Project description:The manufacturing processes of commercial probiotic strains may be affected in different ways in the attempt to optimize yield, costs, functionality, or stability, influencing gene expression, protein patterns, or metabolic output. Aim of this work was to compare different samples of a high concentration (450 billion bacteria) multispecies (8 strains) formulation produced at two different manufacturing sites, USA (US) and Italy (IT), by applying functional proteomics analyses. Several protein-profile differences were detected between IT- and US-made products, with Lactobacillus paracasei, Streptococcus thermophilus, and Bifidobacteria being the main affected probiotics/microorganisms. Integration of omics platforms with in vivo analysis emerged as a powerful tool to assess manufacturing procedures.

Project description:Using Affymetrix data analysis, important signalling pathways and transcription factors relevant to gut inflammation and anti-inflammatory action of probiotics were identified using the clinically validated probiotic VSL#3 and the IL10-knockout mouse, an animal model for inflammatory bowel disease. VSL#3 increased expression of genes involved in PPAR signalling and metabolism of xenobiotics and decreased expression of genes involved in immune response/inflammatory response.

Project description:In animal production, the use of probiotics supplements to promote animal health is increasing. The objective of this study was to assess the impact of probiotics administration on global gene expression in dairy cows. Lactating Holstein-Friesian cows (n=10) from the North Carolina Agricultural and Technical State University dairy herd were used for the study. Treatment was a 50 ml oral drench of FASTtrak microbial pack (Probiotics) (Conklin Company, Kansas City, MO) at the recommended dose in sterile endotoxin-free water or sterile endotoxin-free water only (control). This treatment was carried out for 60 days. Whole blood was collected at the beginning (Day 0) and end of the study (Day 60) for microarray analysis. We employed microarray expression profiling as a discovery platform to identify genes with potential association with probiotics supplementation in cows. Gene expression analysis identified 10,859 differentially expressed genes- 1168 upregulated genes and 9691 downregulated gene. Results for pathway analysis showed significant pathways associated with innate immunity such as the Toll-like receptor (TLR) pathway, inflammation response and Wingless (Wnt) signaling pathway. Real-time PCR was used to validate the expression of the Wnt signaling pathway and immune response genes. Probiotic treatment impacted global gene expression, and particularly, the expression of immune response and Wnt signaling pathway genes. Oral administration of probiotics to dairy cows impacts global gene expression and particularly the expression of innate immune genes in dairy cows.

Project description:Obesity and overweight are closely related to diet, and gut microbiota play an important role in body weight and human health. The aim of this study was to explore how Lactobacillus curvatus HY7601 and Lactobacillus plantarum KY1032 supplementation alleviate obesity by modulating the human gut microbiome. A randomized, double-blind, placebo-controlled study was conducted on 72 overweight individuals. Over a 12-week period, probiotic groups consumed 5×10^9 colony-forming units of HY7601 and KY1032), whereas the placebo group consumed the same product without probiotics. After treatment, the probiotic group displayed a reduction in body weight (p <0.001), visceral fat mass (p <0.025), and waist circumference (p <0.007), and an increase in adiponectin (p <0.046), compared with the placebo group. Additionally, HY7601 and KY1032 supplementation modulated bacterial gut microbiota characteristics and beta diversity by increasing Bifidobacteriaceae and Akkermansiaceae, and decreasing Prevotellaceae and Selenomonadaceae. In summary, HY7601 and KY1032 probiotics exert anti-obesity effects by regulating the gut microbiota; hence, they have therapeutic potential for preventing or alleviating obesity and overweight.

Project description:To comprehensively characterize the intrinsic molecular effects of ADs independently of the specifics of particular disease models, we generated 300 transcriptomic profiles from 10 brain regions in naive rats. Three classical tricyclic antidepressants, Bupropion, desipramine, fluoxetine, and a probiotic formulation (Lacidofil®) can massively alter gene expression in most regions [>1,500 differentially expressed genes (DEGs)], suggesting that traditional single-gene or single-pathway analyses may under-represent the complexity of molecular responses. Using spatial transcriptomics in the prefrontal cortex from an independent set of animals, we further delineated DEGs and molecular pathways that exhibited layer-specific differential regulation by probiotics and fluoxetine.

Project description:The hypocholesterolemic effect of probiotics has been observed, but the molecular mechanism of probiotic-host interaction is still obscure. In this study, DNA microarray technology was used to explore the gene expression profile of liver of hypercholesterolemic rats caused by administration of probiotic Lactobacillus casei Zhang, which can decrease the serum triglyceride, low-density lipoprotein cholesterol, hepatic cholesterol and triglyceride of hypercholesterolemic rats. Six liver samples in high fat and probiotic treated group (3 samples in each group) were randomly selected for RNA isolation and microarray hybridization, the 3 samples in high fat group were used as control.

Blood samples were collected at the end of the study from probiotics-treated and control animals for RNA extraction and microarray analysis. In vitro effect of lipopolysaccharide (LPS) endotoxin treatment was evaluated using blood samples collected from probiotics-treated animals (Day 60 samples) to serve as positive control array. A pooled sample was generated by taking equal concentration of RNA from experimental animals in each group. Pooled samples from each group was hybridized on Agilent one color bovine v2 bovine (v2) 4x44KÂ array slides.

Project description:Probiotics have shown promise in positively altering gut microbiota and can potentially improve the gut flora of individuals with obesity. Recently, the development of probiotics with “Pharmabiotic” properties, which can reduce body fat and inhibit lipid accumulation, has emerged as a notable approach for effectively combating obesity. Nevertheless, owing to the lack of a universal methodology for elucidating the molecular mechanisms of probiotics, their antiobesity effects remain largely unknown. Herein, we developed an advanced multiomics-based strategy to decipher the mechanisms by which probiotics and their derivatives curtail adipocyte lipid production to affirm their antiobesity potential. Our initial investigation assessed the impact of probiotics and their derivatives on adipocyte differentiation and lipid generation at defined differentiation stages. Leveraging these insights, we performed comprehensive multiomics analyses at selected intervals to deepen our understanding regarding the suppression mechanisms of lipid formation. This framework confirmed the antiobesity efficacy of Lactobacillus reuteri lysate, targeting early differentiation to impede branched-chain amino acid (BCAA) catabolism and reduce adipocyte lipid accumulation. Specifically, L. reuteri lysate suppressed Krüppel-like factor 5 expression in early adipocyte differentiation phases, downregulating peroxisome proliferator–activated receptor gamma expression and reducing BCAA catabolism. Concurrently, L. reuteri lysate enhanced hypoxia-inducible factor 1 alpha expression, consequently downregulating lipin-1 expression in initial adipocyte differentiation stages, thus inhibiting adipogenesis. This study underscores the efficacy of our strategy in elucidating the intricate causal dynamics between host and microbiome, advancing therapeutic development and target exploration of probiotics.
