Project description:Background: Hypertension is one of the most common metabolic diseases in the elderly and its pathogenesis is associated with microbiota dysbiosis. Recent evidence suggests that oral microbiota dysbiosis is also an important factor in the development of hypertension. However, the relationship between hypertension and oral flora in the elderly has not been adequately investigated. Objective: The aim of this cross-sectional study was to investigate the structure of the oral microbiota and its correlation with hypertension in elderly hypertensive patients. To provide new ideas for the prevention and treatment of hypertension. Methods: 206 subjects aged 60 ~ 89 years were selected and divided into normal (CON) and hypertensive (HTN) groups, according to the 2018 Chinese Guidelines for the Management of Hypertension. The oral microbiome composition of saliva samples was determined by 16S rRNA gene sequencing. Results: Although there was no significant difference in α and β diversity between the two groups, systolic and diastolic blood pressure were the most important factors influencing the structure of the oral microbiota. At the phylum level, the relative abundance of the spirochete phylum and the mutualistic bacterial phylum was higher in the HT group than in the CON group (p < 0.05). Diastolic blood pressure was negatively correlated with Streptococcus. Furthermore, we analyzed HTN patients with 120 mmHg<systolic blood pressure<160 mmHg and systolic blood pressure>160 mmHg separately and found that the abundance of Saccharibacteria_(TM7) was significantly increased in the HTN_2 group. Conclusions: Our study identified specific oral microbiota in elderly hypertensive patients, confirming the relationship between oral microbiota and hypertension. This enhances our understanding of the important role of oral microbiota in the pathogenesis of hypertension and accumulates more evidence for microbial involvement in the development of hypertension.

Project description:The objective of this study was to examine changes in hypothalamic gene expression of heifer calves offered varying planes of nutrition during early life. Angus X Holstein-Friesian heifer calves (19 ± 5 days of age, BW: 51.2 ± 7.8 kg, mean ± SD) were offered a high (HP, n=14) or moderate plane of nutrition (MP, n=15) from 3 to 21 weeks of age to achieve a target growth rate of 1.2 kg/d and 0.5 kg/d, respectively. At 21 weeks of age, all calves were euthanized and the hypothalamus of each calf recovered. RNA was extracted and hypothalamic gene expression examined using RNAseq. Compared with calves on the moderate plane of nutrition, a HP of nutrition altered the expression of 39 transcripts in the hypothalamus. Biological pathway analysis revealed these differentially expressed genes to be involved in pathways including acute phase response signalling, STAT3 pathway, and tight junction signalling. Additionally, biological functions affected by early life plane of nutrition included cell-to-cell signalling and interaction, molecular transport, small molecule biochemistry, behaviour, nervous system development and function, nucleic acid metabolism, and cellular function and maintenance.

Project description:Purpose: Nutrition is an important factor that regulates reproductive performance of sheep and affects follicle development. However, the correlation between nutrition and follicle development is poorly understood at molecular level. In order to study its possible molecular mechanisms, we performed expression profiling of granulosa cells isolated from sheep that were fed with different nutrition levels during luteal phase. Methods: To do this, ewes received a maintenance diet (M) and their estrus was synchronized by intravaginal progestogen sponges for 12 days. Ewes were randomly divided into the short-term dietary restricted group (R; 0.5×M) and nutrient-supplemented group (S; 1.5×M). RNA samples were extracted from granulosa cells. Transcriptome libraries from each group were constructed by Illumina Sequencing. Results: In 18,468 detected genes, 170 genes were significantly differentially expressed; of which, 140 genes were up-regulated and 30 genes were down-regulated in group S relative to group R. These genes could be candidates regulating follicular development in sheep. GO, KEGG and Clustering analyses were performed. Conclusions: These data in our study are abundant genomic recourse to expand our understanding of the molecular and cellular events underlying follicle development.

