Effect of Annona Muricata Leaves on Colorectal Cancer Patients and Colorectal Cancer Cells

ABSTRACT: Annona muricata L., include the leaves, is found to contain biologically active Annonaceous acetogenins and plant polyphenols that are important components of human diet and a number of them are considered to have chemopreventive and therapeutic properties against cancer. To confirm previous findings in in vitro, animal study and traditionally use, a human, ex vivo and in vitro studies were conducted to evaluate the effects of consecutive ingestion of A. muricata leaves extract for eight weeks.

Project description:Guanabana, known also as Graviola or Annona muricata is a tropical fruit which has been commonly used as complimentary/alternative medicine in Latin American countries. The main compounds in Graviola are the annonaceous acetogenins. These acetogenins have been shown to be selective and toxic against various types of cancer cells in-vitro and in-vivo experimental animal models. In spite of this evidence of anti tumor activity of Graviola, no prospective clinical studies have been carried out to determine if it also has clinical activity.The Investigator have observed two patients at Auxilio Mutuo Cancer Center who experienced significant tumor shrinkage while taking a tea made of Graviola leaves. Neither of these patients were taking any other treatment for their cancer. The investigator propose to conduct a study using guanabana leaves extract in patients with Gastroesophageal junction(GEJ) adenocarcinoma, as well as in Gastric adenocarcinoma, Hepatocellular carcinoma, Pancreatic adenocarcinoma, Low Grade Lymphomas and Colorectal adenocarcinoma.

Healthy adults' blood gene expression who ingested Salacia reticulata plant extract

Project description:In human volunteers, we evaluated changes in gene expression profiles, immunological indices, and intestinal microbiota of blood cells in subjects consuming a S.reticulata extract. Thirty healthy Japanese males were split randomly into a group ingesting 240 mg/day of S.reticulata extract -containing tablets for 4 weeks and a control group ingesting placebo tablets. Ingestion of the S.reticulata extract improved T cell proliferation and other immunological indices, and changed intestinal microbiota, increasing Bifidobacterium and Lactobacillales and decreasing Clostridium bacteria. Expression levels of many immuno-relevant genes were altered. We have shown the S.reticulata extract to enhance human immune functions.

Impact of Orthosiphon stamineus aqueous leave extract on global gene expression of uropathogenic Escherichia coli

Project description:Specific recognition and bacterial adhesion to host cells by uropathogenic E. coli (UPEC) are the first steps towards infection of epithelial tissue of the human urogenital system. Therefore, targeting of UPEC virulence factors, relevant for adhesion, is a promising approach for prevention of recurrent urinary tract infections (UTI). A fully characterized plant-derived aqueous extract from the leaves of Orthosiphon stamineus (OWE), a plant traditionally used in clinical practice in Europe and Asia for UTI, has been shown to exert strong antiadhesive effects under in vitro and in vivo conditions. For improved understanding of the underlying mechanisms transcriptome analysis of OWE-treated UPEC strain UTI89 by Illumina sequencing and cross-validation of these data by qPCR indicated significant down-regulation of bacterial adhesins (curli, type 1-, F1C- and P fimbriae) and of the chaperone-mediated protein folding/unfolding and pilus assembly process; in contrast flagellar and motility-related genes were upregulated. We conclude that OWE transforms the sessile lifestyle of bacteria into a motile one and therefore disables bacterial attachment to the host cell. Additionally, the extract inhibited gene expression of multiple iron acquisition systems (Ent, Fep, Feo, Fhu, Chu, Sit, Ybt) concomitant with an upregulated expression of the ferric uptake regulator (Fur) repressor. The present study explains the antiadhesive and antiinfective effect of the plant extract by pinpointing specific biochemical and molecular targets.

Expression data from micropropagated Vitis vinifera when transferred to ex vitro conditions

Project description:Oxidative stress can arise when in vitro propagated plants developed under low light conditions are exposed to high light during transfer to ex vitro conditions. In such a situation, among the many potential stresses to which the transferred plant can be exposed, oxidative stress is commonly experienced, most likely brought about by absorption of light energy in excess of that required for very low levels of photosynthetic metabolism. In vitro propagated grapevine when transferred to ex vitro conditions with a 4 fold increase in PPFD shows an initial inhibition of PET accompanied by an accumulation of H2O2, suggesting a signal for the upregulation in gene expression and antioxidant enzyme activity, which peaked at 48h after transfer of in vitro grapevine to ex vitro growing conditions. When in vitro propagated plants are exposed to high light upon transfer to ex vitro oxidative stress symptoms occur. To determine whether the underlying pathways activated at the transfer of in vitro grapevine to ex vitro conditions reflect the processes occurring upon light stress we used microarrays. Leaves were harvested from in vitro grown plants immediately prior to transfer to ex vitro condition and 48h after transfer to compare gene expression before and after exposure of these plants to the high light conditions typical of ex vitro growth.

