ABSTRACT: RATIONALE: Chemotherapy may cause blood clots to form in the thigh, leg, and lung. This study may help doctors understand how often blood clots occur in patients undergoing chemotherapy. PURPOSE: This clinical trial is studying how often blood clots occur in patients undergoing chemotherapy for solid tumors, including colorectal cancer, stomach cancer, lung cancer, ovarian cancer, pancreatic cancer, prostate cancer, or metastatic breast cancer

Candidate Biomarkers to Identify Mesothelioma Patients at Risk of Developing Venous Thromboembolism Post-Surgery

Project description:Pleural Mesothelioma (PM) is an aggressive cancer that attacks thousands of people every year. One of the common treatments is surgery to remove the tumor. Unfortunately, around 11% of patients die from blood clots post-surgery. Current predictive factors such as C-reactive protein, D-Dimer, and abnormal platelet count are not specific. To date, there are no specific blood-based protein biomarkers to identify PM patients at risk of developing Venous Thromboembolism (VTE) post-surgery. In this study, we present a set of host-response plasma protein candidate biomarkers that can predict patients at risk of developing VTE. We employed a quantitative mass spectrometry-based proteomics approach integrated with a multilayered, structured, and systematic evaluation of candidate biomarkers in a cohort of 18 patients, comprising six mesothelioma cases, six mesothelioma controls, and six lung cancer controls. This is the first step towards personalized treatment plans for PM patients undergoing surgery. This study's findings can potentially guide subsequent, larger-scale investigations, highlighting the value of small-scale exploratory research

Changes in L-phenylalanine concentration reflect and predict response to anti-PD-1 treatment combined with chemotherapy in patients with non-small cell lung cancer

Project description:Chemotherapy combined with checkpoint blockade antibodies targeting programmed cell death protein (PD-1) has achieved remarkable success in non-small cell lung cancer (NSCLC). However, few patients benefit from long-term treatment. Therefore, biomarkers capable of guiding the optimal therapeutic selection and reducing unnecessary toxicity are of pressing importance. In our research, we gathered serial blood samples from two groups of NSCLC patients: 49 patients received a combination of therapies, and 34 patients went under chemotherapy alone. Utilizing non-targeted metabolomic analysis, we examined different metabolites’ disparity. Among the lot, L-phenylalanine emerged as a significant prognostic in the combination treatment of NSCLC patients, interestingly absent in patients under sole chemotherapy. The reduced ratio of L-phenylalanine concentration (two cycles treatment vs. pre-treatment) was associated with improved progression-free survival (hazard ratio [HR] = 1.8000, 95% confidence interval [CI], 0.8566‒3.7820, p < 0.0001) and overall survival (HR = 1.583, 95% CI, 0.7416‒3.3800, p < 0.005). We further recruited two validation cohorts (cohort 1:40 patients and cohort 2:30 patients) to validate the sensitivity and specificity of L-phenylalanine prediction. Our results demonstrate that a model based on L-phenylalanine variations could serve as an early risk-assessment tool for NSCLC patients undergoing treatment, potentially facilitating strategic clinical decision-making.

The Illumina Infinium EPIC DNA methylation Beadchip was used to obtain DNA methylation profiles Survival has increased in early stage breast cancer (BC), and the late effects of cancer treatment can persist for decades. The molecular mechanisms underlying acceleration of age-related diseases after chemotherapy and radiotherapy are poorly understood. We examined epigenetic changes in peripheral blood in early stage BC patients undergoing either adjuvant radiotherapy, or adjuvant chemotherapy and radiotherapy. DNA methylation experiments were performed on peripheral blood samples from women collected prior to and following completion of adjuvant therapy. Methylation profiles were used to estimate three measures of epigenetic age acceleration - intrinsic, extrinsic, and phenotypic - as well as estimated cell counts.

Temporal characterization of serum metabolite signatures in lung cancer patients undergoing treatment

Project description:Lung cancer causes more deaths in men and women than any other cancer related disease. Currently, few effective strategies exist to predict how patients will respond to treatment. We evaluated the serum metabolomic profiles of 25 lung cancer patients undergoing chemotherapy ± radiation to evaluate the feasibility of metabolites as temporal biomarkers of clinical outcomes . Methods: Serial serum specimens collected prospectively from lung cancer patients were analyzed using both nuclear magnetic resonance (1H-NMR) spectroscopy and gas chromatography mass spectrometry (GC-MS). Multivariate statistical analysis consisted of unsupervised principal component analysis or orthogonal partial least squares discriminant analysis with significance assessed using a cross-validated ANOVA. Results: The metabolite profiles were reflective of the temporal distinction between patient samples before during and after receiving therapy (1H-NMR p<0.001 and GC-MS p<0.01). Disease progression and survival were strongly correlative with the GC-MS metabolite data whereas stage and cancer type were associated with 1H-NMR data. Metabolites such as hydroxylamine, tridecan-1-ol, octadecan-1-ol, were indicative of survival (GC-MS p<0.05) and metabolites such as tagatose, hydroxylamine, glucopyranose, and threonine that were reflective of progression (GC-MS p<0.05). Conclusions: Metabolite profiles have the potential to act as prognostic markers of clinical outcomes for lung cancer patients. Serial 1H-NMR measurements appear to detect metabolites diagnostic of tumor pathology, while GC-MS provided data better related to prognostic clinical outcomes warranting further study in a larger cohort and with various treatment options.

Project description:Lung cancer is one of the leading incident cancers among the various cancer types that occur in human. About 85% of lung cancer is non-small cell lung cancer (NSCLC). NSCLC is often diagnosed at the advanced stage and metastasis occurs in most cases, which explains why the overall survival rates for NSCLC are still low. Hyperthermia (HT) is one of the promising therapies in combination with chemotherapy and radiotherapy, and is known to be effective in improving survival and quality of life in patients with advanced lung cancer. We used microarrays to detail the global program of gene expression underlying HT and ionizing radiation (IR) combination therapy and identified a variety of genes whose expressions were regulated during this process.

RNA-seq of small cell lung cancer [circRNA-Seq]

Project description:We used CCK-8 experiment to determine the chemotherapy tolerance of small cell lung cancer cells, from which we found chemotherapy-sensitive cells (H446) and chemotherapy-tolerant cells (SHP77).By RNA-SEQ, we identified 401 circRNAs with abnormal expression associated with chemotherapy tolerance, including 212 up-regulated circRNAs and 189 down-regulated circRNAs. Through qRT-PCR verification, several circRNAs related to chemotherapy tolerance of small cell lung cancer were found, which helped us to further clarify the mechanism of chemotherapy tolerance of small cell lung cancer.
