Changes in Reproductive and Sexual Health in People With Early Onset Colorectal Cancer

ABSTRACT: The purpose of this study is to find out how cancer treatments (chemotherapy and/or radiation therapy) affect reproductive and sexual health in people with early onset colorectal cancer. The study researchers will observe and track changes in hormone levels and in sexual and reproductive health in people with early onset colorectal cancer. This information will help researchers know more about how cancer treatments affect reproductive and sexual health, including the ability to have children (fertility).

Characterization of m6A in domesticated yak testis before and after sexual maturity

Project description:N6-methyladenosine (m6A) is the most common internal modification of eukaryotic mRNA, and its potential regulatory role has recently been identified in mammals, plants, and yeast. However, little is known about the role of m6A methylation in mammalian sexual maturation. In this study, the testicular tissue of yaks before and after sexual maturation was taken as the research object, and the m6A map of testicular tissue of yaks before and after sexual maturation was obtained through the preliminary experiment and MERIP-seq. The results showed that only spermatogonial cells and some primary spermatocytes could be seen in the testicular tissue of yak before sexual maturation, while spermatogenic cells at different stages could be seen in the testicular tissue after sexual maturation. It was found that the methylation level was higher after sexual maturation than before by detecting the expression of methylation related enzymes and the overall methylation level of testis. There were 1438 differential methylation peaks before and after sexual maturation, of which 1226 were significantly up-regulated and 212 were significantly down-regulated after sexual maturation. Through annotation, it was found that the differential methylation peaks were mainly concentrated in the exon region, followed by 3'UTR, and finally 5'UTR. KEGG pathway analysis showed that Homologous recombination, Notch signaling pathway, Growth hormone synthesis and other signaling pathways may be related to testicular development and maturation. m6A modification was positively correlated with mRNA abundance, suggesting that m6A plays a regulatory role in mammalian sexual maturation. In this study, we reported for the first time the m6A transcriptional map of yak testis, providing a starting point for revealing the function of m6A that may affect the development of yak testis.

Sequencing of murine corticotrophs from male and female POMC-GFP mice.

Project description:Stress-related illness represents a major burden on health and society. Understanding the molecular mechanisms underlying stress responses is an important step to understand these diseases, and design treatments. Sexual dimorphism in stress responses have been extensively reported, with depression and most anxiety disorders being more prevalent in women. Corticotrophs of the anterior pituitary play a central role in the generation of hormonal stress responses, by secreting the stress hormone ACTH in response to stressful stimuli. However, their role in contributing to sexually dimorphic responses of the HPA axis is very poorly understood. We performed bulk RNA-sequencing of FACS-sorted corticotrophs from male and female POMC-GFP mice to determine the molecular determinants of sexual dimorphic traits in the activity of these cells, revealing extensive differential gene expression at the transcriptional level. This includes more than 71 mRNAs encoding ion channel subunits, which could be involved in the generation of the sexual dimorphism in their electrical activity.

Developmental and sexual dimorphic atlas of the prenatal mouse external genitalia at the single cell level

Project description:Before the onset of sexual differentiation, male and female external genitalia are morphologically indistinguishable. After the onset of sex-specific production of sex steroids by the gonads, the male and female external genitalia differentiate into either a penis or clitoris. Here we use single cell sequencing to investigate the similarities and differences of cell populations in the external genitalia during the critical sexual differentiation window in the mouse embryo. We found the male and female genitalia are largely similar in cell composition and gene expression at the bipotential stage of development. As sexual differentiation ensues, sex steroid-dependent and -independent differences in cell populations arise, leading to dimorphic establishment of the external genitalia.

Skin Exposure to UVB Light induces a Skin-Brain-Gonad Axis and Sexual Behavior

Project description:Ultraviolet (UV) light affects endocrinological and behavioral aspects of human sexuality via an unknown mechanism. Using a unique male-female comparative approach, we discovered that the sexual behavioral and hormonal features enhanced by UVB are mediated by the skin. In mice, UV exposure increases hypothalamus-pituitary-gonadal axis hormone levels, resulting in enhanced ovary size, extended estrus days, and anti-Mullerian hormone (AMH) expression. It likewise enhances the sexual responsiveness and attractiveness of females and male-female interactions of both males and females. Conditional knockout of p53 specifically in skin keratinocytes abolished UV’s effects. In humans, UV exposure enhanced romantic passion in both genders increased testosterone levels in men. Our data, revealing that UVB triggers a skin-brain-gonadal axis through skin p53 activation, offers therapeutic opportunities for sex-steroid-related dysfunctions. We speculate that during human furless skin evolution, the skin became the front-line regulator of the response to UVB.

Sexual Dysfunction and the Quality of Sexual Life in Patients With Colorectal Cancer and Their Partners.

Project description:In this project the main focus is on assessing sexual functioning and the quality of sexual life after the treatment of colorectal cancer in patients and their partners. Patients and their partners complete questionnaires concerning sexual functioning, quality of life, body image, fatigue, anxiety, depressive symptoms, personality factors, and demographic factors. Questionnaires are completed before surgical treatment, 6 weeks, 3 months, 6 months, and 12 months after diagnosis. The results of this prospective study will give insight in 1) the incidence of sexual problems and the extent patients with colorectal cancer and their partners are bothered by these problems across time, 2) the effect of different treatment modalities on sexual functioning, 3) the relation between sexual problems and quality of life, 4) the determinants of sexual problems and the quality of sexual life adopting the biopsychosocial approach of patients with colorectal cancer who have been treated with surgery, radiation and/or chemotherapy, and more specifically to the role of personality and patient factors and sexual functioning/the quality of sexual life.

Conditional expression of PfAP2-G for controlled massive sexual conversion in Plasmodium falciparum [array]

Project description:Malaria transmission requires the conversion of some asexual parasites into sexual forms termed gametocytes. The initial stages of sexual development, including sexually-committed schizonts and sexual rings, remain poorly characterized, in part because only a subset of parasites undergo sexual development and they are morphologically identical to their asexual counterparts. Here we present a system based on conditional expression of PfAP2-G, the master regulator of sexual conversion, for controlled sexual induction. Induction resulted in ~90% sexual conversion, which enables the characterization of early sexual stages without further purification. Transcriptomic analysis identified the alterations that follow pfap2-g activation, including identification of genes that are down-regulated. We also show that the inducible system enables the characterization of committed stages at the phenotypic level. Altogether, the inducible lines will facilitate the study of the initial stages of sexual development, which is important for the development of new strategies to stop malaria transmission.