Project description:Recent studies have documented the profound impact of pre-pubertal nutrition on reproductive performance in bulls. Previously, a high plane of nutrition during calfhood (2-30 wk) in beef and dairy bulls hastened puberty explained by an increase in the level of reproductive hormones (LH, testosterone and IGF-I) pre-pubertally, compared to bulls fed less than recommended amounts of energy and protein with no apparent effect on sperm function evaluated by traditional techniques. In addition, upregulated steroid biosynthesis and Sertoli cell maturation was identified in the testicular tissue of high diet bulls at 16 and 24 weeks, respectively. The objective of this study was to evaluate the post- pubertal testes for responses to high (20.0% CP, 67.9% TDN), medium (17.0% CP, 66.0% TDN) and low (12.2% CP, 62.9% TDN) diets fed from 2-30 wk of life. Based on RNA sequencing data, 497 genes were differentially expressed in high vs low diet and 2961 genes in high vs medium diet (P<0.1). According to KEGG analysis, oxidative phosphorylation and ribosome pathways were upregulated in the high diet group (vs medium and low) with majority of the upregulated genes encoding for essential subunits of complex I, III, IV and V of OXYPHOS pathway. In addition, mitochondrial translation, mitotic nuclear division and cell division were enriched in the high vs medium diet group. Supporting the above, the percentage of sperm exhibiting loss of mitochondrial function was lower in the high diet compared to the medium diet (P<0.1). Thus, enhanced early life nutrition upregulated mitochondrial function in both the testes and sperm of post-pubertal Holstein bulls.

Project description:The aim of this study was to investigate the effect of early calf-hood nutrition on the transcriptomic profile of subcutaneous adipose in Holstein-Friesian bulls and possible downstream effects on reproduction. Holstein Friesian bull calves with a mean (±S.D.) age and bodyweight of 19 (±8.2) days and 47.5 (±5.3) kg, respectively, were assigned to either a high (n=10) or low (n=10) plane of nutrition. Calves were fed in order to achieve an overall growth rate of 1.08 and 0.5 kg/day for the high and low plane of nutrition treatments, respectively. At 126 days of age, the bulls were euthanized using an intravenous overdose of sodium pentobarbitone and subcutaneous adipose tissue samples were harvested and RNAseq analysis was performed. At the end of the trial period, there were 745 genes differentially expressed in adipose tissue of bulls on the high plane of nutrition relative to those on the low plane of nutrition (P<0.05; FDR<0.05; fold change >2.0). Plane of nutrition altered the expression of genes involved in energy production and branched chain amino acid degradation. Plane of nutrition was also found to down regulate genes such as leptin and adiponectin which are known to effect reproductive function. This study identifies some of the molecular mechanisms regulated by feeding bulls a high plane of nutrition, which may be incorporated into future breeding programmes for artificial insemination centres.

Project description:Enhanced nutrition during the early calfhood period has been shown to hasten the onset of puberty in bull calves. The hypothalamic-pituitary-ovarian biochemical signalling axis regulates reproductive development in bulls, with the dynamics of gonadotropin pulsatility within the anterior pituitary gland, in particular, central to final sexual maturity. However, the precise molecular mechanisms regulating the influence of metabolic status on this signalling axis in bull calves is yet to be fully elucidated. The objective of this study was to determine the effect of an enhanced plane of nutrition during early life, up to 12 weeks of age, on the anterior pituitary gland proteome of bull calves. Bull calves were offered either a high or moderate plane of nutrition.

Functional analysis of differentially expressed genes revealed functions including organismal development, behaviour, endocrine system development and function, lipid metabolism, molecular transport, small molecule biochemistry, cellular development, cellular growth and proliferation, and nervous system development and function to be affected by early life plane of nutrition in the arucate nucleus of heifer calves.

After 18 weeks of differential feeding all calves were euthanized and skeletal muscle tissue samples collexted from each calf. RNA was extracted and subsequently subjected to RNAseq analysis. 482 genes were significantly differentially expressed between calves on the High plane of nutrition and those on the Moderate plane of nutrition. Biological pathway analysis revealed an enrichement from these genes for processes and pathways related to energy production and cellular growth and proliferation.
