Colorectal Cancer Metastatic

ABSTRACT: Primary Objective: To evaluate the safety of aflibercept in participants with mCRC treated with irinotecan/5-Fluorouracil (5-FU) combination (FOLFIRI) after failure of an oxaliplatin-based regimen (participants similar to those evaluated in the VELOUR trial [EFC10262, NCT00561470]) according to side effects prevention and management guidelines. Secondary Objective: To document the Health-Related Quality of Life (HRQL) of aflibercept in this participant population.

Plasma multi-miRNA models classify Alzheimer’s, Parkinson’s, and Lewy body dementia

Project description:We obtained 289 human plasma samples from national repositories in order to examine whether miRNAs could serve as biomarkers to distinguish diagnostic groups with similar clinical presentations and co-pathologies from each other. The participant groups included Alzheimer’s disease (AD), Parkinson’s disease (PD), and dementia with Lewy bodies (DLB). We also examined plasma miRNAs in normal control participants (NC).

Whole genome transcriptome profiling of Arabidopsis thaliana guard cells under saline growth conditions

Project description:The overall objective was to document transcriptomic changes in the guard cells of Arabidopsis thaliana under short (1x) - and long-term (3x) saline growth conditions. Guard cells of Arabidopsis responded also to the intensity of the salt management by the microarray datasets clearly clustering in (-) salt, 1x salt and 3x salt. Similarly, more GO terms were significantly enriched in differentially expressed guard cell genes of 1 x salt than 3x salt treated plants.

The Clinical impact of Chromosomal microarray on Paediatric care in Hong Kong (CGX-6)

Project description:Objective: To evaluate the clinical impact of chromosomal microarray (CMA) on the management of paediatric patients in Hong Kong Methods: We performed NimbleGen 135k oligonucleotide array on 327 children with intellectual disability (ID)/ developmental delay (DD), autism spectrum disorders (ASD), and/or multiple congenital anomalies (MCAs) in a university-affiliated paediatric unit from January 2011 to May 2013. The medical records of patients were reviewed in September 2013, focusing on the pathogenic/likely pathogenic CMA findings and their “clinical actionability” based on established criteria. Results: Thirty-seven patients were reported to have pathogenic/likely pathogenic results, while 40 had findings of unknown significance. This gives a detection rate of 11% for clinically significant (pathogenic/likely pathogenic) findings. The significant findings have prompted clinical actions in 28 out of 37 patients (75.7%), while the findings with unknown significance have led to further management recommendation in only 1 patient (p<0.001). Nineteen out of the 28 management recommendations are “evidence-based” on either practice guidelines endorsed by a professional society (n=9, Level 1) or peer-reviewed publications making medical management recommendation (n=10, Level 2). CMA results impact medical management by precipitating referral to a specialist (n=24); diagnostic testing (n=24), surveillance of complications (n=19), interventional procedure (n=7), medication (n=15) or lifestyle modification (n=12).

Project description:Whole blood methylomics, transcriptomics and plasma proteomics were obtained for 391 participants of the Progression of Early Subclinical Atherosclerosis (PESA) study. Epigenetic age was calculated from methylomics data for each participant. Its divergence from chronological age is termed epigenetic age acceleration. Subclinical atherosclerosis burden was estimated by multi-territory 2D/3D vascular ultrasound and by coronary artery calcification.

Comparative Proteomic Analysis of Saliva from Chewing Tobacco Users and Oral Cancer Patients Reveals Shared Biomarkers

Project description:Objectives: The aim of this study was to investigate the salivary proteomic profile of smokeless tobacco users in comparison to non-smokers using LC-MS/MS and to investigate the biological changes associated with smokeless tobacco use and oral cancer. Materials & Methods: Saliva samples of 65 participants were collected and divided into three groups: control (25 participants) , smokeless tobacco users group (25 participant), and oral cancer group (15 participant). The mean ages of the control participants, smokeless tobacco users, and oral cancer patients were 37.08 years, 37.8 years, and 52 years, respectively. The saliva samples were then prepared for LC-MS/MS analysis through in-solution digestion. The raw data files were processed using MaxQuant 2.1.0.0 with the human canonical proteome database. Results: The analysis revealed that 343 protein groups exhibited significantly altered abundance in the saliva samples. Among these, 43 out of 51 dysregulated proteins in the smokeless tobacco group were also dysregulated in the oral cancer group. Notably, Apolipoprotein A1 (ApoA1) and Pon1 were found to be significantly increased in both smokeless tobacco users and oral cancer patients. Furthermore, six out of the 20 most significantly altered proteins were mitochondrial proteins, and all of them were decreased relative to controls in both smokeless tobacco users and cancer samples.

Project description:Two cohorts with cerebrovascular disease are included in the present study, i.e., SCD group and VCI group. Participants in SVD group has the normal cognitive function and others in VCI group show the worse cognitive function. The whole-blood is collected from each participant, and RNA sequencing is conducted in whole-blood samples. Characteristic of circRNA, lncRNA, miRNA and mRNA are identified in the present blood samples.
