A Trial of PledOx + FOLFOX6 Compared to Placebo + FOLFOX6 in Patients With Metastatic Colorectal Cancer

ABSTRACT: The present trial is designed to determine whether pre-treatment with PledOx lowers the frequency and severity of side effects from FOLFOX6 administration in patients with metastatic colorectal cancer. The efficacy of PledOx will be assessed when added to FOLFOX6 chemotherapy as first line treatment of metastatic colorectal cancer. This study was performed in multiple parts/phases. Part 1 was an open dose-escalation study with the doses 2, 5 and 10 micromol/kg of calmangafodipir. No study outcomes were planned for this part. In part 2a, participants randomly received either Placebo, 2 or 10 micromol/kg of calmangafodipir. In part 2b, participants randomly received either Placebo, 2 or 5 micromol/kg of calmangafodipir. The overall intent of the study was to compare the effect of antioxidant agent PledOx against placebo in one of three different doses/combinations (2 micromol/kg, 5/10 micromol/kg, 2/5/10 micromol/kg vs. placebo, in the first 8 cycles of FOLFOX6 treatment

Biomarkers to Measure Treatment Response for Alcohol Dependence

Project description:The goal of this study was to identify molecular mechanisms regulated by pharmacological and placebo effects of binge-drinking at a transcriptome-wide level. We performed RNA-seq on PBMC samples collected from healthy heavy social drinkers (N=18) enrolled in an upto 12-day in-patient, randomized, double-blind, cross-over human laboratory trial testing three alcohol doses: Placebo, medium (0.05g/kg (men), 0.04g/kg (women)), and binge (1g/kg (men), 0.9g/kg (women)), administered in three 4-day experiments, separated by minimum of 7-day washout periods. Effects of beverage doses on differential expression of genes (DEGs) within each experiment compared to its own baseline, across experimental sequences in which each beverage dose was administered, and responsiveness to regular alcohol compared to placebo assessing pharmacological effects of alcohol were analyzed.

First-in-Human Study in Healthy Subjects with the Non-Cytotoxic Monoclonal Antibody OSE-127, a Strict Antagonist of the IL-7Rα

Project description:OSE-127 is a humanized monoclonal antibody targeting the IL-7Rα chain (CD127), under development for inflammatory and autoimmune disease treatment. It is a strict antagonist of the IL-7R pathway, is not internalized by target cells and is non-cytotoxic. Here, a first-in-human, phase I, randomized, double-blind, placebo-controlled, single-center study was carried out to determine the safety, pharmacokinetics, pharmacodynamics, and immunogenicity of OSE-127 administration.Sixty-three healthy subjects were randomly assigned to nine groups: six single ascending dose groups with intravenous (IV) administration (0.002-10 mg/kg), a single subcutaneous treatment group (1 mg/kg), and two double IV injection groups (6 or 10 mg/kg). Subjects were followed during < 146 days. OSE-127’s pharmacokinetic half-life after a single dose increased from 4.6 days (1mg/kg) to 11.7 days (10 mg/kg) and, after a second dose, from 12.5 days (6 mg/kg) to 16.25 days (10 mg/kg). Receptor occupancy was ≥ 95% at doses ≥ 0.02 mg/kg and this saturation level was maintained >100 days after two IV infusions at 10 mg/kg. IL-7 consumption was inhibited by OSE-127 administration, as demonstrated by a decreased IL-7 pathway gene signature in peripheral blood cells and by ex-vivo T-lymphocyte restimulation experiments. OSE-127 was well tolerated, with no evidence of cytokine-release syndrome and no significant alteration of blood lymphocyte counts or subset populations.Altogether, the observed lack of significant lymphopenia or serious adverse events, concomitant with the dose-dependent inhibition of IL-7 consumption by target cells highlight that OSE-127 may show clinical activity in IL-7R pathway-involved diseases.

