Study of S 95005 in Combination With Oxaliplatin in Metastatic Colorectal Cancer

ABSTRACT: The main purpose of this study is to assess the safety and tolerability and to determine the recommended phase 2 dose of S 95005 given in combination with oxaliplatin in patients with metastatic colorectal cancer.

Project description:This study aims to determine the efficacy, safety, and tolerability of the sequential addition of immune-modulating therapy to standard-of-care therapy of microsatellite-stable (MSS)/mismatch repair-proficient (pMMR) metastatic colorectal cancer (mCRC). Patients with MSS/pMMR-mCRC harbor tumor that can be transformed into an immunogenic disease by short-course oxaliplatin-based therapy, and may thereby benefit from the addition of immune-modulating therapy to improve outcome of the current oxaliplatin-based standard-of-care. Primary objective: To determine progression-free survival (PFS), in terms of failure of treatment strategy, of sequential treatment with the Nordic FLOX (5-Fluorouracil, oxaliplatin and leucovorin) regimen and nivolumab compared with the standard-of-care Nordic FLOX regimen in previously untreated MSS/pMMR-mCRC. Secondary objectives: To determine safety and tolerability of sequential treatment with the Nordic FLOX regimen and nivolumab compared with the standard-of-care Nordic FLOX regimen. To monitor and compare quality-of-life (QoL) alterations during therapy courses.

Toxicogenomics profiling of bone marrow from rats treated with topotecan in combination with oxaliplatin: a mechanistic strategy to inform combination toxicity.

Project description:Combinations of anticancer agents may have synergistic anti-tumor effects, but enhanced hematological toxicity often limit their clinical use. We examined whether M-bM-^@M-^\microarray profilesM-bM-^@M-^] could be used to compare early molecular responses following a single dose of agents administered individually with that of the agents administered in a combination. Six patterns of co-expressed genes were detected at the 1-hour time point which indicate regulatory expression of genes dependent on the order of the administration. When topotecan is given first, several signal transduction transcription factors associated with cancer or inactivation of a tumor suppressor were co-regulating gene expression. These results suggest alterations in histone biology, chromatin remodeling, DNA repair, bone regeneration, and respiratory and oxidative phosphorylation are among the prominent pathways modulated in bone marrow from animals treated with an oxaliplatin/topotecan combination. mRNA responses were compared within bone marrow of Sprague-Dawley rats after a single 30-minute treatment with topotecan at 4.7 mg/kg or oxaliplatin at 15 mg/kg alone to that of sequentially administered combination therapy or vehicle control Please note that one of the 'Bone Marrow_Oxaliplatin _Vehicle 1' sample (1344-05H_4M 40) was considered an outlier and thus, was excluded from data analysis and GEO records.

Oxaliplatin induces cancer-like cachexia through distinct transcriptional pathways in mouse muscle and adipose depots

Project description:Cancer cachexia is a multifactorial, largely irreversible metabolic disorder characterized by skeletal muscle and adipose tissue depletion that is responsible for 20% of cancer-related deaths. Oxaliplatin, a platinum-based chemotherapeutic, is a common, first-line treatment for metastatic colorectal and other malignancies that result in cancer cachexia. Yet, it is unknown whether oxaliplatin contributes to these effects independently of cancer. To reveal its role in cachexia development, we administered a chronic human-equivalent oxaliplatin dose to non-tumor-bearing C57BL/6 mice. Daily activity, muscle wasting, fat depletion, and metabolic parameters were monitored through metabolic cages and echo-MRI measurements followed by postmortem muscle and fat tissue histology and bulk RNA seq. Our results show that a high accumulative oxaliplatin dose of 80 mg/kg leads to cachexia-like symptoms, characterized by severe loss of body mass, decreased food intake, diminished ambulatory activity accompanied by lowered core body temperature, decreased skeletal muscle mass, and loss of adipose tissue. RNA-seq data from muscle and adipose tissues indicate a complex mechanism that is in part driven by inflammation and adipokine activity. Comparison of oxaliplatin treatment with external human and murine datasets in cancer and fasting settings illustrates that while oxaliplatin shares many differentially expressed genes with these conditions, it also induces unique gene differentiation. These results suggested oxaliplatin alone induces a cachexia phenotype and thus may aggravate cancer-induced cachexia, highlighting the importance of early diagnostic and therapeutic interventions that target cachexia from both cancer and chemotherapy.

Combination with oxaliplatin improves abscopal effect of oncolytic virotherapy through reorganization of intratumoral macrophages

Project description:Oncolytic virotherapy is a unique and promising immunotherapy. In our prior study, a recombinant oncolytic vaccinia virus carrying IL-7 and IL-12 (hIL-7/mIL-12-VV) showed robust antitumor efficacy in both immunogenic CT26.WT and poorly immunogenic Lewis lung carcinoma (LLC) within the treated tumors. However, the efficacy was limited in virus non-treated (distant) LLC tumors. Here we identified oxaliplatin as a powerful combination partner for an oncolytic vaccinia virus to achieve strong efficacy in the virus-injected tumors and the distant tumors. To understand the molecular mechanism of enhanced antitumor efficacy, we evaluated RNA-sequencing signatures in LLC tumors treated with oxaliplatin alone, hIL-7/mIL-12-VV alone and their combination.

Effect of adenoviral E1A on oxaliplatin response in oxaliplatin-sensitive and resistant human colorectal cancer HCT116 cells

Project description:Resistance to chemotherapy drugs, including oxaliplatin, remains a major challenge in the treatment of colorectal cancer, often leading to treatment failure and poor patient outcomes. Overcoming chemoresistance by sensitizing tumor cells represents a critical therapeutic goal. Adenoviral early region 1A (E1A) has been proposed as a promising gene therapy agent capable of modulating cellular pathways to enhance sensitivity to chemotherapeutic agents. This dataset contains RNA-seq profiling of human colorectal cancer cell lines that are either oxaliplatin-sensitive (HCT116) or oxaliplatin-resistant (HCT116 oxpl-R), treated with oxaliplatin, in the context of doxycycline-induced adenoviral E1A expression. The experiment includes eight conditions: untreated controls, E1A expression alone, oxaliplatin treatment alone, and combined E1A expression with oxaliplatin treatment, each in both sensitive and resistant cell lines. The comprehensive experimental design enables the dataset to be divided into three independent analyses: A) Comparison of baseline transcriptomic profiles between oxaliplatin-sensitive and resistant cells (conditions 1 and 5); B) Investigation of the transcriptional response to oxaliplatin in sensitive and resistant cells (conditions 1, 3, 5, and 7); C) Evaluation of the impact of adenoviral E1A expression on the transcriptomes of sensitive and resistant cells alone (conditions 1, 2, 5, and 6) and in combination with oxaliplatin treatment (all eight conditions); This dataset provides valuable insights into the molecular mechanisms underlying oxaliplatin resistance and the potential role of E1A gene therapy in sensitizing colorectal cancer cells to chemotherapy.
