Interferon-Beta Gene Transfer (Ad.hIFN-β) as Treatment for Refractory Colorectal Carcinoma With Liver Metastases

ABSTRACT: This study will be conducted in subjects with refractory colorectal carcinoma with unresectable liver metastases. The purposes of the study are: * to evaluate the safety and any harmful effects of an intravenous injection of Ad.hIFN-β; * help determine whether the virus carrying the interferon-beta gene will enter the bloodstream and liver tumor cells and cause the cancer cells to die.

Expression data of multiple sclerosis patients receiving Interferon-beta therapy

Project description:The purpose of this study was to characterize the transcriptional effects induced by intramuscular IFN-beta-1a treatment (Avonex, 30 Âµg once weekly) in patients with relapsing-remitting form of multiple sclerosis (MS). By using Affymetrix DNA microarrays, we obtained genome-wide expression profiles of peripheral blood mononuclear cells from 24 MS patients within the first four weeks of IFN-beta administration. Keywords: Multiple sclerosis, Interferon, Pharmacogenomics, Affymetrix EDTA blood samples were taken from all patients immediately before first, second and fifth IFN-beta injection. Total RNA of Ficoll-isolated peripheral blood mononuclear cells from each sample was extracted, labelled and hybridized to Affymetrix Human Genome U133 A and B arrays to quantify the mRNA levels.

Project description:The transcriptome of Huh7 and Huh7.5 was sequenced from cells treated with interferon alpha (IFNα), beta (IFNβ) or lambda (IFNλ) for 8 hours. Each treatment was compared to respective mock-treated cells in 6 pairwise comparisons, for a total of 43 samples classified in 12 experimental conditions. Cells were treated for 8h with 10ng/ml of Interferon-α2A, 5000 U/ml of Interferon-β1A or 100ng/ml of interferon-λ3 prior RNA extraction and library preparation. Mock-treated cells were used as control for each treatment. 3 to 4 biological replicates were analysed for each biological condition. Libraries for Illumina sequencing were constructed by polyA selection with the Illumina Kit. A paired end (2x100) run was performed.

Gene expression profiling of interferon-beta stimulated cells

Project description:Cytoplasmic DNA triggers the activation of the innate immune system. While downstream signaling components have been characterized, the DNA sensing components remain largely elusive. We performed a systematic proteomics screen for proteins that associate with DNA, traversed to a screen for IFN-β-induced transcripts. We identified DSIRE (DNA sensor for the IL-1β response, previously called AIM2) as a candidate cytoplasmic sensor. DSIRE showed a marked selectivity for double-stranded DNA. DSIRE can recruit the inflammasome adaptor ASC and gets redistributed to ASC speckles upon coexpression of ASC. RNAi-mediated reduction of DSIRE expression led to an impairment in IL-1β maturation. Reconstitution of unresponsive cells with DSIRE, ASC, caspase 1 and IL-1β showed that DSIRE is sufficient for inflammasome activation. Overall, our data strongly suggest that DSIRE is a cytoplasmic DNA sensor for the inflammasome. In a genomic screen, we used NIH3T3, L929 and RAW264.7 cells to identify genes that were transcriptionally regulated by IFN-β. We stimulated the cells with IFN-β for 4h, isolated the RNA and analyzed global changes in gene expression by microarray analysis. Overall, 225 genes were upregulated at least 3fold, among which numerous well-known interferon-induced genes were found
