Project description:To characterize the impact of neoadjuvant ABFOLFOX (Atezolizumab, Bevacizumab, and FOLFOX) therapy on the tumor microenvironment of colorectal liver metastases (CRLM), we performed a comprehensive molecular analysis using bulk RNA-seq. The study comprised two distinct cohorts: a cross-sectional cohort (Cohort A, n=60) and a prospective longitudinal cohort (Cohort B, n=20). Cohort A included patients who underwent hepatic surgery, categorized into three groups (n=20 each): those who underwent upfront surgery (G1 group); those treated with neoadjuvant chemotherapy (NAC; FOLFOX or FOLFIRI) (G2 group); and those treated with NAC plus bevacizumab (G3 group). In Cohort B, CRLM specimens were analyzed at three serial time points: T0 (baseline liver biopsy), T1 (biopsy after atezolizumab monotherapy), and T2 (surgical or biopsied specimen after ABFOLFOX). Our findings suggest a progressive increase in immunogenicity within the CRLM tumor ecosystem following treatment. In particular, the monocyte lineage and SP140 regulon appear to serve as potential mediators of the observed therapeutic benefits. This submission presents the transcriptomic data supporting the interplay between systemic therapy and the immune landscape in CRLM.

Liver metastases license neutrophils throygh IL-1 to potentiate cancer progression

Project description:Liver metastases are associated with poor cancer outcomes in many solid malignancies, but the factors influencing the trajectory of patients with liver metastases are poorly defined. It is known that liver metastases suppress systemic antitumor immunity; however, the underlying mechanisms remain incompletely described. We report that liver metastases promote disease progression in patients and preclinical models. Patients with liver metastases progress rapidly, regardless of primary tumor type. In multiple murine models, we find that liver metastases potentiate neutrophil migration and activity. Neutrophils licensed by liver metastasis augment metastatic colonization in an IL-1 dependent fashion. Thus, liver metastasis rewires systemic immunity to promote cancer progression. This work has implications for novel treatment strategies to address the poor clinical outcomes associated with liver metastasis.

Proteomics analysis of the matrisome from MC38 experimental mouse liver metastases

Project description:Dissemination of primary tumors to distant anatomical sites has a substantial negative impact on patient prognosis. The liver is a common site for metastases from colorectal cancer, and patients with hepatic metastases have generally much shorter survival, raising a need to develop and implement novel strategies for targeting metastatic disease. The extracellular matrix (ECM) is a meshwork of highly crosslinked, insoluble, high molecular weight proteins maintaining tissue integrity and establishing cell-cell interactions. Emerging evidence identifies the importance of the ECM in cancer cell migration, invasion, intravasation, and metastasis. Here, we isolated the extracellular matrix from MC38 mouse liver metastases using our optimized method of mild detergent solubilization followed by biochemical enrichment. The matrices were subjected to label-free quantitative mass spectrometry analysis, revealing proteins highly abundant in the metastatic matrisome. The resulting list of differentially expressed proteins significantly predicted survival in patients with colorectal cancer but not other cancers with strong involvement of the extracellular matrix component. One of the proteins upregulated in liver metastatic ECM, Annexin A1, was not previously studied in the context of cancer-associated matrisome. Here we show that Annexin A1 was markedly upregulated in colon cancer cell lines compared to cancer cells of other origin, and also overrepresented in human primary colorectal lesions as well as hepatic metastases in comparison with their adjacent healthy tissue counterparts. In conclusion, our study provides a comprehensive ECM characterization of MC38 experimental liver metastases and proposes Annexin A1 as a putative target for this disease.
