Project description:Arterial hypertension is a leading cause of cognitive impairment, attributed to hemodynamic insufficiency, blood-brain barrier disruption, and white matter damage. However, the molecular mechanisms by which hypertension affects brain cells remain unclear. Using scRNA-seq in a mouse model of hypertension induced by angiotensin II, we mapped neocortical transcriptomic changes before (3 days) and after (42 days) onset of neurovascular and cognitive deficits. Surprisingly, evidence of endothelial transport disruption and senescence, stalled oligodendrocyte differentiation, interneuronal hypofunction and network imbalance emerged after just 3 days. By 42 days, when cognitive impairment becomes apparent, deficits in myelination and axonal conduction, as well as neuronal mitochondrial dysfunction, developed. These findings reveal a previously unrecognized early vulnerability of endothelial cells, interneurons, and oligodendrocytes, and provide the molecular bases for the subsequent neurovascular dysfunction and cognitive impairment in hypertension. In addition, the data constitute a valuable resource for future mechanistic studies and therapeutic target validation.

Project description:Nocturnal hypertension is a strong predictor of cardio cerebrovascular disease (CVD), yet its underlying mechanisms remain largely unknown. In this study, we identified CD36 as a key regulator of blood pressure (BP) rhythms and as a gatekeeper of nocturnal hypertension. Its deficiency disrupts circadian BP rhythms.We continuously monitored 48-hour BP using telemetry in CD36 global knockout (KO) mice and renal proximal tubule-specific CD36 KO mice. We also collected serum samples and ambulatory blood pressure monitoring (ABPM) data from patients with nocturnal hypertension to assess the relationship between CD36 expression and nocturnal hypertension. A comprehensive screening of various factors affecting BP and circadian rhythms was conducted to explore the molecular mechanisms involved, utilizing RNA sequencing, calorimetry, enzyme-linked immunosorbent assay (ELISA), co-immunoprecipitation, immunofluorescence assays, and echocardiography. Our telemetry data revealed significantly elevated BP during the daytime (inactive period) in both CD36 global KO and renal proximal tubule specific KO mice compared to their control counterparts. Furthermore, levels of platelet CD36 negatively correlated with the severity of nocturnal hypertension in humans. Mechanistically, CD36 modulates BP rhythms through the AMPK/HDAC5/PER1 pathway: its deficiency activates phosphorylated AMPK, leading to the dissociation of the HDAC5-PER1 complex, enhancing PER1 nuclear accumulation, and desynchronizing the oscillations of intrarenal reninangiotensin-aldosterone system (RAAS) genes and clock-controlled genes (CCGs), ultimately disrupting BP rhythm. Our findings highlight CD36 as a critical integrator that links renal clocks to BP homeostasis, establishing it as a potential predictor and therapeutic target for nocturnal hypertension.

Project description:<p>Background: Hypertension remains a significant global health challenge, with limited treatment efficacy in many patients. Emerging evidence suggests that gut microbiota plays a crucial role in blood pressure regulation. This study investigates the anti-hypertensive effects of Achyranthes bidentata (Ab) extract and its interaction with the gut microbiota, particularly focusing on the enrichment of Dubosiella sp. and its impact on arachidonic acid metabolism.</p><p>Results: Here, we identified a novel microbiota-driven anti-hypertensive mechanism mediated by the Ab extract and Dubosiella sp. Ab significantly ameliorated hypertension and kidney damage by enriching Dubosiella in hypertensive mice. Metabolomics analysis revealed that Ab enhances lipid metabolic pathways, particularly arachidonic acid metabolism. In vitro, Ab significantly promoted the biofilm formation of D. newyorkensis DSM103457. Further gavage experiments confirmed that D. newyorkensis DSM103457 alleviates hypertension, cardiac and kidney injury. Combined microbial and metabolic profiling via 16S rRNA sequencing, metagenomics and metabolomics demonstrated that D. newyorkensis DSM103457 modulates gut microbiota, arachidonic acid metabolism, restoring prostaglandins and leukotrienes essential for cardiovascular homeostasis. Notably, arachidonic acid significantly promotes D. newyorkensis DSM103457 growth, enabling rapid generation and transformation of HETEs and HODEs in vitro.</p><p>Conclusions: This study identified Ab-Dubosiella-arachidonic acid axis as a novel anti-hypertensive pathway, offering promise for the development of integrated herbal prebiotics and microbial probiotics for targeted hypertension therapy.</p>

The transcript profiles of PBMCs from 30 idiopathic pulmonary arterial hypertension patients (IPAH), 19 patients with systemic sclerosis without pulmonary hypertension (SSc), 42 scleroderma-associated PAH patients (SSc-PAH), and 8 patients with SSc complicated by interstitial lung disease and PH (SSC-PH-ILD) were compared to the gene expression profiles of PBMCs from 41 healthy individuals. Gene expression is compared at a global level using total RNA from BPMC for pateints and controls using the Illumina microarray platform.

Project description:<p>Arterial stiffening is a hallmark of early vascular aging (EVA) syndrome and an independent predictor of cardiovascular morbidity and mortality. In this case-control study we sought to identify plasma metabolites associated with EVA syndrome in the setting of hypertension. An untargeted metabolomic approach was used to identify plasma metabolites in an age-, BMI- and sex-matched groups of EVA (n = 79) and non-EVA (n = 73) individuals with hypertension. After raw data processing and filtration, 497 putative compounds were characterized, out of which 4 were identified as lysophosphaditylcholines (LPCs) [LPC (18:2), LPC (16:0), LPC (18:0) and LPC (18:1)]. A main finding of this study shows that identified LPCs were independently associated with EVA status. Although LPCs have been shown previously to be positively associated with inflammation and atherosclerosis, we observed that hypertensive individuals characterized by 4 down-regulated LPCs had 3.8 times higher risk of EVA compared to those with higher LPC levels (OR = 3.8, 95% CI 1.7 – 8.5, P < .001). Our results provide new insights into a metabolomic phenotype of vascular aging and warrants further investigation of negative association of LPCs with EVA status. This study suggests that LPCs are potential candidates to be considered for further evaluation and validation as predictors of EVA in patients with hypertension </p>

Project description:Salt-sensitive hypertension is a common form of hypertension characterized by an exaggerated blood pressure response to dietary salt intake. The molecular mechanisms linking salt-induced hypertension to multi-organ damage remain poorly defined. To address this issue, we generated a longitudinal multi-organ transcriptomic atlas by performing RNA sequencing on the kidney cortex, kidney medulla, liver, and heart across four stages of hypertension in male Dahl salt-sensitive rats. These data provide a valuable resource for identifying organ-specific transcriptional responses involved in the pathogenesis of salt-induced hypertension.
