Analysis of melanosomal, exosomal and cellular miRNAs of melanoma cells

Project description:miRNA microarray profiling was performed for exosomes and secreted melanosomes of MNT-1 cells as well as WM3682 and WM3314 cells and melanosomes isolated from cell homogenates. The aim of this study is to compare the miRNA content of exosomes and secreted melanosomes and to assess differences in the miRNA profiles of different melanoma cell lines and their melanosomes. Total RNA was extracted from MNT-1 exosomes and secreted melanosomes (2 replicates each) that were isolated from the conditioned medium of MNT-1 melanoma cells by differential ultracentrifugation. In addition, total RNA was isolated from WM3682 and WM3314 melanoma cells and melanosomes (1 replicate each). These latter melanosomes were isolated from cell homogenates by filtration through a 0.45 Âµm filter, followed by FACS sorting. Transcriptome profiles derived from parallel venipuncture, dried blood spot (DBS), and saliva samples

Project description:Comparison of transcript abundance estimates derived from DBS vs saliva vs gold standard peripheral blood mononuclear cell (PBMC) samples. Gene expression profiling was conducted using parallel collection and assay of venipuncture blood samples (PBMC), DBS samples, and saliva. Data come from 58 community dwelling adults recruited from the Los Angeles metropolitan area. Analyzed data include basic demographic characteristics (age, sex, race/ethnicity) and body mass index (BMI) as well as RNA indicators of major leukocyte subset prevalence (i.e., estimated abundance of mRNA for CD3, CD4, CD8, CD19, FCGR3A/CD16, NCAM1/CD56, and CD14; all provided here as log2-transformed values of normalized gene expression data). Categorial variables were coded 0=no/absent and 1=yes/present.

A serum-induced transcriptome and serum cytokine signature obtained at diagnosis correlates with the development of early pancreatic ductal adenocarcinoma metastasis

Project description:Immune indicators of PDAC progression were obtained from peripheral blood and analyzed in the context of each other by calculating a serum-induced transcriptome index and a serum cytokine index. Significant correlation of the transcriptome index with the inflammatory transcriptome index suggest that inflammtion has a role in PDAC progression. The segregation of metastatic progressed and not progressed patients when the inflammatory transcriptome index was analyzed in the context of the serum cytokine index suggest that the serum cytokine index may be a systemic biomarker of PDAC progression . Our data illustrates both the transcriptome and cytokine indices have promise as biomarkers of cancer immunity.
