Toxicity/Benefit Ratio Optimization of Chemotherapy in Colorectal Cancer (CRC) Patients by Determination of Individual Genotypic Determinants

ABSTRACT: This study intends to optimize a fluorouracil/irinotecan chemotherapy regimen by the identification of individual thymidylate synthase (TS) and UDP-glucuronosyltransferase 1 (UGT1A1) polymorphisms before the first administration. The results of this identification determine the chemotherapy type: high-dose irinotecan or not.

Phase II study of therapy selected by tumor molecular profiling in patients with previously treated metastatic pancreatic cancer. A study of the Stand Up to Cancer (SU2C) study consortium

Project description:The study accrued 49 subjects, and 48 underwent a tumor biopsy. and MP based therapy was administered to 35 patients. Study therapy was not given in 13 subjects either due to insufficient tumor on biopsy (n=8) or due to worsening cancer related symptoms after biopsy (n=5) . The demographics of evaluable patients (n=35) are M/F (59%/41%), with and age range was 34-78 (median 63 years). Time from first diagnosis of mPC to biopsy ranged from 5.8-26.7 months (median 16.1 months). Patients had 1-6 prior regimens with a median of 2. The most common IHC targets were topoisomerase 1 or 2, thymidylate synthase, ERCC1 and SPARC. Only commercially available agents were prescribed. Common regimens/agents recommended were FOLFIRI, FOLFOX, irinotecan and doxorubicin. Response rate was 9% , overall median survival of 6 months and 1 year survival was 21%.

Contact-based thymidylate transfer promotes collective tumor growth

Project description:Sustained cell proliferation is a fundamental hallmark of cancer, yet its mechanism remains elusive. While tumor cells often cooperate to enhance survival, the specific molecular forces driving this behavior are unknown. We uncover a novel mechanism of tumor growth, that cancer cells proliferate through a contact-dependent transfer of thymidylate (dTMP) via gap junctions with neighboring cells, bypassing canonical biosynthetic and salvage pathways driven by thymidylate synthase (TS) and thymidine kinase (TK1). Importantly, using a genetic mouse model of lung cancer harboring dual TS/TK1 tumor-specific knockout, we show that cancer cells lacking canonical synthesis maintain proliferation. These findings advance the current dogma of a ubiquitous nucleotide-driven activation in cancer, suggesting that a programmed dTMP re-equilibration sustains tumor growth. This novel mechanism could be exploited in cancer therapy.

Whole transcriptome sequencing of HCT116 cells with altered DNA repair capacities treated with RTX or 5FdUR

Project description:Thymidylate synthase (TS) is a key enzyme in thymidylate biosynthesis and an established target of chemotherapeutics such as 5-fluoro-2’-deoxyuridine (5FdUR) and raltitrexed (RTX). Inhibition of TS disrupts the dUTP:dTTP balance, leading to uracil misincorporation, futile base excision repair cycles, DNA strand breaks, and ultimately cell death. However, active U-DNA repair is apparently not required for the cytotoxic effect of these drugs, indicating that alternative pathways contribute to the observed S-phase arrest. To dissect these mechanisms, we investigated the transcriptomic effects of TS inhibition in HCT116 colon cancer cell lines with altered DNA repair capacities treated with 5-fluoro-2′-deoxyuridine (5FdUR) or raltitrexed (RTX). Both drugs induced similar DNA damage responses and S-phase arrest, yet displayed distinct transcriptional signatures. Our findings underscore the multifaceted impact of TS inhibition and reveal drug-specific differences in cellular responses.

Beyond its catalytic role, TS also binds RNA, autoregulating its own translation and interacting with transcripts such as p53 and c-myc, thereby linking TS activity to broader post-transcriptional regulatory networks. To better understand how TS inhibitory drugs influence this network, we performed TS-coupled RNA-coimmunoprecipitation (TS-RIP) and sequencing from mismatch repair-deficient, UNG-inhibited HCT116 colon cancer cells treated with 5FdUR or RTX. TS-RIP revealed RNA-binding targets of TS, in addition to the known p53 and TS mRNAs. Our findings underscore the multifaceted impact of TS inhibition, linking enzymatic disruption to RNA-level regulation and revealing drug-specific differences in cellular responses.

Molecular characterization of Plasmodium falciparum Bruno/CELF RNA binding proteins

Project description:The human malaria parasite Plasmodium falciparum employs intricate post-transcriptional regulatory mechanisms in different stages of its life cycle. Despite the importance of post-transcriptional regulation, key elements of these processes, namely RNA binding proteins (RBPs), are poorly characterized. In this study, the RNA binding properties of P. falciparum proteins were characterized including two putative members of the Bruno/CELF family of RBPs (PfCELF1 and PfCELF2), dihydrofolate reductase-thymidylate synthase (PfDHFR-TS), and adenosine deaminase (PfAda).The mRNA targets of these P. falciparum proteins were investigated by ribonomics using DNA microarrays.
