Project description:ChAdOx1 nCoV-19, a chimpanzee adenovirus (ChAd)-vector vaccine expressing the SARS-CoV-2 spike protein, has elicited robust immunological responses in large populations. Despite its proven efficacy and safety, some recipients have reported immediate inflammatory reactions post-vaccination. We comprehensively examined the immune landscape following ChAdOx1 nCoV-19 vaccination based on the epigenomic profiles of monocytes. Glucocorticoids are wildly used as steroid anti-inflammatory drugs to modify acute and chronic inflammation. Recently, dexamethasone is used to modify inflammation in COVID19 and side-effects after COVID19 vaccination. ChAdox1 nCoV-19 induces interferon responses and inflammation in human. In addition to IFNa, TNF and IL1b treatment to mimic inflammatory environment gives general effects of glucocorticoids in complex inflammatory context.

Project description:Nevirapine is a non-nucleoside reverse transcriptase inhibitor, a class of antiretroviral drug, used for the treatment of HIV-1 infection. Despite its wide use, nevirapine treatment has been associated with a significant incidence of different kind of hypersensitivity reactions (HSRs). We used microarrays to find significant genes that can relate to Nevirapine-persuaded hypersensitivity reactions in ‘acute’ patients compared to ‘recovered’ and/or ‘tolerant’ patients.

Project description:The anthracycline doxorubicin (Doxo) is one of the most effective and commonly used anti-cancer drugs. However, its clinical application is limited by severe side effects, such as cardiotoxicity, therapy-related tumors and infertility. Doxo induces both DNA damage through double-strand breaks and chromatin damage through histone eviction. We examined whether separating these activities would detoxify Doxo, while maintaining its chemotherapeutic efficacy. Here we present anthracycline variants harboring the histone eviction activity alone which remain potent anti-cancer drugs, while greatly alleviating cardiotoxicity and therapy-related tumor formation. This suggests that the treatment-limited side effects of Doxo in mice can be strongly reduced by modifying the chemical structure. The modified anthracycline anti-cancer drugs may provide effective chemotherapeutics allowing prolonged treatment and a higher quality of life post-treatment.

Project description:The majority of people in the U.S. manage health through at least one prescription drug. Drugs classified as non-antibiotics can adversely affect the gut microbiome and disrupt intestinal homeostasis. Here, we identified medications associated with an increased risk of GI infections across a population cohort of more than 1 million individuals monitored over 15 years. Notably, the cardiac glycoside digoxin and other drugs identified in this epidemiological study are sufficient to alter microbiome composition and risk of Salmonella enterica subsp. Typhimurium (S. Tm) infection in mice. The impact of digoxin treatment on S. Tm infection is transmissible via the microbiome, and characterization of this interaction highlights a digoxin-responsive b-defensin that alters microbiome composition and consequent immune surveillance of the invading pathogen. Combining epidemiological and experimental approaches thus provides an opportunity to uncover drug-host-microbiome-pathogen interactions that increase infection risk in human populations.

Pediatric neuro-ectodermal tumors originate from neural crest cells and show epigenetic defects of apoptotic pathways, which points to sensitivity towards epigenetic drugs in this patient group. The young age of these patients is accompanied by However, ongoing developmental processes regulated by epigenetic mechanisms may be deregulated by epigenetic drugs in this patient group that is characterized by young age. This prompted us to study molecular effects and side-effects of low dosage epigenetic drugs in neuro-ectodermal tumor cell lines of pediatric origin. Short term combination treatment of 5-aza-2`-deoxicytidine (DAC) and Trichostatin A (TSA) at nanomolar dosages reduced proliferation, induced wide-spread demethylating effects in 17 NBL and 5 PNET cell lines, and was accompanied by large effects on gene-expression profiles. Approximately half of the genes that were significantly upregulated upon treatment demonstrated significant demethylating effects in their promoter regions. In NBL cell lines, almost every cellular pathway (193/200) investigated demonstrated altered expression upon treatment, and resulted in upregulation of known epigenetically regulated genes such as X-chromosomal, tissue-specific, and a limited number of imprinted genes, but also known tumor suppressor genes and oncogenes. In conclusion, genome-wide methylation and gene expression changes are induced DAC and TSA treatment at nanomolar dosages. This treatment affected more than 97% of cellular pathways investigated and further studies towards the effectiveness and side-effects of epigenetic drugs are desirable in pediatric tumors. Project description:Development of chemotherapy has led to a high survival rate of cancer patients; however, severe side effects of anti-cancer drugs, including organ hypoplasia, persists. This study examined the molecular mechanism of the side effects of cyclophosphamide (CPA), a commonly used anti-cancer drug for the treatment of leukemia, and determined how to suppress the side effects using an organ culture system. Treatment with CPA disturbed the growth of tooth germs by inducing apoptosis and suppressing cellular proliferation and differentiation. Furthermore, organs cultured at low-temperature could avoid the CPA-mediated inhibition of organ morphogenesis and differentiation. Cap analysis of gene expression of CPA-treated tooth germ revealed that the expression of genes related to the G1/S cell cycle checkpoint were down-regulated in the CPA-treated tooth germ cultured at low-temperature. In vitro analysis revealed that low-temperature impeded Rb phosphorylation and caused cell cycle arrest at the G1 phase. This can prevent the CPA-mediated cell damage of DNA replication caused by the cross-linking reaction of CPA. Our results revealed that the side effects of CPA on organ development can be avoided by maintaining a low temperature. Further, this study provides a new screening model for assessing the effects of anticancer drugs on organ development.

