First Line Metastatic Colorectal Cancer Therapy in Combination With FOLFOX

ABSTRACT: The purpose of this study is to see if Cediranib in combination with FOLFOX is effective in treating metastatic colorectal cancer and to see how it compares with Avastin (Bevacizumab) in combination with FOLFOX.

Single-Cell Analysis Reveals Cellular Reprogramming in Advanced Colon Cancer Following FOLFOX-Bevacizumab Treatment

Project description:The combination of FOLFOX and bevacizumab (FOLFOX-Bev) is a promising treatment for advanced colorectal cancer (CRC). To gain insights into the cellular changes associated with FOLFOX-Bev treatment, we conducted single-cell transcriptomic analysis of CRC samples derived from a patient before and after treatment. Our results show that cancer cells with high proliferative, metastatic, and pro-angiogenic properties respond better to FOLFOX-Bev treatment. Moreover, FOLFOX-Bev enhances CD8+ T cell cytotoxicity, thereby boosting the anti-tumor immune response. Conversely, FOLFOX-Bev impairs the functionality of tumor-associated macrophages, plasma cells, and cancer-associated fibroblasts, leading to a decrease in VEGFB-mediated angiogenesis. Furthermore, FOLFOX-Bev treatment reset intercellular communication, which could potentially affect the function of non-cancer cells. Our findings provide valuable insights into the molecular mechanisms underlying the response of advanced CRC to FOLFOX-Bev treatment and highlight potential targets for improving the efficacy of this treatment strategy.

Identification of SNP candidates associated with chemo-responsiveness in colorectal cancer patients

Affymetrix SNP 6.0 chips were used to determine genotypes of the same colorectal cancer patients. Single-cell RNA sequencing of MC38 subcutaneous allograft tumors in C57BL/6J mice treated with FOLFOX alone or in combination with a composite probiotics

Project description:This study employs single-cell RNA sequencing (scRNA-seq) to investigate the impact of a probiotic consortium, alone or in combination with the chemotherapeutic regimen FOLFOX, on the tumor microenvironment (TME) of MC38 colorectal adenocarcinoma allografts in C57BL/6J mice. MC38 tumor-bearing mice were divided into three experimental groups: untreated control, FOLFOX-treated, and FOLFOX plus composite probiotics-treated. Tumors were harvested at a defined endpoint, dissociated into single-cell suspensions, and subjected to scRNA-seq using the 10x Genomics platform. The resulting data enable high-resolution characterization of transcriptional changes across immune, stromal, and malignant cell populations within the TME. This dataset aims to elucidate how combinatorial probiotic and chemotherapy intervention reshapes cellular composition, cell-state transitions, and intercellular communication networks, potentially revealing mechanisms of enhanced anti-tumor immunity or therapy resistance.

Probiotics Complex Synergizes with FOLFOX to Enhance Antitumor Response and Ameliorate Chemotherapy-induced Intestinal Toxicity via Upregulation of NAD+ Metabolism

Project description:Modulating the gut microbiota can improve chemotherapy efficacy, but strategies that simultaneously reduce toxicity and enhance therapeutic outcomes remain understudied. This study developed a NAD⁺-enhanced composite probiotic formulation (derived from fermented yak milk from the Qinghai-Tibet Plateau) to alleviate FOLFOX-induced intestinal toxicity and enhance efficacy in a colitis-associated colorectal cancer (CAC) mouse model. Metabolomics revealed significant contributions of nicotinic acid and nicotinamide metabolism to the body's response to chemotherapy. Consequently, RNA-seq was employed to validate the criticality of nicotinamide metabolism at the gene expression level: probiotics counteracted FOLFOX-induced NAD⁺ depletion, restored SIRT1/3 activity, and suppressed FoxO3a-mediated chemotherapy resistance. A clinical cohort of 53 gastrointestinal cancer patients confirmed serum NAD⁺ levels correlate with gut microbiota and intestinal toxicity. Collectively, we propose a microbiota-targeting strategy synergizing with FOLFOX, providing a translatable paradigm for precision oncology.

Global proteome changes in liver tissue 6 weeks after FOLFOX treatment of colorectal cancer liver metastases.

Project description:Study: Oxaliplatin-based chemotherapy for colorectal liver metastasis is associated with sinusoidal injury of liver parenchyma and may result in increased morbidity after liver surgery. Preclinical and human liver parenchyma studies have proposed several pathways of oxaliplatin-induced injury, but the effects on protein level remains unknown. Protein expression in liver tissue from eight patients treated with FOLFOX and seven controls without treatment was analyzed by label-free liquid chromatography mass spectrometry. Conclusion: Six weeks after FOLFOX treatment less than 1% of identified proteins showed changes in expression. The changes were associated with DNA replication, cell cycle entry and innate immune response. We hypothesize that the changes are residual after recovery from FOLFOX treatment injury.
