Project description:G protein-coupled receptors (GPCRs) are the largest class of cell surface signaling proteins; they participate in all physiological processes and are the targets of 30% of marketed drugs. Typically, nanomolar-micromolar concentrations of ligand are used to activate GPCRs in experimental systems. However, by measuring cAMP with increased spatial and temporal resolution, we can now detect GPCR responses to an extraordinarily wide range of ligand concentrations: from attomolar to millimolar. Mathematical modeling shows that the addition of femtomolar concentrations of ligand can activate a significant proportion of cells provided that a cell can be activated by 1-2 binding events. In addition to cAMP, activation of the endogenous b2-adrenoceptor (b2AR) and muscarinic M3R by femtomolar concentrations of ligand in cell lines and human cardiac fibroblasts causes sustained increases in nuclear ERK or cytosolic PKC, respectively. These responses are spatially and temporally distinct from those that occur at higher concentrations of ligand, and result in a unique proteomic profile. This highly sensitive signaling is dependent on the GPCRs forming pre-assembled higher-order signaling complexes at the plasma membrane. Recognizing that GPCRs respond to ultra-low concentrations of neurotransmitters and hormones challenges established paradigms of drug action and provides a new dimension of GPCR activation that is quite distinct from that typically observed.

Project description:Molecular mechanism underlying regeneration process, triggered by stem cells in tissue engineered urinary bladder, is still poorly explained. The study aimed to explore underlining pathways associated with regeneration process in urinary bladder reconstructed with stem cell seeded graft. The study was performed on 110 Wistar rats. Urinary bladders were augmented with bladder acellular matrix (BAM)(n=52) or BAM seeded with adipose derived stem cells (ADSCs)(n=52). The process of bladder healing was analyzed at 7, 30, 90 and 180 days postoperatively. Gene expression was evaluated using microarrays and analyzed in GeneSpring Software. Gene ontology (GO) and pathway enrichment analyses of differentially expressed genes (DEGs) were performed. A total of 4023, 4674, 7997 and 1120 of DEGs between the bladders augmented with ADSCs seeded BAM and BAM only were identified at 7, 30, 90 and 180 days postoperatively, respectively. The DEGs were enriched in GO terms associated with cellular and intercellular events, morphogenesis, epithelium, smooth muscles and nerves regeneration, angiogenesis, inflammatory response and wound healing. Numerous differentially expressed pathways between the bladders augmented with ADSCs seeded BAM and BAM only were identified. In conclusion, this study provided the unequivocal evidence that stem cells changed healing milieu in tissue engineered urinary bladder and indicated underlying pathways that can be associated with regeneration process triggered by stem cells.

Project description:Transportation stress causes significant changes in physiological responses in goats; however, studies exploring the transcriptome of stress are very limited. This study was conducted to analyze the transcriptome of stress and meat quality in goats using RNA-seq technology. Fifty-four male Spanish goats (8-mo old; BW = 29.7 ± 2.03 kg) were randomly subjected to one of three treatments (TRT; n = 18 goats / treatment): (i) transported for 180 min, (ii) transported for 30 min, or (iii) held in pens (control). Blood samples were collected before and after treatment for stress hormone, metabolite, and transcriptomic analysis. RNA-Seq technology was used to obtain the transcriptome profiles of blood. Analysis of physiological data using SAS showed that plasma cortisol concentrations were higher (P < 0.01) in 180 min and 30 min groups compared to the control group. A similar effect was noticed in plasma non-esterified fatty acid concentrations. Glucose concentrations were the lowest in the control group, highest in the 180 min group, and intermediate in the 30 min group (P < 0.01, while plasma creatine kinase concentrations were not significantly influenced by TRT. Neutrophil counts were higher (P < 0.01) and lymphocyte counts were lower (P < 0.01) in 180 min group compared to 30 min or control groups. The DESeq2 software identified a total of 430 DEGs for control vs. 30 min comparison, of which 127 genes were upregulated. For 180 min vs. control comparison, 741 DEGs were upregulated. Enrichment analysis of DEGs related to transportation stress through Gene Ontology and KEGG databases revealed that the DEGs related to inflammatory pathways, caspases, and apoptosis such as IL1R2, CASP14, CD14, TLR4, and MAPK14 were highly expressed in the transported group of goats compared to non-transported goats. Stress in goats leads to a sequence of events at cellular and molecular levels that causes inflammation and apoptosis and is also reflected in blood metabolites and leukocyte counts.

Project description:Background: Biomarkers that reflect glioblastoma tumour activity and treatment response are urgently needed to help guide clinical management, particularly for recurrent disease. As the urinary system is a major clearance route of circulating extracellular vesicles (EVs; 30-1000 nm nanoparticles) we explored whether sampling urinary-EVs could serve as a simple and non-invasive liquid biopsy approach for measuring glioblastoma-associated biomarkers. Methods: Fifty urine specimens (15-60 ml) were collected from 24 catheterised glioblastoma patients immediately prior to primary (n=17) and recurrence (n=7) surgeries, following gross total resection (n=9), and from age/gender-matched healthy participants (n=14). EVs isolated by differential ultracentrifugation were characterised and extracted proteomes were analysed by high-resolution data-independent acquisition liquid chromatography tandem mass spectrometry (DIA-LC-MS/MS). Results: Overall, 6857 proteins were confidently identified in urinary-EVs (q-value≤0.01), including 94 EV marker proteins. Glioblastoma-specific proteomic signatures were determined, and putative urinary-EV biomarkers corresponding to tumour burden and recurrence were identified (FC≥|2|, adjust p-val≤0.05, AUC>0.9). Conclusion: In-depth DIA-LC-MS/MS characterisation of urinary-EVs substantiates urine as a viable source of glioblastoma biomarkers. The promising ‘liquid gold’ biomarker panels described here warrant further investigation.

Project description:N-nitroso compounds (NOCs) can be formed through endogenous nitrosation in the human body and are known to induce micronuclei (MN) formation in vitro. Since lymphocytic MN represent a well-validated biomarker of effect with regard to carcinogenic risk in multiple target organs, establishing a link between NOCs and MN in humans could provide evidence for a carcinogenic risk. Investigating gene expression modulations in relation to NOC exposure could provide further crucial information on underlying molecular mechanisms-of-action. The purpose of this study is therefore to establish the relationship between human NOC exposure under daily life conditions and MN formation in relation to associated transcriptomic changes, using lymphocytes as a surrogate tissue for analyzing carcinogenic events in target organs. We analyzed gene expression levels and MN frequency in lymphocytes from adults participating in a European cohort from the multidisciplinary research project NewGeneris. For assessing exposure to NOCs, urinary samples were analyzed for marker nitrosamines by GC-MS. NOC exposure was subsequently linked to peripheral blood transcriptomics. We found an association between MN frequency and urinary NOCs, indicating that NOC exposure under daily life circumstances may play an important role in human cancer development. Furthermore, we have identified modifications in pathways which indicate a molecular response to NOC-induced genotoxicity. From this, we derived a small group of genes which could be suitable as mechanistic-based transcriptomic biomarkers of NOC exposure-related cancer risk. The modified genetic processes and genes found in this study may be of interest for future investigations into NOC-associated carcinogenicity in humans. The study investigated transcription levels in human whole blood from 30 pregnant mothers in a Danish NewGeneris cohort. For each subject, cRNA copies of isolated mRNA were labeled with one dye (Cy3) and each sample was hybridized on separate arrays. One replicate per subject (so 30 arrays in total).
