Project description:A majority of patients undergoing chemotherapy treatment develop side effects that delay further cancer treatment. Antimetabolites, like 5 Fluorouracil (5 FU), are known to induce gut and oral inflammation, highlighting the importance of chemotherapy toxicity management. Here, we aimed to study whether probiotic bacterium Limosilactobacillus reuteri DSM 17938 (LR)-secreted components, including cell-free supernatant (CFS), exopolysaccharides (EPS), and extracellular membrane vesicles (MV), can improve gut barrier integrity following 5 FU exposure. Collectively, 5 FU altered the viability, metabolic activity, barrier integrity, and functional response of Caco-2 cells. EPS stimulation after 5 FU removal significantly improved the integrity and permeability of intestinal barrier through modulation of the transcriptional program associated with extracellular matrix and structure organization. Further, we found that CFS, MV and EPS differentially influenced monocyte polarization pathways, when monocytes were cultured with the supernatant from 5 FU exposed Caco-2 cells. Together, our findings suggest that the incorporation of bacterial metabolites, such as EPS, during chemotherapy could accelerate intestinal barrier recovery without suppressing the immune system, potentially allowing for a more effective scheduling of therapy in cancer patients.

Project description:NAD+ is a crucial cofactor for the activity of various enzymes, including sirtuins and ADP-ribosyl transferases, and its decline is associated with aging and metabolic-related diseases. Therefore, a strong interest has been raised in the therapeutic use of NAD+ precursors (Vitamin B3s), but many suffer from poor bioavailability and adverse effects. This study characterizes the metabolic impact of dihydronicotinamide riboside, a recently identified novel form of NAD+ precursor. Upon oral administration in mice, NRH reaches all tissues examined. Chronic administration in low-fat diet-fed mice showed negligible metabolic effects, while high-fat diet-fed mice were protected against body weight gain and glucose intolerance. However, our study also unveiled potential side effects at higher doses. Thus, NRH could constitute an alternative NAD+ boosting strategy to prevent diet-induced metabolic complications and conditions associated with low NAD+ levels, but the therapeutic window must be optimized to maximize benefits and minimize risks.

Project description:Colorectal cancer is a common yet survivable malignancy, partly due to the addition of chemoradiation to treatment regimens. Many patients suffer treatment-related side effects such as pain, diarrhea, and myelosuppression, and increasing toleration of treatment has the potential to improve outcomes. BMX-001, a superoxide dismutase mimetic, is currently in clinical trials as a selective radioprotector for anal and rectal cancer; however, the mechanism by which BMX-001 exerts selective radioprotection of healthy tissue over cancer tissue, particularly when administered at physiologically achievable doses, is not well understood. BMX-001 was given before and during chemoradiation and did not interfere with chemoradiation-induced cancer cell killing in mouse models. In vitro, BMX-001 still induced cancer cell killing in the presence of exogenous catalase, suggesting that BMX-001’s anti-tumor activity is not solely reliant on hydrogen peroxide production. BMX-001 exerted robust acute radioprotection in radiation mouse models five and nine days post-radiation. To assess whether BMX-001’s protection is contingent on Nrf2 expression, we evaluated the effect of BMX-001 on 5-FU treated bone marrow, where Nrf2 is crucial to maintaining hematopoietic stem cell niches. Interestingly, BMX-001 could still exert some chemoprotection in mice without functional Nrf2. To assess whether BMX-001 had any functional effects on protein oxidation status, we performed mass spectrometry on sulfenylated proteins in bone marrow treated with 5-FU in wildtype mice and mice without Nrf2. We found that BMX-001 given before 5-FU treatment resulted in the sulfenylation of CRTC2 and CBP, two proteins involved in CREB signaling and known to enhance Nrf2 activity. We also found that BMX-001 with 5-FU treatment enhanced DPYD sulfenylation, a protein known to detoxify 5-FU, and ALDH7A1 sulfenylation, a protein known to detoxify aldehydes. Together, these results suggest that BMX-001 can protect against oxidative stress in normal tissue via Nrf2 dependent and independent mechanisms.

Project description:Developing safe and effective pain medications is an ongoing challenge for human health. Mu opioid receptor (MOR) agonists are essential pain medications, but their high intrinsic efficacy induces adverse side effects, including respiratory depression, constipation, tolerance, dependence, withdrawal, and addiction. Strategies to limit such effects traditionally include developing MOR agonists that have low intrinsic efficacy or preferentially activate G protein over β-arrestin signaling. We report the discovery of a novel MOR superagonist with supraphysiological intrinsic efficacy and a unique pharmacological profile that produces effective analgesia in rodents with minimal adverse side effects. N-desethyl-fluornitrazene (DFNZ) was derived from a class of synthetic benzimidazole opioids called Nitazenes. DFNZ has a unique spatiotemporal MOR cellular signaling profile that favors G protein activation over β-arrestin recruitment, impaired brain penetrance, and diminished efficacy at the MOR-galanin 1 receptor (Gal1R) heteromer. DFNZ does not induce respiratory depression, tolerance, or MOR desensitization after repeated exposure. Compared to other MOR agonists, DFNZ does not increase dopamine transmission in nucleus accumbens and has weaker reinforcing effects in the drug self-administration procedure. These results provide novel insights on MOR pharmacology, have important implications for pain and addiction treatment, and challenge the prevailing dogma that MOR superagonists cannot constitute safe and effective therapeutics.

Abstract:
Background and objectives: In recent decades, cancer has become one of the most fatal and destructive diseases which is threatening humans life. Accordingly, different types of cancer treatment are studied with the main aim to have the best treatment with minimum side effects. Anti-angiogenic is a molecular targeted therapy which can be coupled with chemotherapy and radiotherapy. Although this method does not eliminate the whole tumor, but it can keep the tumor size in a given state by preventing the formation of new blood vessels. In this paper, a novel model-free method based on reinforcement learning (RL) framework is used to design a closed-loop control of anti-angiogenic drug dosing administration.
Methods: A Q-learning algorithm is developed for the drug dosing closed-loop control. This controller is designed using two different values of the maximum drug dosage to reduce the tumor volume up to a desired value. The mathematical model of tumor growth under anti-angiogenic inhibitor is used to simulate a real patient.
Results: The effectiveness of the proposed method is shown through in silico simulation and its robustness to patient parameters variation is demonstrated. It is demonstrated that the tumor reaches its minimal volume in 84 days with maximum drug inlet of 30 mg/kg/day. Also, it is shown that the designed controller is robust with respect to ± 20% of tumor growth parameters changes.
Conclusion: The proposed closed-loop reinforcement learning-based controller for cancer treatment using anti-angiogenic inhibitor provides an effective and novel result such that with a clinically valid and safe dosage of drug, the volume reduces up to 1mm3 in a reasonable short period compared to the literature.
