Identification the Expression of Sox2 in Colorectal Cancer Tissues

ABSTRACT: Sox2 has been studied in several types of human solid tumors. The investigators found that Sox2 had higher expression level in colorectal cancer and metastatic tissues than normal tissues. So the investigators assumed that whether Sox2 plays an important role in the progression and migration of colon cancer.

Gene expression in normal mouse tissues

Project description:Gene expression was studied using PancChip5.0 in 15 tissues. Most tissues were from the adult mouse, except for fetal pancreas (e18.5) and placenta. Adult tissues studied were: adrenal gland, pancreas, brain, heart, kidney, liver, lung, ovary, parotid gland, pituitary gland, small intestine, stomach and testis. All were studied in duplicate except for the pituitary, the liver, and the stomach. All samples were pooled samples. Tissues were taken from 3 males and 3 females, with the exception of the gonadal tissues, and the fetal tissues were sex was not determined.

Identification of tRFs&tiRNAs Expression Profile in Breast Cancer Tissues

Project description:Purpose:In recent years, tRFs(transfer RNA-Derived Fragments) & tiRNAs (transfer RNA-Derived Stress-induced RNAs or tRNA halves) have been shown to have vital roles in cancer biology. We aimed to reveal the expression profile of tRFs&tiRNAs in breast cancer tissues in the study, and to explore their potential as biomarkers of breast cancer. Methods:We characterize tRFs&tiRNAs expression profiles in 6 pairs of breast cancer tissues and adjacent normal samples.Then we selected six tRFs&tiRNAs with significant expression and added 10 pairs of samples to verify the selected genes by qPCR in 16 pairs of breast cancer tissues along with the previous 6 pairs. Results:We found 31 differentially expressed tRFs&tiRNAs across our dataset, out of which 17 were up-regulated, 14 were down-regulated. Compared with 16 breast cancer tissues and healthy controls by qPCR, the results demonstrated that AS-tDR-001430, AS-tDR-001130 and AS-tDR-000779 were significantly expressed in breast cancer tissues (p < 0.001). AS-tDR-000779 was significantly up-regulated, AS-tDR-001430 and AS-tDR-001130 were significantly down-regulated which the expression patterns were similar to the sequencing results. Conclusions:Our studies have demonstrated that there were significantly expressed tRFs&tiRNAs in breast cancer tissues. They are hopefully to become biomarkers and would be valuable researches in this area.

Extrarenal ?-klotho expression in human tissues

Project description:?-Klotho has emerged as a powerful regulator of the ageing process. To-date, the expression profile of ?-Klotho in human tissues is unknown and its existence in some human tissue types is subject to much controversy. Objective: This is the first study to characterize system-wide tissue expression of transmembrane ?-Klotho in humans. We have employed next generation targeted proteomic analysis using Parallel Reaction Monitoring (PRM) in parallel with conventional antibody-based methods to determine the expression and spatial distribution of human ?-Klotho expression in health. Results: The distribution of ?-Klotho in human tissues from various organ systems, including arterial, epithelial, endocrine, reproductive and neuronal tissues was first identified by immunohistochemistry. Kidney tissues showed strong ?-Klotho expression, while liver did not reveal a detectable signal. These results were next confirmed by western blotting of both whole tissues and primary cells. To validate our antibody-based results, ?-Klotho expressing tissues were subjected to PRM mass spectrometry identifying peptides specific for the full length, transmembrane ?-Klotho isoform. Conclusions: The data presented confirms ?-Klotho expression in the kidney tubule and in artery, and provides evidence of ?-Klotho expression across organ systems and cell-types that have not previously been described in humans.

Project description:The tumor microenvironment (TME) plays an important role in tumorigenesis1. The TME is usually studied in tumor tissue and in relation to tumor progression2,3, which can limit understanding of how the TME is involved in processes associated with initial tumorigenesis as well as tumor recurrence and metastasis after surgery. To describe the underlying mechanisms of human hepatocellular carcinoma (HCC) occurrence and progression, we propose a new concept, the peritumor microenvironment (PME). We collected normal (all biochemical indicators of liver function, imaging examination and histopathology examination were normal) and peritumor (The liver tissues adjacent to the tumor were approximately 2 cm away from the tumor and were taken from patients with HCC who did not receive tumor radiotherapy, tumor chemotherapy or targeted drug therapy before surgery) to perform a proteomic characterization of the PME in peritumor liver tissues.

Tissue-level dissection of obesity-exercise axis in adipose-muscle tissues

Project description:We studied the opposing effects of exercise training and high-fat diet at tissue-level resolution to reveal gene expression and pathway changes in subcutaneous white adipose tissue, visceral white adipose tissue, and skeletal muscle. We revealed gene modules consistently up-regulated by exercise training, which include fatty acid biosynthesis/beta-oxidation/metabolism, oxidative phosphorylation, TCA cycle, and ROS response. Modules down-regulated by exercise training in all three tissues are antigen presentation, neutrophil degranulation, immune cell migration, phagosome-related genes, ECM-related genes, proliferation, and ribonucleotide biosynthesis. Taken together, exercise training improves fatty acid metabolism and cellular respiration similarly across the three metabolic tissues, and represses immune, ECM, proliferation and tissue-specific pathways.
