Low intensity proton beam irradiation downregulates antioxidative protection in Aspergillus nidulans

Project description:The effect of a simulated solar particle event on the fungus Aspergillus nidulans was investigated by exposing surface cultures to a low intensity proton beam (10 pA) and comparing their transcriptomes with those of untreated cultures. Results showed upregulation of DNA repair, ribosome biogenesis, and protein ubiquitination genes, while glycolysis, and respiration genes were downregulated. Surprisingly, antioxidative enzyme genes were bulk-downregulated, increasing susceptibility to oxidative stress induced by menadione sodium bisulfite (MSB). Experiments with an oxidative stress-sensitive mutant and MSB-pretreated cultures suggested that oxidative stress protection was crucial for surviving proton radiation. Therefore, down-regulation of antioxidant enzyme genes was not an adaptive response, but probably a consequence of a disturbance in redox sensing caused by reductive pre-hydrated electrons from water radiolysis. These results shed light on the complex interaction between radiation stress and fungal responses, which could lead to the potential exploitation of the underlying mechanisms in real space environments.

Lymphoma transcriptome analysis after X-ray or Proton beam irradiation

Project description:Proton (PT) therapy represents an alternative to conventional X-ray therapy, and its clinical application for cancer treatment is on the rise due to associated dose deposit advantages. However, our knowledge of biological responses to PT, in comparison to X-ray, remains in its infancy. Identification of PT specific molecular signals is an important opportunity for the discovery of biomarkers and synergistic drugs. We have profiled the transcriptome regulation of lymphoma cells exposed to clinical sources of PT and X-ray radiation, respectively. Subsequent analysis demonstrated both common and radiation-specific deregulation of gene expression. Gene set enrichment discovered pathways unique to PT \ that contribute to the unfolded protein response (UPR) and mitochondrial transport.

Arabidopsis mutant plants with reduced proton pump activity grow normal under ideal growth conditions; however their growth are reduced compared with wildtype plants when placed under the conditions that stress on protonmotive force (high external pH or high external potassium). Distinct vascular genomic effects of proton and gamma radiation

Project description:We performed comparative RNA sequencing of the early (4 hrs) dose response (0.5 – 200 cGy whole body dose, 10 dose levels) of the mouse aorta to proton and gamma radiation. Total-body proton radiation of conscious animals was performed using a proton beam produced by a cyclotron system, while total-body gamma radiation of animals was performed using a Caesium-137 gamma source. A trend analysis identified genes that showed a dose response, using data permutation to estimate a false discovery rate (q-value) for each gene. We identified 29 and 194 genes (q-value ≤ 0.1) that were upregulated with increasing doses of proton and gamma radiation, respectively. No genes were down-regulated. While fewer genes were dose-responsive to proton radiation, the magnitude of the effect was greater than with gamma radiation. These highly responsive genes were enriched for pathways involved in the response to DNA damage, apoptosis, cellular stress and inflammation (p < 0.01). Gamma radiation responsive genes included the same pathways, but extended to genes in vasculature specific pathways. Genes responsive to both radiation types (19 genes at q-value ≤ 0.1) showed almost perfectly superimposable dose-response relationships. We observed the same superimposable dose response relationship of gamma and proton radiations in a subset of genes validated by quantitative PCR not only in the aorta but also in liver, lung, heart and kidney. Despite a relative similar relative biological effectiveness of protons and gamma photons and the activation of canonical radiation response pathways by both radiation types, we detected marked differences in the genomic response. It seems plausible that these genomic differences translate into differences in the biological processes leading to cardiovascular pathologies.
