Project description:B7-H3 (CD276), part of the B7 superfamily, has been shown to play an immunomodulatory role, however its regulation, receptor, and mechanism of action remain unclear. Protein levels of B7-H3 have been previously shown to relate to prostate cancer outcomes and currently, humanized monoclonal antibodies are being investigated for clinical therapeutic use (enoblituzumab, MGA271, Macrogenics). Here we use genomic expression data to examine the relationship of B7-H3 to prostate cancer clinical characteristics and molecular profile.

Project description:To understand the changes in the expression of genes in U87MG cells due to ID Family, we analyzed the transcriptome of U87MG cells overexpressing ID Family. As a result, we identify differentially expressed genes patterns so that the signaling mechanism regulated by ID Family in GBM and changes in cells were observed. These mRNA expression profiling analysis results enable an understanding of the signaling mechanism by ID Family in GBM.

Project description:Patients with autoimmune disorders exhibit highly reproducible gene expression profiles in their peripheral blood mononuclear cells. These signatures may result from chronic inflammation, other disease manifestations, or may reflect family resemblance. To test the latter hypothesis, we determined gene expression profiles in unaffected first-degree relatives of individuals with autoimmune disease. Gene expression profiles in unaffected first-degree relatives resembled the profiles found in individuals with autoimmune diseases. A high percentage of differentially expressed genes in unaffected first-degree relatives were previously identified as autoimmune signature genes. Examination of the linear regression relationship of gene transcript levels between parent-offspring pairs revealed that autoimmune signature genes display high levels of family resemblance. Taken together, these results support the hypothesis that these variations in gene transcript levels are associated with family resemblance rather than clinical manifestations of disease. Project description:Naïve T cell activation involves at least two signals from an antigen presenting cell (APC), one through the T cell receptor via interaction with APC peptide-MHC complexes and a second through interaction of CD28 with APC B7 ligands. Following activation, T cells up-regulate a host of other membrane-bound costimulatory molecules which can either promote or inhibit further T cell maturation and proliferation. In some cases, it is necessary to attenuate T cell activation to prevent deleterious inflammation, and inhibitory members of the B7/Butyrophilin family of ligands have evolved to balance the strong stimuli the activating B7 ligands confer. Human genetic association and in vitro studies have implicated one such ligand, BTNL2, in controlling inflammation at mucosal surfaces. Here we show that recombinant mouse BTNL2 modifies B7/CD28 signaling to promote expression of Foxp3, a transcription factor necessary for murine Treg development and function. BTNL2 blocks Akt-mediated inactivation of Foxo1, a transcription factor necessary for Foxp3 expression. Immunophenotyping and gene profiling reveal that BTNL2-induced Treg share many properties with natural Treg, and in vivo they suppress enteritis induced by mouse effector T cells. These findings describe a mechanism by which environmental antigen-driven formation of Treg may be orchestrated by APC expressing specific modulators of costimulatory signals. CD4+CD25- cells isolated from spleen were stimulated on plates coated with anti-mouse CD3 and human IgG (negative control), anti-mouse CD3, rB7-2, and IgG (positive control), and anti-mouse CD3, rB7-2, and rBTNL2 (experimental). Culturing was performed in duplicate for each condition.

These data were used to evaluate differences in GRAMD1B gene expression in whole blood comparing healthy and affected subjects of the family. Functional experiments have been subsequently performed to evaluate the involvement of this gene in multiple sclerosis.

Project description:Oral mucosa has a potential to maintain immunological homeostasis since little severe inflammation happens in the continuous antigen exposure in the oral cavity. SLIT applied for the allergen-specific immune suppression is performed by repeated antigen (Rpt Ag)-painting to sublingual mucosa (SLM). Among a series of immune cells at submucosa, heterogeneous DC/MF subsets contribute to the orchestration of mucosal immune responses. Previously, We found that Rpt Ag-painting to SLM exhausted typical CD11c+ DCs and induced round-type Macrophage-like CD206hi cells. In this study, we defined three major DC/Macrophage fraction in SLM after Rpt Ag-painting by the expressions of several surface molecules. Fraction 1 (Fr-1): CD206- CD11c+ F4/80lo; Fraction 2 (Fr-2): CD206- CD11c- F4/80+ and Fraction 3 (Fr-3): CD206hi CD11clo F4/80+. Microarray analyses revealed that total of 7282 up-regulated and 7389 down-regulated genes in Fr-3 were enriched compared with Fr-2, respectively. Genes in Fr-3 preferentially expressed molecules related to homeostatic process and negative regulation of T cell activation. Furthermore, new B7 family of immune checkpoint molecules express significantly higher on Fr-3. When we checked the expressions of genes related to M2 MF, Fr-3 cells preferentially express fizz1, aldh1a1 and aldh1a2 but not Ym-1 and arginase-1 compared to in vivo induced M2-like cells. These results indicate that Fr-3 has distinct features compared with M2 like cells and may has T cell regulation potential. Functional investigation imply that CD206hi cell dominant status induces suppression of Ag-specific T cell responses. In conclusion, these results suggest that CD206hi cells from SLM involved in immune tolerogenic inducement by expressing new B7 family molecules and modulate T cell immune responses.