Biofabricated patient-derived mini-colons as next-generation colorectal cancer organoids

Project description:The compromise to reduce, replace, and refine (3Rs) animal usage in cancer research demands the development of biologically capable ex vivo cancer models. The establishment of three-dimensional organoid-based systems has paved the way in this direction by bringing some realistic in vivo features to the in vitro context. However, faithful modeling of cancer requires a degree of tissue-level organization, multi-cellular diversity, biological durability, and experimental flexibility that far exceeds the capabilities of any existing in vitro systems. Here we implemented tissue engineering and microfabrication technologies to develop topobiologically complex patient-specific avatars of colorectal cancer able to overcome those limitations. These miniature tissues consist of long-lived gut-like-shaped healthy colon epithelia (“mini-colons”) that allow the stable integration of cancer cells and their native tumor microenvironment on a platform designed for the real-time high-resolution evaluation of cellular dynamics. This provides an unprecedented repertoire of ex vivo experimental possibilities, which we illustrate through different applications, including the discovery of a cancer associated fibroblast-triggered mechanism driving colorectal cancer invasion. As a whole, our mini-colon system pushes the boundaries of ex vivo cancer research in both basic and pre-clinical settings.

Prognostic Liver Signature profiles of Jc1-infected Huh7.5.1dif cells treated with various drugs

Project description:Hepatocellular carcinoma (HCC) is the second leading cause of cancer death worldwide, indicating urgent need for preventive strategies. Cancer chemoprevention discovery is challenged by the absence of tractable and clinically relevant human model systems and the complex cell circuits supporting carcinogenesis. Here, we developed a simple and robust human liver cell-based system in which persistent hepatitis B/C virus infection or ethanol induce an HCC risk signature robustly predicting long-term HCC risk in cirrhotic patients. Using a computationally enriched small molecule cell-based screen followed by ex/in vivo validation in human liver tissues and animal models, we identified captopril as an HCC chemopreventive agent that antagonizes the EGF/EGFR pathway, a key driver of liver disease and hepatocarcinogenesis. Our system, modeling the cell circuits encoded in the clinical HCC risk signature, enables fast-track cancer chemoprevention discovery and provides an urgently needed approach for improving the dismal prognosis of patients at risk of HCC.

Transcription profiling of human normal and malignant RWPE-1 cells treated with the soybean peptide lunasin reveals it up-regulates tumor suppressor and other chemopreventive genes in prostate epithelial cells

Project description:Numerous studies have identified certain components of soybean as having anticancer properties. Lunasin, a unique 43-amino acid soybean peptide, has been shown to suppress carcinogenesis in mammalian cells and mouse models. It has been hypothesized that lunasin modulates changes in chromatin organization by modifying histone tails, thereby, resulting in the expression of chemopreventive genes. In this report, microarray analysis was used to reveal a novel property of lunasin â?? its ability to up-regulate tumor suppressor and other chemopreventive genes in prostate epithelial cells. The effects of exogenous lunasin on the comprehensive gene expression profiles revealed that a total of 123 genes had a greater than twofold change in expression in the lunasin-treated cells as compared to the mock-treated cells. Of these genes, 121 genes were up-regulated in normal RWPE-1 cells and only two genes were up-regulated in malignant RWPE-2 epithelial cells. The genes up-regulated in RWPE-1 cells include those involved in tumor suppression, apoptosis, and the control of cell division. The results shed new light on lunasinâ??s previously observed chemopreventive properties. We propose that lunasin prevents cancer in vitro and in small animal models by up-regulating the expression of genes that prevent the onset of the disease. Experiment Overall Design: Four microarrays were analyzed. Two samples are RWPE1 human non-tumorigenic prostate epithelial cells and the other two samples are the genetically identical RWPE2 prostate epithelial cancer cells. In each cell line, one of the samples was treated with lunasin at 2 microM for 24 hr.
