DO CARCINOMAS OF THE RECTOSIGMOID JUNCTION BELONG TO THE UPPER RECTUM OR SIGMOID COLON?

ABSTRACT: Interventions: Group 1: A retrospective analysis of all patients operated on for a carcinoma in the sigmoid colon, the rectosigmoid junction and the upper rectum between the years 2004 and 2014 will be performed. The objective of this study is to determine whether carcinomas of the rectosigmoid junction have to be considered independently or can be assigned to the upper rectum or to the sigmoid colon taking into account tumor characteristics and the oncological outcome. Primary outcome(s): Patient and tumor characteristics: evaluation of perioperative findings; oncological outcome (tumor recurrence and death); Query by a standardized questionnaire (via mail or telephone interview) or by a query from the local cancer registry (follow-up in the median: 44 months) Study Design: Allocation: ; Masking: ; Control: ; Assignment: ; Study design purpose: other

Peptidomics profiling of human intestine

Project description:The aim of the project is to generate a peptidomics map of gut hormone peptides along the gastrointestinal tract, starting with the stomach and including the duodenum, jejunum, ileum, ascending colon, sigmoid colon and rectum. The tissues would be collected after surgery and the peptide fraction extracted and anlysed by nano LC-MS to identify what peptide hormones are present. These data will then be used to compare against the human transcriptome, and also for comparison against equivent peptides from murine intestinal extracts.

Sigmoid Take-off, New Rectum Definition

Project description:The diversity of definitions for the rectosigmoid junction is becoming a major obstacle to the standardization of optimal treatment of rectal cancers. The aim of this study was to determine the average height of the sigmoid take-off and its association with individual factors. Patients diagnosed with rectal and sigmoid colon cancer in our center from January 2010 to December 2018 were retrospectively enrolled in the cancer group. The results of 200 controls without colorectal disease were also reviewed (normal group). The distance of different landmarks and margins of cancer from the anal verge were retrieved from computed tomography (CT), magnetic resonance imaging (MRI), and endoscopy findings.

Biogeography of the human colorectum

Project description:We applied numerical ecology methods to data produced with a human intestinal tract-specific phylogenetic microarray (the Aus-HIT Chip) to examine the biogeography of mucosa-associated bacteria along the human colon. The microbial DNA associated with matched biopsy tissue samples taken from the cecum, transverse colon, sigmoid colon and rectum of 10 healthy patients was examined. Consistent with previous studies, the profiles revealed a marked inter-subject variability; however, the numerical ecology methods of analysis allowed the subtraction of the subject effect from the data and revealed, for the first time, evidence of a longitudinal gradient for specific microbes along the colorectum: with Streptococcus, Comamonadaceae, Enterococcus and Lactobacillus in greatest abundance at the cecum, with a gradual decline in their relative abundance through to the rectum. Conversely, the analyses suggest that members of the Enterobacteriaceae increase in relative abundance towards the rectum. These differences were validated by quantitative PCR. We were also able to identify significant differences in the profiles, especially for the Streptococci, on the basis of gender. The results derived by these multivariate analyses are biologically intuitive, and suggestive that the biogeography of the colonic mucosa can be monitored for changes via cross-sectional and/or inception cohort studies. 10 patients, 5 males and 5 females. Comprehensive molecular classification for adenocarcinoma of gastroesophageal junction between esophageal and gastric adenocarcinomas

Project description:Background and aims: The aim of this study was to analyze the molecular characteristics of adenocarcinoma of the gastroesophageal junction (AGEJ) compared to esophageal (EAC) and gastric adenocarcinomas (GCFB) from The Cancer Genome Atlas (TCGA) and Seoul National University (SNU) cohorts using next-generation sequencing. Methods: Based on mRNA expression of EAC (n=78) and GCFB (n=102) from the TCGA cohort, a molecular classification model using the Bayesian compound covariate predictor classified AGEJ/cardia (n=48) from the TCGA cohort and AGEJ/upper third gastric adenocarcinoma (n=46) from the SNU cohort into the EAC-like or GCFB-like groups. The molecular characteristics between the EAC-like and GCFB-like groups were compared.
