Samples were taken prospectively from 18 patients undergoing surgical resection for intraoral SCC (excluding lip, pharynx and hypopharynx). Biopsies were taken from the tumour and from 5mm beyond each of the abnormal tissue margins defined by WL and NBI. Transcriptional profiling was performed by microarray.

Project description:This study provides a comparison of gene expression in two tomato cultivars, Moneymaker and M82, focusing on first internode and its innermost cell type pith—under two light conditions: white light with far-red supplementation (WL+FR) and the control condition of white light (WL). The samples were harvested at four time points leading to the point where internode elongation could be measured in response to WL+FR. The timepoints were 6, 24, 30, and 48 hours after treatment initiation.

Project description:We have investigated both the response to prolonged darkness and the re-acclimation to M-bM-^@M-^\moderate intensityM-bM-^@M-^] white irradiance (E = 100 M-BM-5mol m-2 s-1) in the diatom Phaeodactylum tricornutum, using an integrated approach involving global transcriptional profiling, pigment analyses, imaging and photo-physiological measurements. The responses were studied during continuous white light, after 48 h of dark treatment and after 0.5 h, 6 h, and 24 h of re-exposure to the initial irradiance. The cultures were incubated at 15C under cool white fluorescent light at a scalar irradiance (EPAR) of ~100 M-BM-5mol m-2 s-1 under continuous white light (CWL) conditions. The cultures were dark-treated for 48 h (DT) before re-exposure to the previous light conditions for 0.5 h, 6 h or 24 h (WL 0.5, 6 and 24 h). Sampling of CWL cultures and DT cultures were performed in addition to the sampling at the WL incubation times of 0.5 h, 6 h, and 24 h. Three biological replicas for each of the five treatments were harvested.

Project description:Chroococcidiopsis thermalis is an extremophilic cyanobacterium that is adaptable to a widerange of environments including deserts, where it can maintain viability after desiccation. This species is also capable of photoautotrophic growth under far-red light (FRL, 750 nm) The proteomic comparison in this study, in conjunction with topographical imaging of thylakoid membranes by atomic force microscopy, supports earlier findings that FRL growth is accompanied by the synthesis of alternative photosynthesis-related proteins. Under white light (WL), a number of phycobilisome (PBS) and photosystem (PS) subunits were predominant at levels 2 - 14-fold greater compared to FRL growth. Two antenna family proteins were markedly more abundant under WL, with a fold-difference of approximately 80. While WL-associated proteins persisted at relatively low abundance under FRL, an exclusive set of PBS and PS subunit isoforms was expressed under FRL. These proteins were undetectable under WL, in many cases encoded in a discrete FRL-associated gene cluster. In line with the expected higher energy capture rate under WL, complexes associated with electron transport (eg. cytochrome b6f) and the ATP synthase were 1.2 – 3.4 times more abundant than under FRL.

Right-sided tumor location was associated with an inferior overall survival and progression-free survival. We aimed to perform a detailed analysis of the diversity in exosomes between left- and right-sided colon cancer using quantitative proteomics. Experimental Design: We isolated exosomes from left- and right-sided colon cancer patients and healthy volunteers and treated colorectal cancer cell line with serum-derived exosomes. Then we performed quantitative proteomics analysis of the serum-derived exosomes and cell line treated with exosomes, respectively. Results: The expression profile of the serum exosome proteome in patients with right-sided colon cancer is different from patients with left-sided colon cancer. Serum-derived exosomes of right-sided colon cancer promote metastasis via up-regulation of extracellular matrix-related proteins, especially proteoglycans like SPARC and glycoprotein like LRG1. Exosomal SPARC and LRG1 were closely correlated with progression-free survival. Conclusions: Proteomic analysis identified different exosomal protein profiling between left- and right-sided colon cancer. Serum-derived exosomes of right-sided colon cancer promote metastasis via overexpression of SPARC and LRG1.

Project description:To study the transcriptomic profile of wt and brc1 mutant axillary buds during the shade avoidance response, we simulated a canopy shade with a low R/FR light ratio. We treated plants with white light supplemented with far-red light (Red light = 29 μeinstein · m-2 seg-1, Far-Red light= 146 μeinstein · m-2 seg-1) for 8 hours. Control plants were left for 8 hours in white light (Red light = 29 μeinstein · m-2 seg-1, Far-Red light= 2.2 μeinstein · m-2 seg-1) . Six biological replicates of 7-8 plants were collected for each genotype and condition (wt WL, wt FR, brc1 WL, brc1 FR). Samples were compared wt WL vs wt FR and brc1 WL vs brc1 FR.