Project description:Small molecule ligands often have multiple effects on the transcriptional program of a cell: they trigger a receptor specific response and additional, indirect responses ("side effects"). Distinguishing those responses is important for understanding secondary effects of drugs and for elucidating molecular mechanisms of toxic chemicals. We addressed this problem by exposing cells to the environmental contaminant benzo-a-pyrene (B[a]P). B[a]P exposure activates the aryl hydrocarbon receptor (Ahr) and causes toxic stress resulting in transcriptional changes that are not regulated through Ahr. We sought to distinguish these two types of responses based on a time course of expression changes measured after B[a]P exposure. Using Random Forest machine learning we classified 81 direct Ahr targets and 1,308 genes regulated as secondary exposure effects. Subsequent weighted clustering gave further insight into the connection between expression pattern, mode of regulation, and biological function. Finally, the accuracy of the predictions was supported through extensive experimental validation. This study presents a strategy for distinguishing receptor dependent and secondary responses based on expression time courses. 36 samples were analyzed in total. Time-course data for four different time points, two different benzo(a)pyrene concentrations, including a vehicle control for each time points. Three biological replicates were generated for each treatment and time point.

Project description:Cardiovascular toxicity causes adverse drug reactions and may lead to drug removal from the pharmaceutical market. Cancer therapies can induce life-threatening cardiovascular side effects such as arrhythmias, muscle cell death, or vascular dysfunction. New technologies have enabled cardiotoxic compounds to be identified earlier in drug development. Human induced pluripotent stem cell (hiPSC)-derived cardiomyocytes (CMs) and vascular endothelial cells (ECs) can screen for drug-induced alterations in cardiovascular cell function and survival. However, most existing hiPSC models for cardiovascular drug toxicity utilize two-dimensional, immature cells grown in static culture. Improved in vitro models to mechanistically interrogate cardiotoxicity would utilize more adult-like, mature hiPSC-derived cells in an integrated system whereby toxic drugs and protective agents can flow between hiPSC-ECs that represent systemic vasculature and hiPSC-CMs that represent heart muscle (myocardium). Such models would be useful for testing the multi-lineage cardiotoxicities of chemotherapeutic drugs such as VEGFR2/PDGFR-inhibiting tyrosine kinase inhibitors (VPTKIs). Here, we develop a multi-lineage, fully-integrated, cardiovascular organ-chip that can enhance hiPSC-EC and hiPSC-CM functional and genetic maturity, model endothelial barrier permeability, and demonstrate long-term functional stability.

Possible causes include allergic, cytotoxic or immunologic reactions to this agent. We examined the consequences of the mechanism of amiodarone-induced pulmonary toxicity gene expression in BEAS-2B cells, huma bronchial cell line, by microarray. The expression of these genes are potential biomarker of amiodarone-induced pulmonary toxicity. Also, We provide a clue about mechanism of pulmonary toxic action by these clinical chemotherapeutic agents.

Project description:Chronic graft-versus-host disease (cGvHD) is a frequent complication of allogeneic hematopoietic stem cell transplantation (alloSCT) and is associated with high morbidity and mortality. First drugs have been recently approved for the treatment of cGvHD, but the response rates are suboptimal and treatment discontinuation due to side effects remains frequent. Here, we evaluated the efficacy and safety profile of porcupine inhibition by Wnt-C59 in monotherapy or in combination with ruxolitinib, ibrutinib or belumosudil in experimental sclerodermatous GvHD (sclGvHD). Treatment was well tolerated in all groups with improvements in clinical and histological features. Of these, ruxolitinib in monotherapy, and the combination therapies demonstrated the highest efficacy. The combination therapies of Wnt-C59 with ibrutinib or belumosudil showed additive antifibrotic effects in skin and lungs compared to the respective monotherapies. Our study may have direct translational relevance, since: 1) core signaling pathways targeted by these drugs were enriched in both human and murine sclGvHD, and 2) ruxolitinib, ibrutinib and belumosudil are already approved for the treatment of cGvHD, and porcupine inhibitors are in clinical trials for other fibrotic diseases. Thus, simultaneous targeting of different pathogenetically relevant pathways might be a safe and effective therapeutic approach in sclGvHD.
