Nitrous Oxide and Risk of Cancer Recurrence After Colorectal Surgery: A Randomized, Blinded Study

ABSTRACT: This study was developed to test the hypothesis that the risk of colorectal cancer recurrence was similar in patients who were randomly assigned to 65% nitrous oxide or nitrogen during colorectal surgery.

To test this, obese Zucker rats were randomly assigned to 2 groups of 12 rats each with similar body weights (BW) and fed diets either without (OC) or with 5 % PSC (OPSC) for 4 weeks and hepatic transcript profiling was performed. Transcriptomics of the liver revealed a total of 580 transcripts differentially expressed between OPSC and OC.

Project description:The hypothesis was tested that insect meal (IM) as protein source influences intermediary metabolism of growing pigs. To test this, 5-week-old crossbreed pigs were randomly assigned to 3 groups of 10 pigs each with similar body weights (BW) and fed isonitrogenous diets either without (CON) or with 5 % IM (IM5) or 10 % IM (IM10) from Tenebrio molitor L. for 4 weeks and liver was analyzed using transcriptomics. Transcriptomics of the liver revealed a total of 166 transcripts differentially expressed between IM10 and CON.

Influence of form of Se supplementation and tall fescue endophyte toxicity on the adrenal gland transcriptome of grazing beef steers

Project description:To test the hypothesis that average daily gain (ADG) and clinical parameters of steers grazing novel non-toxic (NTE) or toxic KY-31 (TE) endophyte-infected tall fescue would be improved by ad libitum intake of vitamin-mineral mixes (V-M) that contain 27 ppm Se as a 1:1 blend of SELPLEX:sodium selenite (MIX) vs. sodium selenite (ISe), 32 fescue-naïve beef steers depleted of Se were randomly assigned to ad libitum consumption ISe vs MIX for 35 d and fed enough of a NTE/alfalfa/grain diet to achieve 0.57 kg BW gain/d. Within Se-form treatments, 2 steers were randomly assigned to each of 4 NTE (ISe = 316 ± 31 kg, MIX = 315 ± 22 kg) or TE (ISe = 316 ± 37 kg, MIX = 314 ± 39 kg) paddocks for 84 d and had ad libitum access to their respective V-M. Steers were slaughtered over a 23-d period from day 90 to day 113 (August 27, 2019 to September 19, 2019) of the study and the left adrenal gland removed.

Expression data from corpora lutea from cows supplemented with different forms of selenium (Se)

Project description:The goal of this study was to test the hypothesis that sodium selenite (ISe) vs. a 1:1 blend (MIX) of ISe and SEL-PLEX in a basal vitamin-mineral (VM) mix would differentially alter corpora lutea transcriptome profiles in non-lactating beef cows. Predominately-Angus cows were randomly selected from herds of fall-calving cows consuming VM mixes that contained 35 ppm Se as ISe and MIX forms. Cows were depleted of Se for 45 d followed by a 45 d repletion period with ISe to bring all cows to Se adequate status. Cows were then randomly assigned to treatment (ISe, n=5; or MIX, n=5) with treatments administered for at least 90 days using in-pasture Calan gates. Corpora lutea samples were then collected on Day 7 post-estrus and examined for changes in global expression patterns by microarray analysis.

A 4-gene signature associated with recurrence risk in patients with low and intermediate-risk endometrial cancer

Project description:The molecular profile of endometrial cancer has become an important tool in determining patient prognosis and their optimal adjuvant treatment. In addition to The Cancer Genome Atlas (TCGA), simpler tools have been developed, such as the Proactive Molecular Risk Classifier for Endometrial Cancer (ProMisE). We attempted to determine a genetic signature to build a recurrence risk score in patients diagnosed with low- and intermediate-risk endometrial cancer. A case-control study was conducted. The eligible patients were women diagnosed with recurrence low- and intermediate-risk endometrial cancer between January 2009 and December 2014 at a single institution; the recurrence patients were matched to two nonrecurrence patients with the same diagnosis by age and surgical staging. Following RNA isolation of 51 cases, 17 recurrence and 34 nonrecurrence patients, the expression profile was determined using the nCounter® PanCancer Pathways Panel, which contains 770 genes. The expression profile was successfully characterized in 49/51 (96.1%) cases. We identified 12 genes differentially expressed between the recurrence and nonrecurrence groups. The ROC curve for each gene was generated, and all had AUCs higher than 0.7. After backward stepwise logistic regression, four genes were highlighted: FN1, DUSP4, LEF1, and SMAD9. The recurrence risk score was calculated, leading to a ROC curve of the 4-gene model with an AUC of 0.93, sensitivity of 100%, and specificity of 72.7%. We identified a four-gene signature that is associated the risk of recurrence in patients with low- and intermediate-risk endometrial cancer. This finding suggests a new prognostic factor in this poorly explored group of patients with endometrial cancer.
