Project description:Hypoxia-ischemia (HI) brain damage is one of the most common causes of neonatal brain injuries, amidst other conditions such as intrauterine infection and perinatal cerebral hemorrhage (Bracci et al., 2006). HI, occurring during the perinatal period, severely affects brain integrity resulting in detrimental long-term neurological morbidity in terms of motor, intellectual, educational and neuropsychological performance deficits (e.g. cerebral palsy, mental retardation, learning disability and epilepsy), and even neonatal mortality (Cowan et al., 2003; Ferriero, 2004; van Handel et al., 2007; Shalak and Perlman, 2004). Current therapeutic interventions fail to provide substantial reversal of HI brain injuries and improvement in overall cognitive function. Recent clinical studies demonstrated that post-HI hypothermia provide moderate neuroprotection but fail to show any significant reduction in neonatal morbidity and mortality (Shankaran et al., 2005). We would like to investigate the transcriptional effects of HI on neonatal brain, and if hypoxic pre-conditioning is beneficial to the reduction of brain damage.

Project description:Nr5a2 (also known as liver receptor homolog-1, Lrh-1) has been shown to bind both the proximal enhancer and proximal promoter regions of Pou5f1 and regulate Pou5f1 in the epiblast stage of mouse embryonic development (Gu et al., 2005). Nr5a2-null embryos display a loss of Oct4 expression in the epiblasts (Gu et al., 2005) and die between E6.5 and E7.5 (Gu et al., 2005; Pare et al., 2004). To identify the targets of Nr5a2, we generated a stable ES cell-line that expresses HA-tagged Nr5a2. Anti-HA antibody was used to immunoprecipitate HA-Nr5a2 for ChIP-seq analysis. Project description:One of the most robust behavioural phenotypes in the Fmr1-/ymouse is an increased susceptibility to AGS, which models the sensory hypersensitivity and epileptiform activity seen in FX patients (Berry-Kravis, 2002; Musumeci et al., 1991; Yan et al., 2004; Yan et al., 2005). Multiple therapeutic strategies that alleviate AGS have gone on to be tested in clinical studies, suggesting this assay has predictive value (Liu and Smith, 2014; Michalon et al., 2012; Osterweil et al., 2013; Pacey et al., 2009). Recent studies implicate the inferior colliculus (IC) as a central mediator of the AGS phenotype. The IC is a central hub for auditory processing, relaying information from brainstem nuclei to the auditory cortex and other brain regions (Ayala et al., 2016). In the Fmr1-/ymouse, auditory stimulation causes an enhanced activation of IC neurons as assessed by cFos+ staining and in vivosingle-unit recordings (Nguyen et al., 2020). Moreover, studies using conditional expression of Fmr1in specific neuronal populations show that loss of FMRP in glutamatergic VGlut2+ neurons in the IC is both necessary and sufficient for AGS induction (Gonzalez et al., 2019). We aim to test whether changes in proteasome activity is seen in the Fmr1-/y IC, and if so whether modulating UPS activity could resolve pathological changes.

Project description:Effect of the low phytic acid mutation on gene expression of developing seeds of M955, a low phytic acid barley genotype with >90% reduction in phytic acid in mature seeds. Expression analysis using the Barley1 GeneChip was performed on total RNA from developing seeds (7DAA) of M955 lpa and wt sib-selections. Sib-selections were from BC2 populations of M955 with Harrington as the recurrent parent. Material was grown in field trials in two years 2003 and 2004.
