Evaluation of Tumour Growth and Oncological Treatment in Patients With CRLM Using Zebra Fish Embryo Model

ABSTRACT: In order to improve and individualize cancer treatment, personalized treatment needs to be developed much further. Liver metastasizing colorectal cancer is treated with a combination of oncological and surgical interventions. The selection of chemotherapy is today mainly done according to best guess. Today only a small fraction of oncological treatment may be known to be effective in a person before treatment start, most often it is trial and error. A fast reliable system for looking at response to different treatments in each unique patient is much needed and would, if successful, completely change the way we give oncological treatment today. Patient’s tumor tissue will be evaluated with use of zebrafish embryo avatars to evaluate tumour growth and response to different combinations of chemotherapy. If successful interventional studies are planned.

Translation Error Clusters Induced by Aminoglycoside Antibiotics

Project description:Aminoglycoside antibiotics (AGAs) target the ribosome and induce mistranslation, yet which translation errors induce bacterial cell death is unclear. The analysis of cellular proteins by quantitative mass spectrometry shows that bactericidal AGAs induce not only single translation errors, but also clusters of errors in full-length proteins in vivo with as much as four amino acid substitutions in a row. The downstream errors in a cluster are much more frequent than the first error (0.4 vs. 10-3, respectively) and independent of the intracellular AGA concentration. The prevalence, length, and composition of error clusters depends not only on the misreading propensity of a given AGA, but also on its ability to inhibit ribosome translocation along the mRNA. Error clusters constitute a new class of misreading events in vivo that may constitute the predominant source of proteotoxic stress at low AGA concentration, which is particularly important for the autocatalytic uptake of the drugs.

Investigating the Mechanisms of Silver Nanoparticle Toxicity in Daphnia magna: A Multi-Omics Approach

Project description:Silver nanoparticles (AgNPs), one of the most common nanomaterials in use today, have been shown to enter the aquatic environment from use of commercially obtained products. Although acute toxicity has been shown in vitro and in vivo, there is still a great deal of uncertainty surrounding the molecular mechanism of AgNP-induced toxicity. Much of the current literature centres on the question of whether toxicity is induced by the AgNPs themselves or by ionic silver released from the nanoparticle surface. Here, I present research investigating AgNP toxicity to Daphnia magna, a keystone species of freshwater ecosystems the world over, using a multi-omics platform of transcriptomics and metabolomics techniques. Initially, the AgNPs were fully characterised and the exposure conditions optimised to accurately assess the impact of AgNP exposure to D. magna. This allowed for the design of sub-lethal AgNP exposures that had been fully controlled for the fraction of dissolved silver ions. Secondly, using this multi-omics platform, the role of Ag+ ions in AgNP toxicity to D. magna was determined. Finally a metabolic pathway perturbed by AgNP exposure was identified, as well as two constituent metabolites of key importance. Overall, this research advances the mechanistic understanding of AgNP toxicity to aquatic organisms, and further highlights the need for greater consideration of the nanoscale in regulative legislature.

The adjuvants studied, CpG-ODN and α-GalCer have previously been shown to be potent mucosal adjuvants in mice when administrered together with a glycoprotein from HSV-2.

Gene expression profiling of white bass when fed fish meal or fish meal replacement diets

Project description:White bass (Morone chrysops) are a popular sportfish throughout the southern United States, and one parent of the commercially successful hybrid striped bass (M. chrysops x M. saxatilis). Currently, white bass are cultured using diets formulated for other carnivorous fish, such as largemouth bass (Micropterus salmoides) or hybrid striped bass and contain a significant percentage of marine fish meal. Since there are no studies regarding the utilization of alternative proteins in this species, we evaluated global gene expression of white bass fed diets in which fish meal was partially or totally replaced by various combinations of soybean meal, poultry by-product meal, canola meal, soy protein concentrate, wheat gluten, or a commercial protein blend (Pro-Cision). Significant differential expressed genes and gene ontology of pairwise comparisons between control diet and each test diet are presented and discussed.

Towards Single Bacterium Proteomics

Project description:Bacteria are orders of magnitude smaller than mammalian cells, and while single cell proteomics (SCP) detects and quantifies today several thousands of proteins per mammalian cell, it is not at all clear whether conventional SCP methods will be suitable for bacteria. Here we report on the first successful attempt to detect proteins from individual Escherichia coli bacteria, with validation of our findings by comparison with bulk proteomics data.
