Project description:Interventions: split-dose PEG-ELS combination with Lubiprostone 3 times by specifying as follows; 2 days before colonoscopy 1 capsule of Lubiprostone at 6.00 a.m. then took 1 capsule of Lubiprostone at 6 p.m. Day before colonoscopy - 1 capsule of Lubiprostone at 6.00 a.m. The subjects would receive 1 sachet of PEG-ELS mixed with 2 liters of water, which the patients had to take all within 2 hours at 6:00 p.m. Lastly, on the colonoscopy day, the subjects received 1 sachet of PEG-ELS mixed with 2 liters of water and took it all in 2 hours at 4.00 a.m. on the day of the colonoscopy.,split-dose PEG-ELS before colonoscopy day, patients had to take 1 sachet of PEG-ELS with 2 Liter of water at 6 p.m. and 1 sachet of PEG-ELS with 2 Liter of water at 4 a.m. on the day of the colonoscopy.;Active Comparator Drug,Placebo Comparator Drug;split-dose PEG-ELS combination with Lubiprostone 3 times ,split-dose PEG-ELS combination Primary outcome(s): the Ottawa bowel preparation score Colonoscopy day proportion of adequate bowel cleansing Study Design: Randomized

Transcription profiling by array of mouse intestinal epithelial m-ICcl2 cells stimulated with lipopolysaccharide

Project description:m-ICcl2 cells (Bens et al., Am. J. Physiol., 1996) were grown for 6 days on collagen coated cell culture flasks in medium as recommended (see ference above). Medium was changed every 48 hours. The cells were mycoplasma tested (PCR) and found negative. The medium and PBS was endotoxin tested (ELISA) and found to be negative (< 50pg/mL). Unstimulated cells (control) were left cultured in medium only. Lipopolysaccharide (100 ng/mL in water, D31m4 E. coli LPS from List Biochemicals) was sonicated (10 min_ prior to use. Cells were stimulated 6 hours with 100 ng/mL LPS and harvested or stimulated with LPS (100 ng/mL) and washed with warm phosphate buffered saline (PBS) after 6 hours, followed by medium change every 48 hours. Cells were harvested after 96 hours (6 hours incubated in the presence of LPS, 90 hours after stimulation). RNA was isolated after repeated (3x) washing with PBS using Trizol reagent (1 mL/75 cm2) using the standard protocol and resuspended in Millipore water. Quantification was done by spectroscopy and the RNA was stored at -80C.

Genotoxicity of nitrosamines

Project description:We profiled the gene expression in eight chicken embryo livers in the 11th day of incubation. The eggs were injected on postincubation days 9, 10 and 11 with 4mg activation-dependent DNA reactive diethylnitrosamine (DEN) or its structural non-carcinogenic comparator, N-nitrosodiethanolamine (NDELA)DEN in 0.15 ml/egg deionized water. We found that both compounds induced simmilar massive transcriptomic alterations in the xenobioatic metabolism pathways when comparing with reference data in eggs injected with the vehicle only (deionized water).

RNA-BisSeq for YBX1 RIP in normal T24 cell

Project description:Flag-YBX1 overexpressed T24 cells pellets were resuspended with 2 volume of lysis buffer (150 mM KCl, 10 mM HEPES pH 7.6, 2 mM EDTA, 0.5% NP-40, 0.5 mM DTT, 1:100 protease inhibitor cocktail, 400 U/ml RNase inhibitor), and incubated at 4 °C for 30 min with rotation. Then the lysate was centrifuged at 15 000 g for 20 min. Before incubating the lysate with Flag beads, 100ul were taken as input. The anti-Flag M2 magnetic beads (Sigma, 10 μl per mg lysate) were washed with NT2 buffer (200 mM NaCl, 50 mM HEPES pH 7.6, 2 mM EDTA, 0.05% NP-40, 0.5 mM DTT, 200 U/ml RNase inhibitor) four times. Cell lysate was mixed with M2 beads and incubated at 4 °C for 4 h with rotation. The beads were washed two times with 1 ml ice-cold NT2 buffer. Then the beads were subject to Micrococal nuclease (NEB) digestion (1:1 000 000 dilution) for 8 min at 37 °C. The beads were cooled on ice immediately for 5 min and washed two times with 1 ml ice-cold 1× PNK+EGTA buffer (50 mM Tris-HCl pH 7.5, 20 mM EDTA, 0.05% NP-40, 200 U/ml RNase inhibitor) and two times with 1 ml ice-cold 1× PK buffer (50 mM NaCl, 100 mM Tris-HCl pH 7.5, 10 mM EDTA, 0.2% SDS, 200 U/ml RNase inhibitor). Then the beads were digested with 200 μl pre-heated (20 min at 50 °C) Proteinase K and RNAs were extracted with an equal volume of Acid-Phenol: Chloroform, pH 4.5 (Ambion). The RNAs were subjected to rRNA removal and Bisseq. The libraries were sequenced on the Illumina HiSeq X-Ten platform at Novogene (Tianjin, CA) with paired-end 150 bp read length.The m5C sites were called using meRanCall from meRanTK (FDR < 0.01).

A clinical trial to study the effects of coffee and chewing gum on return of normal bowel function after undergoing surgery for malignant tumors of female reproductive system.

Project description:Intervention1: Chewing gum: sugarless chewing gums to chew 3 times daily at 15:00, 19:00, and, 10.00 hours until initiation of solid diet or, for 2 days whichever is earlier, starting from the day of surgery itself, each session of chewing to last 30 minutes. Intervention2: Coffee: Coffee will be dispensed from a single vending machine using same brand of coffee capsules, containing fixed and known proportion of caffeine and other ingredients : to drink three cups of caffeinated coffee daily (100 mL at 08:00, 12:00, and 16:00), beginning on Post-operative day (POD) 1. Patients have to drink the entire 100 mL amounts within 20 min under the supervision of a nursing officer. This will be contined for 2 days or upto initiatiation of solid diet, whichever is earlier. Control Intervention1: nil intervention: This group will receive post-operative care in form of hydration, antibiotics, anti-emetics, thrombo-prophylaxis, ambulation as per department protocol.
