ABSTRACT: Interventions: Group 1: Arm 1 - 0.33 L Anthocyanin-rich blueberry / grape juice daily for 28 days. This is followed by a wash-out period of 10 days and a run-in phase of 7 days. Group 2: Arm 1 - 0.33 L Anthocyanin-reduced blueberry / grape juice daily for 28 days. This is followed by a wash-out period of 10 days and a run-in phase of 7 days. Primary outcome(s): Inhibition of tumor cell migration in vitro after application of plasma-isolated anthocyanins and metabolites. The endpoint is recorded after 28 days compared to day 1. Effects of bedrest and hypoxia on human skeletal muscle miRNA expression

Project description:The present study concerns the effects of 21 days of sustained recumbence (bedrest) and hypoxia, alone and in combination, on skeletal muscle microRNA expression. 14 male subjects participated in 3 experimental campaigns in a counterbalanced fashion: normoxic bedrest (NBR), hypoxic bedrest (HBR) and hypoxic ambulatory confinement (HAMB), both hypoxic conditions with FO2 = 0.141 and PIO2 = 90 ± 0.4 mmHg, equivalent to an altitude of ≈ 4000 m). Each intervention (bedrest or ambulatory confinement) lasted 21 days, and the interventions were separated by a 4-month wash-out/recovery period. The order in which each subject undertook the interventions is denoted 'Intervention_order' Throughout both bedrest interventions, each subject remained in a horizontal position at all times. He was allowed one pillow for the head and to occasionally lean on an elbow while eating or being transferred to a gurney. Muscular exercise was prohibited. During the HAMB confinement, each subject was allowed to move freely within the hypoxic area. To mimic a normal level of physical activity subjects performed 30 minutes of low-to-moderate-intensity exercise twice daily. Muscle samples from Vastus lateralis.

RNA-seq analysis of differential gene expression in liver from Holstein Friesian bulls undergoing diet restriction and compensatory growth

Project description:The objective of this study was to examine changes in hepatic gene expression of growing bulls during a period of dietary energy restriction followed by a period of subsequent realimentation and compensatory growth. Purebred Holstein Friesian bulls (n=20) were assigned to one of two feeding treatments (i) restricted feed allowance for 125 days (n=10) followed by ad libitum access to feed for a further 55 days or (ii) a control group with ad libitum access to feed through out the 180 days trial (n=10). The first 125 days of the trial were denoted as Peirod 1, during which treatment groups were fed differentially. The subsequent 55 days, denoted as Period 2 during which all bulls were fed ad libitum. All bulls received the same diet of 70% concentrate 30% grass silage through out the experimental trial,with the amount of feed provided different dependnet on each treatment group. At the end of each period, 10 animals from each treatment group (10, restricted fed and 10 ad libitum fed) were slaughtered and hepatic tissue collected within thirty minutes of slaughter. RNA was extracted and hepatic gene expression was examined using RNAseq technology and bioinformatic analysis. During the differential feeding period, over-represented pathways including amino acid and lipid metabolism, protein synthesis and cell cycle/proliferation were identified, which indicate alterations in metabolic and growth rate in the liver during dietary restriction . During the realimentation period, pathways involved in cell cycle and cell growth as well as protein synthesis were over-represented, with the direction of fold changes opposite to that of these pathways in Period 1. This information can be exploited in genomic breeding programmes to assist selection of cattle with a greater ability to compensate following a period dietary restriction.

Mytilus galloprovincialis gills exposed to TDCPP

Project description:Mussels (Mytilus galloprovincialis) were exposed during 7 and 28 days in seawater (control), seawater + acetone (solvent control, SC), and 10 micrograms per Litre of tris(1,3-dichloro-2-propyl) phosphate (TDCPP). TDCPP was added from a stock solution prepared in acetone, the volume added being 100 µL per 30 L aquaria. The same volume of acetone was added to SC aquaria. Mussel density was 20 mussels per each 30 L aquaria at the beginning of the experiment and varied between 15 ‒ 20 mussels/30 L aquaria during the 28 days exposure period. Exposure conditions were the following: T = 15.6 ± 0.7 ºC, S = 35.5 ± 0.5 ppt, pH = 7.9 ± 0.1, O2 = 7.9 ± 0.6 mg L-1 and 10:14h light:dark photoperiod. Water was renewed twice per week and mussels were fed before every water renewal with a mixture of phytoplankton representing 1% of mussel tissue dry weight.

