A Phase II Clinical Trial of Panitumumab in Combination with Infusional Oxaliplatin and Oral Capecitabine (XELOX) Chemotherapy as 1st line Therapy in Patients with Colorectal Cancer and Advanced Liver Metastases

ABSTRACT: Primary objectives: To assess the objective response rate (ORR) over the entire treatment period when panitumumab is administered in combination with XELOX chemotherapy as 1st-line therapy in patients with previously untreated colorectal cancer and advanced liver metastases. Primary endpoints: Objective Response Rate (ORR) over the entire treatment period (the proportion of subjects in the ITT analysis set with a confirmed complete or partial response). Subjects who do not respond will be considered a non-responder in the analysis.

Project description:We have established epidermal expression of chil-27p::GFP as the marker for activation of oomycete recognition response (ORR) in C. elegans. Through a forward genetic screen, we identified clec-27, clec-35, and ceh-37 to be the key players required for activating chil-27p::GFP upon exposure to the oomycete Myzocytiopsis humicola extract. To determine if activation of entire ORR is affected in these mutants, we analyzed their transcriptome in the presence and absence of extract and compared with that of wildtype C. elegans.

Genomic CpG methylation and TKI-response of the patient with advanced stage lung adenocarcinoma

Project description:The DNA methylation signature involved 87 differentially methylated regions (DMR) that distinguished patients’ response to EGFR-TKIs. The methylation level of HOXB9 (cg13643585) which was negatively correlated with gene expression and annotated as function of transcription factor displayed 88% and 92% of sensitivity with respect to objective response rate (ORR, OR=9.25, p=0.0002) and disease control rate (DCR, OR=6.64, p=0.0009), respectively. Importantly, HOXB9 gained methylation in patients with progressive disease (PD) relative to partial response (PR) patients specific to those who harbored EGFR-activating mutations (ORR, OR=8.8, p=0.0045; DCR, OR=6.63, p=0.018).

Sitravatinib Plus Nivolumab in Patients Undergoing Nephrectomy for Locally Advanced Clear Cell Renal Cell Carcinoma

Project description:Sitravatinib is an immunomodulatory tyrosine kinase inhibitor that can augment responses when combined with programmed death-1 (PD-1) inhibitors such as nivolumab. We report a phase 2 study of neoadjuvant sitravatinib in combination with nivolumab in patients with locally advanced clear cell renal cell carcinoma (ccRCC) prior to curative nephrectomy (NCT03680521). The primary endpoint was objective response rate (ORR) prior to surgery with a null hypothesis ORR = 5% and the alternative hypothesis set at ORR = 30%. Secondary endpoints included correlative immune effects and disease-free survival (DFS). Twenty patients were evaluable for safety and 17 for efficacy. The ORR was 11.8%, and 24-month DFS probability was 88·0% (95% CI 61.0 to 97.0). There were no grade 4/5 treatment-related adverse events. Correlative blood and tissue analyses showed changes in the tumour microenvironment resulting in an immunologically active tumour by the time of surgery. These data suggest that short-term neoadjuvant sitravatinib and nivolumab does not substantially increase ORR but can modulate the immune microenvironment and yield high DFS probabilities in patients with locally advanced ccRCC.

Expression data of fruits in the wildtype and ORR tomato plants

Project description:An Ac/Ds transposon tagged mutant population was screened for changes in visible fruit phenotypes. One line showed orange, yellow sectors in the fruit and was named Orange ripening (Orr), its transposase free offspring showed Mendelian segregation yielding red, yellow and orange fruit bearing plants in a ratio of 1:2:1. Crossing the an orange fruit plant line to the wild-type yielded only plants bearing yellow fruit. A cross between the yellow fruit bearing progeny yielded 26 plants having red and 17 plants having yellow fruit, suggesting an over-dominant allele. Using inverse PCR analysis showed an insertion in the putative subunit M of the tomato Ndh complex. Subsequently, an Orr Ds transposon excision line was recovered which only showed red pigmented fruit. Here, we describe microarray profiling of tomato fruits from wild-type, heterozygous and homozygous Orr insertion plants and from fruits harvested from the Orr excision line. Gene expression analysis of DAC treated AML: high impact or tumor suppressor gene expression changes

Project description:DAC represents a therapeutic option for elderly AML patients. However, there is still a lack of data for valid biomarkers in respect to response. We executed a gene expression analysis prior to treatment to evaluate gene expression patterns associated with response that might be used to predict DAC therapy outcome. In our cohort an objective ORR of 27% was seen. In a class comparison analysis 333 genes were identified that correlated significantly with response. In this gene signature genes that were prior associated with adverse outcome to regular chemotherapy were enriched in the response group. Interistingly for the non response cohort TSG showed an increased expression, suggesting that epigenetic silencing due to promoter hypermethylation might play a lesser role in theses leukemia pathogenesis.

ORR rate is defined as the proportion of patients whose tumor regresses or does not progress under treatment.

A proof-of-concept phase 2 study of the CHK1 inhibitor prexasertib with translational study outcomes in platinum-resistant recurrent, BRCA wild-type high-grade serous ovarian carcinoma

Project description:This proof-of-concept phase 2 trial (NCT02203513) evaluated the clinical activity of the CHK1 inhibitor (CHK1i), prexasertib, in platinum-resistant (PR) recurrent high-grade serous ovarian carcinoma (HGSOC) patients without BRCA mutation. The primary endpoint was objective response rate (ORR). Secondary outcomes were safety and progression-free survival (PFS). Translational research was an exploratory endpoint. Potential biomarkers were investigated using pre-treatment fresh biopsies and serial blood samples. 49 heavily-pretreated patients were enrolled. Among the 39 RECISTv1.1-evaluable patients, ORR was 30.7%, the median PFS was 5.8 months. Toxicity was manageable. Transcriptomic analysis revealed high levels of DNA replication-related genes (POLA1, POLE, GINS3, MCM7) associated with lack of clinical benefit. Subsequent preclinical experiments demonstrated synthetic lethality of POLA1 inhibition in combination with CHK1i in PR-HGSOC cell line models. Therefore, POLA1 expression may be predictive for CHK1i resistance, and the concurrent POLA1 inhibition may improve the efficacy of CHK1i monotherapy in this hard-to-treat population, deserving further investigation.

Identification of genes contributing to a long circadian period in Drosophila melanogaster

Project description:Our objective was to identify candidate genes that contribute to the long 31-hour circadian period previously observed in DGRP_892. We performed transcriptional profiling of whole fly heads from two genotypes: DGRP_892, and Canton-S B, a line with a normal 24-hour circadian period. We collected fly heads every two hours over a 24-hour period. We quantified differential expression among genotype, time, and sex.