A Mechanistic Study on the Synergistic Antihypertensive Effects of Dendrobium officinale Compound Combined with Conventional Antihypertensive Drugs in Spontaneously Hypertensive Rats, Based on Metabolomics

Project description:This dataset comprises metabolomic profiling data of serum and gut metabolites from 6 female Wistar rats and 42 female spontaneously hypertensive rats (SHR). Six Wistar rats were assigned to the normal control group (Normal), and 42 SHR rats were randomly divided into seven groups with six rats in each group: model group (Model), low-dose Dendrobium mixture group (LF), medium-dose Dendrobium mixture group (MF), high-dose Dendrobium mixture group (HF), irbesartan group (IRB), dual antihypertensive drug combination group (IAC), and herbal formula combined with dual antihypertensive drugs group (FIAC). The gavage volume for all groups was 2 mL/100 g body weight, administered once daily for six consecutive weeks. Rats in the LF, MF, and HF groups received the crude herbal formula at doses of 0.7 g/kg, 1.4 g/kg, and 2.8 g/kg, respectively. The IRB group received irbesartan at 15 mg/kg, the IAC group received a combination of irbesartan (15 mg/kg) and amlodipine (0.5 mg/kg), and the FIAC group received the herbal formula (2.8 g/kg) plus irbesartan (15 mg/kg) and amlodipine (0.5 mg/kg). The Normal and Model groups were administered an equivalent volume of physiological saline.

Hepatic premalignant alterations triggered by human carcinogen aristolochic acid I in canines

Project description:Ten-month-old male beagle canines were purchased from the ANNIMO Science and Technology Ltd (Nanjing, China, Certificate No. SCXK (Su) 2010-0002), and maintained in a specific pathogen-free environment. Canines were randomly assigned to two groups (4 in each group) and received capsules with control filler or Aristolochic Acid I (AAI) filler (3 mg/kg/day, equivalent dose of mouse) for 10 days. Canines were sacrificed at 11 days after initiation of the treatment. Livers were excised immediately after sacrifice. Part of the livers were immediately snap-frozen in liquid nitrogen and kept at -80°C for total RNA isolation and microRNA microarray analysis.

Effect of telmisartan on prostate carcinogenesis in TRAP rats

Project description:We investigate the direct effects of ARBs on prostate cancer growth and progression. A total of 36 heterozygous male TRAP rats of 3 weeks of age were randomly divided into three groups. Rats in group 1 as a control received basal diet and tap water. The rats in groups 2 and 3 continuously received 2 or 10 mg/kg/day telmisartan in drinking water for 12 weeks, respectively. Total RNAs of both control and telmisartan (10 mg/kg/day) were applied to microarray analysis.

Microarray Gene Expression Data from Early Skin Biopsies of a Secukinumab Clinical Trial in Psoriasis

Project description:<p>The phase II clinical study CAIN457A2223 (<a href="https://clinicaltrials.gov/ct2/show/NCT01537432">NCT01537432</a>) was designed to evaluate the proportion of patients achieving reversal of chronic plaque psoriasis at weeks 4 and 12 following multiple doses of secukinumab, administered subcutaneously, compared to placebo. 36 patients were enrolled in this study, with 24 being randomly assigned to the treatment arm and 12 to the placebo arm. Starting from week 13, all patients received multiple doses of secukinumab up to week 52 to study long term effects of secukinumab. The microarray gene expression data deposited in this dbGaP study are derived from the early (baseline to week 12) skin biopsies of this clinical trial.</p>

Predicting drug response in human prostate cancer from preclinical analysis of in vivo mouse models (II)

Project description:Analysis of the transcriptome of allografted mouse tumors after treatment with rapamycin and PD0325901. Nkx3.1CreERT2/+; Ptenflox/flox; KrasLSL-G12D/+ (NPK mice) were induced and their tumors removed to generate allograft lines by implanting a 1.5 mm3 tumor fragment in the subcutaneous space of athymic nude mice. Allografted NPK tumors were allowed to grow until they reached a volume of 1 cm3, at which moment they were randomly assigned to either vehicle (Veh) or combination therapy using rapamycin and PD0325901 (RAPPD). Allografted mice were administered rapamycin (10 mg/kg) and PD0325901 (10 mg/kg) during five consecutive days (Allo SHORT). Mice were euthanized in the fifth day 6 hours after having received the last treatment and the tumors were harvested and snap frozen for subsequent molecular analysis.
