Latinos CARES (Colorectal Cancer Awareness, Research, Education and Screening)

ABSTRACT: The purpose of this study is to conduct 4 focus groups and recruit for 12 participants per focus group to explore community members’ views about colorectal cancer screening (CRCS) processes and perceived benefits and impediments, what information they believe should be communicated, communication preferences (style, tone) and perceptions of screenings.

Novel stool-based protein biomarkers for improved colorectal cancer screening

Project description:Fecal immunochemical tests (FIT) detecting hemoglobin in stool are widely used for non-invasive colorectal cancer (CRC) screening, but their sensitivity leaves room for improvement. Our aim is to identify novel protein biomarkers in stool that outperform or complement hemoglobin in detecting CRCs and advanced adenomas (AAs). Stool samples (one series of 12 CRCs and 10 controls, and a second series of 81 CRCs, 40 AAs, 43 non-advanced adenomas and 129 controls) were analyzed by mass-spectrometry and searched for human proteins. Classification and regression tree and logistic regression analyses were performed to identify protein combinations that differentiated CRCs and/or AAs from controls. Antibody-based assays for four selected proteins were performed on an independent series of FIT samples (14 CRCs, 16 AAs, 18 non-advanced adenomas and 24 controls) Results: In total, 834 human proteins were identified, of which 29 were significantly enriched in CRCs versus controls in both stool sample series. Combinations of four proteins reached sensitivities of 80% and 45% for detecting CRCs and AAs, at 95% specificity, which was higher than hemoglobin alone.

BAF60/SWP73 Subunits Define Subclasses of SWI/SNF Chromatin Remodelling Complexes in Arabidopsis

Project description:• Evolutionarily conserved SWI-SNF ATP-dependent chromatin remodeling complexes (CRCs) change nucleosome positioning and chromatin states, affecting gene expression to regulate important regulatory processes controlling such as proper development and hormonal signalling pathways. • In our study we used transcript profiling, chromatin immunoprecipitation (ChIP), exhaustive protein-protein interaction study including mass-spectrometry, yeast-two-hybrid and bimolecular fluorescence complementation (BiFC) along with hormone and metabolite profiling, and phenotype assessments to distinguish the functions of Arabidopsis SWP73A and SWP73B subunits in Arabidopsis. • We identified a novel subclass of SWI/SNF chromatin remodelling complexes defined by the presence of SWP73A subunit. Therefore, we propose a refined classification of SWI/SNF CRCs in Arabidopsis, introducing BAS-A (SWP73A-containing) and BAS-B (SWP73B-containing) subclasses. The SWP73A and SWP73B-carrying subclasses of SWI/SNF CRCs exhibit differential properties demonstrated by distinct chromatin binding patterns and divergent effect on hormone biosynthesis and metabolism. We found that SWP73A plays a specific role that cannot be fully compensated by SWP73B. We recognized that some atypical subclasses of SWI/SNF CRCs may be likely formed in mutant lines with inactivated SWP73 subunits. • Our study reveals that the duplication of the SWP73 subunit contributes to unique and shared functions of subclasses of SWI/SNF CRCs in the control of various regulatory processes in Arabidopsis.

scRNAseq of orthotopic mismatch repair deficient (dMMR) or chromosomally instable (CIN) colorectal cancer

Project description:Colorectal cancers (CRCs) deficient in DNA mismatch repair (dMMR) contain abundant CD8+ tumor infiltrating lymphocytes (TILs) responding to the abundant neoantigens from their unstable genomes. Priming of such tumor-targeted TILs first requires recruitment of CD8+ T cells into the tumors, implying that this is an essential prerequisite of successful dMMR anti-tumor immunity. We have discovered that selective recruitment and activation of systemic CD8+ T cells into dMMR CRCs strictly depends on overexpression of CCL5 and CXCL10 due to endogenous activation of cGAS/STING and IFN signaling by damaged DNA. TIL infiltration into orthotopic dMMR CRCs is neoantigen-independent and followed by induction of a resident memory-like phenotype key to the anti-tumor response. CCL5 and CXCL10 could be upregulated by common chemotherapies in all CRCs, indicating that facilitating CD8+ T cell recruitment underlies their efficacy. Induction of CCL5 and CXCL10 thus represents a tractable therapeutic strategy to induce TIL recruitment into CRCs where local priming can be maximized even in neoantigen-poor CRCs.

Project description:Floral organ identities are specified by few transcription factors which act as master regulators. Subsequently, specification of organ axes programs the distribution of distinct tissue types within the organs that themselves develop unique identities. The C-class, AGAMOUS-clade MADS box genes are primary promoters of the gynoecium which is divided into a distal style and a subtending ovary along the apical-basal axis. We show that members of a clade of B3-domain transcription factors, NGATHA1 to NGA4 (NGA1-4), are expressed distally in all lateral organs, and all four have a redundant and essential role in style development. Loss of all four genes results in gynoecia where style is replaced by valve-like projections and a reduction in style-specific SHATTERPROOF1 (SHP1) expression. In agreement, floral misexpression of NGA1 promotes ectopic style and SHP1 expression. STYLISH1, an auxin biosynthesis inducer, conditionally activated NGA genes, which in turn, promoted distal expression of other STY genes in a putative positive feed back loop. Inhibited auxin transport or lack of YABBY1 gene activities resulted in a basally expanded style domain and broader expression of NGA genes. We speculate that early gynoecium factors delimit NGA gene response to an auxin-based signal, elicited by STY gene activity, to restrict the activation of style program to a late and distal carpel domain.