Modulation of the post meal PBMC transcriptomic response of shift workers after four weeks of a dietary intervention implementing a dedicated overnight fast

Project description:In a cross-over randomised controlled trial,19-night shift workers with abdominal obesity, completed two diet conditions separated by a two week wash out period; a) the control period = four weeks of habitual eating overnight and b) the intervention period = four weeks of avoiding food intake between 1 AM – 6 AM. At the end of each intervention period participants were given a high fat/high carbohydrate challenge meal in the morning and peripheral blood mononuclear cells were isolated from whole bood sampled at fasting, 2 and 4 hours after eating the meal.

Project description:Fifty 8-week-old C57BL/6J mice were randomly divided into five groups of ten mice. The study duration was 28 days and involved three phases, namely an adaptation phase (days 1M-^V7), a treatment phase (days 8-21), and a washout period (days 22-28). During the 14-days treatment period each mouse in the L. gasseri group, the pre-incubation group and the co-incubation group received a daily dose of 1 x 109 viable K7 cells, determined as colony forming units (cfu) on MRS agar, re-suspended in 100 M-5L of saline solution. The suspension was applied orally by the use of plastic feeding tubes for rodent oral gavage. A single dose of E. coli O157:H7 was applied on day 8 of trial (E. coli group, co-incubation group) and day 15 of trial (pre-incubation group). Each mouse received a dose of 5 x 108 viable E. coli O157:H7 cells, determined as colony forming units (cfu) on E. coli O157:H7 agar, re-suspended in 100 M-5L of saline solution. The suspension was applied intra-gastrically by the use of plastic feeding tubes for rodent oral gavage. Throughout the three phases of the study the animals were fed ad libitum a standard mouse diet and had access to water at all times. All mice were weighted weekly on days 0, 7, 14, 21, 28. Their feed intake was assessed weekly by measuring the remaining (unconsumed) food. Mice were also monitored twice daily for the clinical status and behavioral changes. End of the study period, mice were euthanized with CO2 and blood samples were obtained from the heart by cardiac puncture. For microarray analysis 300ul of blood from 4 mice in each group were transferred immediately after the cardiac puncture to tubes containing a pre-loaded RNA-later solution.

Comparative proteome of a marine dinoflagellate under different temperatures

Project description:Batch cultures of P. donghaiense were maintained in 0.22 filtered and autoclaved seawater (30 psu) with K medium, and grown under cool white fluorescent light at an irradiance of 100 μmol quanta m-2 s-1 with a 14:10 h light: dark cycle at 25 ℃. Tripicated experiments at 19, 22, 25 and 28 ℃ were performed in 2 L acid-washed and autoclaved polycarbonate bottles containing 1.6 L medium. To obtain stable metabolic activity of cells, P. donghaiense at each temperature were cultured and semi-continuously diluted daily for approximately 20 days before samplings.

Strategies to enable large-scale proteomics for reproducible research.

Project description:An experiment was designed to assess the reproducibility of SWATH-MS measurements collected from different mass spectrometers in a single facility over a period spanning approximately four months. Data were acquired with 90-minute gradient lengths at the Australian Cancer Research Foundation International Centre for the Proteome of Human Cancer (ProCan) on six SCIEX TripleTOF 6600 QTOF mass spectrometers. Multiple replicate aliquots were prepared for eight samples. These comprised a dilution series of ovarian cancer tissue (0%, 3.125%, 6.25%, 12.5%, 25% and 50%) offset by yeast and a fixed proportion (50%) of prostate cancer tissue (Samples 1-6), a 1:1 mix of ovarian cancer tissue and yeast cells (Sample 7), and a human cell line (HEK293T; Sample 8). On each mass spectrometer, sets of 20 replicate aliquots (three aliquots of Samples 2-5, and two aliquots of Samples 1, 6-8) were run during each of thirteen 48-hour periods. Experimental data were acquired in 48-hour time periods on each instrument continuously for eight days (with sets of 20 replicates commencing on days 1, 3, 5 and 7), once per week for the remainder of the month (commencing on days 14, 21 and 28), and then once per month for the remainder of the first three months (commencing on days 56 and 84). After each instrument underwent a major clean, the sets of 20 replicates were again run continuously for a further eight days (commencing on days 101, 103, 105 and 107). Data were therefore acquired during a total of thirteen 48-hour periods over approximately four months, during which time the mass spectrometry facility was fully operational. Mass spectrometer maintenance schedules varied according to each individual instrument's performance, and each instrument commenced data acquisition asynchronously within 28 days from the experiment start.
