Project description:This study is being done to select an appropriate TF2 bsMAb dose suitable for pretargeting the 111In/90Y-labeled hapten-peptide (IMP-288). Eligible patients will receive a fixed dose of 90Y-IMP-288 4 days after the TF2 antibody injection. Two different dose levels of TF2 will be studied in the first part.
Once an appropriate TF2 dose is selected based on information learned from the first 2 dose levels, patients will be enrolled onto several different increasing dose levels of 90Y-IMP-288.

Project description:Clinical trials of MSCs in the treatment of OA have been carried out and good results have been obtained. However, these data on the dose of transplanted cells, the number of transplants, the therapeutic effect especially cartilage regeneration and safety are incomplete. Therefore, we constructed a cynomolgus macaque model consistent with the pathogenesis of OA. We have also established clinical-grade human umbilical cord-derived mesenchymal stem cell line. We selected two cell doses (2.5*10e7 cells per knee and 5*10e6 cells per knee, respectively) to assess differences in treatment effects and to assess the systemic safety of MSC therapy. High-dose hUC-MSCs can regenerate cartilage, increase joint space, and improve bone condition by ultrasound and MRI scans. But low-dose hUC-MSCs simply prevented further deterioration of the joint

Project description:BACKGROUND: Preclinical studies have demonstrated that pharmacological mobilization and recruitment of endogenous bone marrow stem cells and immunoregulatory cells by a combination of plerixafor and low-dose tacrolimus (MRG-001) improves wound healing, promotes tissue regeneration and prevents allograft rejection. This first‐in‐human phase I dose‐escalation study evaluates the safety, tolerability, pharmacokinetics and pharmacodynamics of MRG-001, a novel fixed-dose combination drug. METHODS: In this Phase 1, double‐blind, randomized, placebo-controlled study, multiple ascending dose (MAD) cohorts are randomized to receive MRG-001 containing up to 0.02 mL/kg (plerixafor 24 mg/mL and tacrolimus 0.5 mg/mL) or saline placebo, subcutaneously every other day (SC, QAD) for 5 days (ClinicalTrials.gov: NCT04646603)The primary outcome is safety and tolerability. Safety and functional assessments are performed throughout the study. Blood samples are collected to evaluate systemic exposure. Fluorescence-activated cell sorting analysis and RNA expression of peripheral blood mononuclear cells (PBMCs) are used to evaluate the pharmacodynamics. RESULTS: Fourteen subjects received MRG-001 and 7 received a placebo. MRG-001 is safe and well-tolerated over the selected dose range. No deaths or severe adverse events are reported. There are no clinically significant laboratory changes after MRG-001 administration, apart from the predicted generalized leukocytosis. The intermediate dose group (0.01 mL/kg) showed the most significant white blood cell mobilization over time and increased by 2-4 fold from baseline and returned to baseline levels prior to the next injection. Circulating immune cells including FOXP3+ regulatory T cells and hematopoietic stem cells (CD45IntCD34+) increased significantly after MRG-001 injection. PBMC RNA sequencing and gene set enrichment analysis revealeds 31 down-regulated pathways in the intermediate dose MRG-001 group compared to no changes in the placebo group. CONCLUSION: MRG-001 is safe and well-tolerated across the full dose ranges tested. MRG-001 may be a clinically useful therapy for immunoregulation and tissue regeneration. A Phase II trial to treat severely and critically ill COVID-19 patients with MRG-001 has been initiated (NCT04646603) and a second phase II trial will explore the potential of MRG-001 to accelerate wound healing (NCT05844527),

Project description:Food safety evaluation of new, genetically modified (GM) plant varieties has led to basic questions regarding the safety assessment of new plant varieties and whole foods derived thereof. An important part of the hazard identification in the European approach is a targeted compositional analysis of new GM plant varieties compared to one or more conventional reference varieties. Comparative analysis will become much more informative with unbiased analytical approaches, such as omics profiling. Analysis tools that estimate the similarity of new varieties to the reference would in turn greatly facilitate hazard identification. Further in-depth biological, functional and eventually toxicological analysis of the data would then only be necessary for varieties that fall outside the scale of those with a history of safe human consumption. For this purpose, the use of a one-class classifier tool was explored in this study to assess and classify transcriptome profiles of potato varieties. Five potato varieties were grown in the Netherlands during the same year (NL samples) and included four biological replicates for four varieties or two biological replicates for the fifth one. They were all analysed in 2011. A sixth variety was grown in the UK in a previous study and a previous year, for which the data are submitted in E-MTAB-605. The two UK samples were analysed in the original study in 2008 and again together with the NL samples in the present study, resulting in four profiles for two samples.

Project description:The ability to differentiate between threat and safety cues is essential for adaptive behavior, yet this process is disrupted in psychiatric disorders such as post-traumatic stress disorder (PTSD), where safety cues fail to suppress fear expression. To uncover the neurobiological mechanisms of safety learning, we compared safety conditioning (SC) and contextual fear conditioning (CFC) in the infralimbic cortex (IL) of the medial prefrontal cortex (mPFC) across recent and remote memory retrieval in mice. We identify a novel mechanism whereby safety learning promotes neuro-glial interactions that may facilitate plasticity in the mPFC. SC promotes long-term suppression of contextual fear compared to CFC. At remote retrieval, SC animals show increased IL parvalbumin (PV) interneuron activity and reduced perineuronal net (PNN) density, suggesting that extracellular matrix (ECM) remodeling could support enhanced plasticity in this group. We further identify a critical role for oligodendrocyte lineage cells (OLCs) in PNN remodeling. Notably, safety retrieval recruits satellite oligodendrocyte lineage cells (i.e., satOlig2+ cells) to PV interneurons. In SC animals, this recruitment correlates with reduced PNN accumulation around PV-satOlig2 pairs, whereas CFC animals exhibit increased PNN density at remote retrieval. Open-source single-cell RNA sequencing reveals that oligodendrocyte precursor cells (OPCs) express PNN-building genes, mature oligodendrocytes (OLs) express PNN-degrading genes, and newly-formed oligodendrocytes (NFOLs) express both. Bulk RNAseq from SC animals shows reduced expression of OPC-related structural genes and increased expression of OL-related PNN-degrading genes in the mPFC. Notably, satOlig2 cells recruited during remote safety retrieval are predominantly CC1+ mature satellite OLs (satOLs), which we hypothesize degrade PNNs through expression of PNN degradation enzymes. Optogenetic inhibition of PV activity during safety learning blocks satOlig2 recruitment, maturation, and PNN degradation while restoring long-term contextual fear expression. These findings suggest a role for satOlig2 cells in promoting safety learning through dynamic PNN regulation. By maintaining PV interneuron activity and excitatory-inhibitory balance, satOlig2 cells may facilitate PNN remodeling, in support of fear suppression and adaptive behavior. This study provides critical insights into the mechanisms of safety learning and highlights potential OL- and ECM-based therapeutic targets for PTSD and related psychiatric disorders.
