Volatile Markers in Digestive Cancer

ABSTRACT: The study is aimed to determine the potential of volatile marker testing for identification of gastrointestinal cancers (in particular - colorectal and gastric cancers), the related precancerous lesions in the stomach and colon. The study will be addressing the role of confounding factors, including lifestyle factors, diet, smoking as well as addressing the potential role of microbiota in the composition of exhaled volatile markers.

Volatile communication in plants relies on a KAI2-mediated signaling pathway

Project description:Plants are targets of volatile organic compounds (VOCs) released as a part of plant-plant communication, within-plant self-signaling and plant-microbe interactions. Therefore, understanding VOC perception and downstream signaling is vital for unraveling the mechanisms behind information exchange in plants, which remain largely unexplored. Using the hormone-like function of volatile terpenoids in reproductive organ development as a system with a visual marker for communication, we demonstrated that among the four petunia karrikin-insensitive receptors, PhKAI2ia stereo-specifically perceives the (-)-germacrene D signal, triggering a karrikin-like signaling cascade. This study provides new insights into plant olfaction, uncovers the role(s) of the unique intermediate clade of KAI2 receptors, illuminates the involvement of KAI2ia-dependent signaling pathway in volatile communication and gives insight into the long-standing question about the nature of potential endogenous karrikin-like ligand(s).

Immature leaves are the dominant volatile sensing organs of maize

Project description:Plants perceive herbivory induced volatiles and respond to them by upregulating their defenses. So far, the organs responsible for volatile perception remain poorly described. Here, we show that responsiveness to the herbivory induced green leaf volatile (Z)-3-hexenyl acetate (HAC) in terms of volatile emission, transcriptional regulation and jasmonate defense hormone activation is largely constrained to younger maize leaves. Older leaves are much less sensitive to HAC. In a given leaf, responsiveness to HAC is high at immature developmental stages and drops off rapidly during maturation. Responsiveness to the non-volatile elicitor ZmPep3 shows an opposite pattern, demonstrating that this form of hyposmia (i.e. decreased sense of smell) is not due to a general defect in jasmonate defense signaling in mature leaves. Neither stomatal conductance nor leaf cuticle composition explain the unresponsiveness of older leaves to HAC, suggesting perception mechanisms upstream of jasmonate signaling as driving factors. Finally, we show that hyposmia in older leaves is not restricted to HAC, and extends to the full blend of herbivory induced volatiles. In conclusion, our work identifies immature maize leaves as dominant stress volatile sensing organs. The tight spatiotemporal control of volatile perception may facilitate within-plant defense signaling to protect young leaves, and may allow plants with complex architectures to explore the dynamic odor landscapes at the outer periphery of their shoots

Volatile compounds emitted by Plant Growth Promoting Fungi Tolypocladium inflatum GT22 alleviates copper and pathogen stress

Project description:The intricate interactions between plants and microorganisms have garnered substantial scientific interest. While previous studies have highlighted the potential influence of various fungal volatile compounds(VCs) on plant growth and development, the precise mechanisms underlying this modulation still need to be discovered. In this study, we discovered that fungal volatile organic compounds from the soil-borne fungus Tolypocladium inflatum GT22 enhance the growth of Arabidopsis. Remarkably, following the priming of Arabidopsis with GT22 VC, it displayed an enhanced immune response, thereby mitigating the detrimental effects caused by both pathogenic infections and copper stress. Transcriptomic analyses of Arabidopsis seedlings treated with GT22 VCs revealed the differential expression of 90, 83, and 137 genes after 3, 24, and 48 hours of volatile exposure, respectively. These responsive genes are involved in growth, hormone regulation, defense mechanisms, and signaling pathways. Notably, the induction of genes related to innate immunity, hypoxia, salicylic acid (SA) biosynthesis and camalexin biosynthesis by GT22 VCs were reported. Among the VCs emitted by GT22, limonene is particularly noteworthy. Arabidopsis seedlings exposed to limonene exhibited not only growth promotion effects but also alleviation of copper stress, indicating that limonene may play a role in the interaction between GT22 and plants. Overall, the findings of this study provide evidence supporting that fungal VCs can promote plant growth and enhance both biotic and abiotic tolerance. Furthermore, our results suggest that seedlings exposed to T. inflatum GT22 VCs holds promising potential for harnessing beneficial effects to improve crop productivity.

Radiation responses in peripheral white blood cells of smokers and non-smokers

Project description:Understanding the possible impact of potential confounding factors is necessary for any approach to radiation biodosimetry. Potential confounding factors have not been fully addressed for gene expression-based biodosimetry approaches, such as we are developing. To begin addressing this need, we have used an ex vivo irradiated peripheral blood cell model to investigate the potential effect of smoking on the global radiation gene expression response, and looked for genes that respond to radiation differently in smokers and non-smokers, and also in males and females. The results indicate that only a small number of genes may be significantly confounded by either factor, supporting the idea of developing peripheral blood gene expression strategies for radiation biodosimetry.

Headspace volatile organic compounds from bacteria implicated in ventilator-associated pneumonia analysed by TD-GC/MS

Project description:Ventilator-associated pneumonia (VAP) is a healthcare-acquired infection arising from the invasion of the lower respiratory tract by opportunistic pathogens in ventilated patients. The current method of diagnosis requires the culture of an airway sample such as bronchoalveolar lavage, which is invasive to obtain and may take up to seven days to identify a causal pathogen, or indeed rule out infection. While awaiting results, patients are administered empirical antibiotics; risks of this approach include lack of effect on the causal pathogen, contribution to the development of antibiotic resistance and downstream effects such as increased length of intensive care stay, cost, morbidity and mortality. Specific biomarkers which could identify causal pathogens in a timely manner are needed as they would allow judicious use of the most appropriate antimicrobial therapy. Volatile organic compound (VOC) analysis in exhaled breath is proposed as an alternative due to its non-invasive nature and its potential to provide rapid diagnosis at the patient's bedside. VOCs in exhaled breath originate from exogenous, endogenous, as well as microbial sources. To identify potential markers, VAP-associated pathogens Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Staphylococcus aureus were cultured in both artificial sputum medium and nutrient broth, and their headspaces were sampled and analysed for VOCs. Previously reported volatile markers were identified in this study, including indole and 1-undecene, alongside compounds that are novel to this investigation, cyclopentanone and 1-hexanol. We further investigated media components (substrates) to identify those that are essential for indole and cyclopentanone production, with potential implications for understanding microbial metabolism in the lung.

Project description:In this study, we showed that three bacteria were able to inhibit the mycelial growth of the phytopathogenic fungus Thielaviopsis ethacetica, by the emission of microbial volatile organic compounds (mVOCs). Aiming to understand the molecular mechanisms of these interactions, we evaluated the transcriptomic response of T. ethacetica to the mVOCs produced by one of these bacterial isolates.
