A preliminary survey on the baseline characteristics and preferences for a multimodal prehabilitation in patients scheduled for elective colorectal cancer resection

ABSTRACT: Interventions: Patients scheduled for bowel resection for colorectal cancer will be recruited. They will be asked to complete questionnaires about their health conditions, fitness levels, psychological status,nutritional status and preferences for a proposed prehabilitation programme assisted by the cancer care co-ordinator. This would require approximately an hour of their time. That would be the end of their participation in the research project. They will not be required to participate in any programme as a part of this project. End of Life Treatment Preferences of Latino Medicare Beneficiaries With Cancer

Project description:Objectives: The primary objective of the research project is to investigate how declining health status influences the end-of-life (EOL) treatment preferences in Latino Medicare beneficiaries with cancer and to examine if and how these preferences are modified by predisposing characteristics (i.e., acculturation, education, medical mistrust, perceived racism) and/or provider-related characteristics (i.e., ethnicity, language used, provision of interpreters, or patient navigators). The specific aims are: 1. To determine if declining functional status influences the end-of-life treatment preferences of older Latino Medicare beneficiaries with cancer. 2. To determine if predisposing characteristics (i.e., acculturation, age, education, medical mistrust, perceived racism) and/or provider-related characteristics (i.e., ethnicity, language used, provision of interpreters, or patient navigators) modify the association between end-of-life treatment preferences and declining health status in older Latino Medicare beneficiaries with cancer. 3. To identify if the end-of-life treatment preferences of Latino Medicare beneficiaries with cancer differ from those of older Medicare beneficiaries without cancer.

Comprehensive profiling of Roquin binding preferences for RNA stem-loops

Project description:Roquin proteins target specific RNA stem-loop motifs with their unique ROQ domain for mRNA degradation. The cellular levels of mRNAs are controlled by these post-transcriptional cis-regulatory stem-loop elements in the 3'-untranslated region (3'-UTR). Here, we first provide an RBNS (RNA Bind-n-Seq) experiment using pre-structured RNA libraries as input. This sRBNS (structured RBNS) was used to map Roquin's RNA binding preferences at nucleotide resolution, revealing preferences for loop sizes and consensus motifs for CDEs and ADEs.

Prehabilitation as a biologically active intervention reprograms pancreatic tumor-immune microenvironment to enhance antitumor immunity

Project description:This dataset contains NanoString GeoMx Digital Spatial Profiling data generated from surgically resected pancreatic ductal adenocarcinoma (PDAC) specimens collected from patients who underwent a multimodal prehabilitation program and matched untreated controls. Spatial transcriptomic profiling was performed across immune (CD45 positive), tumor (PanCK positive), and stromal regions of interest to investigate the biological effects of prehabilitation on the PDAC tumor microenvironment. Differential expression, pathway enrichment, and immune deconvolution analyses were used to evaluate transcriptional changes associated with cytotoxic immune activation, stromal remodeling, and oncogenic signaling. The goal of this exploratory dataset is to provide an initial resource for investigating how prehabilitation may influence immune–tumor interactions in PDAC and to support future studies integrating biological and clinical endpoints.

Decoding RNA binding protein's binding preferences using a general generative model

Project description:Current computational approaches struggle to detect the binding preference of novel or data-scarce RNA binding proteins (RBPs). Here we present Protein Guided Ribonucleic Acid Generator (ProRiboGen), an integrated generative diffusion model that predicts RBPs binding preferences directly from their protein sequences. ProRiboGen leverages insights from protein language models and CLIP-seq interaction data, generating RNA sequences that align with the binding preferences of target RBPs. We demonstrated its capacity for recognizing RNA-binding domains and cross-species generalization ability. Using this approach, we experimentally identified binding motifs for five human and zebrafish RBPs in the absence of CLIP-seq data. Furthermore, its binding site prediction was validated in vivo, revealing a previously uncharacterized intrinsic Cas13d-RNA interaction across the transcriptome, suggesting a potential link to off-target and bystander effects. This study advances our understanding of RBP-RNA interaction and highlights the model’s potential in RNA post-transcriptional regulation.
