. RNA was isolated from these serum and serum EV samples, and 61 microRNAs were quantified per sample on a custom qPCR plate. The microRNA data was normalized using NormFinder. The normalized microRNA data was then compared to adverse pathology and prostate biopsy Gleason grade group.

The Diagnostic Value of Salivary microRNA in Esophageal Cancer

Project description:Tissue microRNAs (miRNAs) can detect cancers and predict prognosis. Several recent studies reported that tissue, plasma, and saliva miRNAs share similar expression profiles. In this study, we investigated the diagnostic value of salivary miRNAs (including whole saliva and saliva supernatant) for detection of esophageal cancer. By Agilent microarray, six deregulated miRNAs from whole saliva samples from seven patients with esophageal cancer and three healthy controls were selected. The six selected miRNAs were subjected to validation of their expression levels by RT-qPCR using both whole saliva and saliva supernatant samples from an independent set of 39 patients with esophageal cancer and 19 healthy controls.

Extracellular miRNAs in serum and feces of mice exposed to high-dose radiation

Project description:Exposure to high-dose radiation causes life-threatening serious intestinal damage. Histological analysis is the most accurate method for judging the extent of intestinal damage after death. However, it is difficult to predict the extent of intestinal damage to body samples. Here we focused on extracellular microRNAs (miRNAs) released from cells and investigated miRNA species that increased or decreased in serum and feces using a radiation-induced intestinal injury mouse model. A peak of small RNA of 25–200 nucleotides was detected in mouse serum and feces 72 h after radiation exposure, and miRNA presence in serum and feces was inferred. MiRNAs expressed in the small intestine and were increased by more than 2.0-fold in serum or feces following a 10 Gy radiation exposure were detected by microarray analysis and were 4 in serum and 19 in feces. In this study, miR-375-3p, detected in serum and feces, was identified as the strongest candidate for a high-dose radiation biomarker in serum and/or feces using a radiation-induced intestinal injury model.

Deep sequencing identified serum extracellular vesicle delivered miR-181a-5p as an indicator for bone-metastatic prostate cancer (microarray)

Project description:Purpose: The goals of this study are to compare the serum extracellular vesicle (EV) delivered miRNA levels of patients with bone-metastatic prostate cancer (PCa), non-bone -metastatic PCa and benign prostatic hyperplasia (BPH), and to identify EV-delivered microRNAs in patient’s serum as indicators for bone-metastatic PCa. Methods:Serum extracellular vesicle delivered miRNA profiles of patients with bone-metastatic PCa or non-bone -metastatic PCa or BPH were generated by miRNA chip array, using Agilent-070156 Human_miRNA_V21.0_Microarray plateform. Results: Differential analysis showed the expressions of 27 EV delivered miRNAs were significantly different between serum of patients with bone-metastatic PCa and non-bone-metastatic PCa with a p value <0.05. the expressions of 5 EV delivered miRNAs were confirmed with qRT–PCR. Conclusions: Serum EV-delivered miR-181a-5p is a promising diagnostic biomarker for bone-metastatic PCa.

Differential analysis showed the expressions of 35 EV delivered miRNAs were significantly different between serum of patients with PCa and BPH, with a p value <0.05. the expressions of 5 EV delivered miRNAs were confirmed with qRT–PCR. Conclusions: Serum EV-delivered miR-181a-5p is a promising diagnostic biomarker for bone-metastatic PCa.
