ABSTRACT: Primary outcome(s): The primary outcomes are Overall survival period, relapse free survival period, response rate, Adverse event rate. We are going to investigate the correration the sarcopenia, the rate of skeletal muscle loss and subcutaneous or visceral fat with the primary outcomes.

Eicosapentaenoic and docosahexaenoic acid - enriched high fat diet ameliorates sarcopenia-associated changes in the skeletal muscle transcriptome of C57BL/6J mice

Project description:Effects of EPA/DHA enriched high fat diets (HFD) compared to HFD-Corn oil after 8 weeks intervention on age/obesity induced sarcopenia. Results provides novel information on the signalling pathways regulated by EPA/DHA enriched HFD in delaying the onset of sarcopenia in C57Bl/6J mice compared to HFD-Corn oil. Four mice representing each diet group on the basis of body weight, plasma triglyceride and plasma cholesterol levels were selected for RNA isolation and microarray analysis. Total RNA from skeletal muscle of these selected animals was purified using the RNeasyÂ® Plus Universal Mini kit (Qiagen Nordic, Sollentuna, Sweden) by following the manufacturer?s instruction carefully. The extracted RNA was quantified by NanoDrop 2000c UV-Vis Spectrophotometer (Thermo Scientific, USA) and the quality of the RNA was estimated by RNA 6000 Nano LabChip for Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, California, USA).

Chromatin landscape of human visceral and subcutaneous adipocytes

Project description:Obesity is characterized by the excess of body fat leading to impaired health. Abdominal fat is particularly harmful and is associated with cardiovascular and metabolic diseases and cancer. In contrast, subcutaneous fat is generally considered less detrimental. The mechanisms that establish the cellular characteristics of these distinct fat types in humans are not fully understood. Here, we explored whether differences of their gene regulatory mechanisms can be investigated in vitro. For this purpose, we in vitro differentiated human visceral and subcutaneous pre-adipocytes into mature adipocytes and obtained their gene expression profiles and genome-wide H3K4me3, H3K9me3 and H3K27ac patterns. Subsequently, we compared those data with public gene expression data from visceral and subcutaneous fat tissues. We found that the in vitro differentiated adipocytes show significant differences in their transcriptional landscapes, which correlate with biological pathways that are characteristic for visceral and subcutaneous fat tissues, respectively. Unexpectedly, visceral adipocyte enhancers are rich on motifs for transcription factors involved in the Hippo-YAP pathway, cell growth and inflammation, which are not typically associated with adipocyte function. In contrast, enhancers of subcutaneous adipocytes show enrichment of motifs for common adipogenic transcription factors, such as C/EBP, NFI and PPARgamma, implicating substantially disparate gene regulatory networks in visceral and subcutaneous adipocytes. Consistent with the role in obesity, predominantly the histone modification pattern of visceral adipocytes is linked to obesity-associated diseases. Thus, this work suggests that the properties of visceral and subcutaneous fat tissues can be studied in vitro and provides preliminary insights into their gene regulatory processes.

Chronic circadian misalignment accelerates sarcopenia progression in mice

Project description:In today's 24/7 society, circadian misalignment caused by environmental and lifestyle factors is associated with various adverse health consequences, underscoring the need to understand tissue-specific pathology. A potential association of environmental circadian misalignment with sarcopenia, or accelerated loss of skeletal muscle strength and mass, is poorly documented. Here, we exposed wild-type C57BL/6J male mice to a chronic (64 weeks) jet lag (CJL) paradigm. CJL caused significant reductions in muscle strength and weight relative to untreated mice. Transcriptomic and histological analysis showed activation of TWEAK/Fn14 signaling and reduced myofiber cross-sectional area, hallmark features of sarcopenia. Furthermore, CJL mice showed increased centrally nucleated fibers, myosin heavy chain co-expressing fibers, and myogenic gene expression, suggesting that insufficient muscle regeneration likely contributes to muscle atrophy. These findings demonstrate that circadian misalignment is an independent risk factor for sarcopenia, underscoring circadian rhythms as a key regulator and actionable target for sarcopenia prevention.

Mouse Skeletal Muscle Sarcopenia

Project description:Our study aimed to characterize sarcopenia in C57BL/6J mice using a clinically relevant definition and age range and investigate the underlying mechanisms leading to sarcopenia. Aged male C57BL/6J mice (23-32 months) were classified as non-sarcopenic (no deficit), probable sarcopenic (1 deficit), and sarcopenic (2-3 deficits) based on assessments of grip strength, muscle mass, and treadmill running time and using 2 standard deviations below the mean of the young (4-9 months) as cut-off points. A 9-22% prevalence of sarcopenia was identified in 23-26-month-old mice. Age-related declines in muscle function were more severe than in muscle mass and outcomes of all three assessments were positively correlated in aged mice. As sarcopenia progressed, there were decreases in specific force as well as fiber size and number of IIB skeletal muscle fibers. Mitochondrial biogenesis, oxidative capacity, and AMPK-autophagy signaling decreased significantly while no increase in atrogenes was detected. Our study is the first to characterize sarcopenia in C57BL/6J mice using a clinically relevant definition and highlights the different trajectories of age-related declines in muscle mass and function. These critical insights into the molecular changes associated with sarcopenia progression will facilitate future development of therapeutic interventions.
