Significance of Immunohistochemical Expression of Fascin-1 in Colorectal Carcinoma

ABSTRACT: Colorectal carcinoma is a major health problem. As a malignant tumor, the malignant potential of colorectal carcinoma is based mainly on its ability to metastasis to different sites. Fascin-1 is an actin binding protein which is involved in reconstruction of intracellular actin network, the latter enforces the neoplastic cell to invade surrounding structures.

Significance of Cortactin Expression in Colorectal Adenocarcinomas

Project description:Colorectal carcinoma is one of the most aggressive malignant epithelial neoplasms affecting the gastrointestinal tract. The incidence of colorectal carcinoma is obviously increasing in developing countries, where the physical inactivity and the consumption of animal fat-rich food became more evident. Colorectal tumorigenesis is a multistep process which is initiated by adenoma and is terminated by carcinoma, the latter shows variable degrees of tumor differentiation and invasiveness. During the adenoma-carcinoma process; a series of genetic mutations occur. Detection of these genetic mutations will help in the development of novel therapeutic agents, which in turn will improve patients’ outcomes. Cortactin (CTTN) is a Src kinase substrate, encoded by a gene located on chromosome 11. CTTN binds to and activates Arp 2/3 and stabilizes the dynamic actin assembly after its formation. So, it become clear that CTTN is involved in the formation of the leading-edges cellular protrusions.

Tobacco smoking transcriptional imprinting contributes to urothelial cancer

Project description:Smoking is a major risk factor for Urothelial carcinoma (UC). However the complex mechanisms, how smoking promotes carcinogenesis and tumour progression, remain obscure. A microarray based approached was therefore performed to detect the smoking derived gene expression alteration in non-malignant and malignant urothelial tissues from patients with superficial or invasive UC. Smoking enhanced cell migration and response to tissue damages. In non-malignant tissues smoking induced immune response and altered the cytoskeleton. In urothelial carcinoma, smoking altered extracellular and chromosome structures. Smoking affected tissues from patients with invasive carcinomamore strongly, up-regulating particularly growth factors and oncogenes in non-malignant tissue of patients with invasive but not with superficial carcinoma. In former smokers, comparable changes were seen in tissues form patients with invasive disease while they were minor or reversed in tissue of patients with superficial disease. Best but not complete tissue repair was suggestedfor non-malignant tissue from patients with superficial tumours. we used microarray techniques to define gene expression profile which may explain the contribution of smoking to urothelial carcinoma. Gene expression profiling using GeneChip HG-U133A Plus 2.0 (Affymetrix) was performed on non-malignant and carcinogenic urothelium from six patients with superficial and six with invasive urothelial carcinoma, each group including two current, former, and non-smokers.

Transcription profiling of breast tumors and surrounding tissue to detect pre-malignant histological and genetic changes

Project description:The aim of our study is to examine the region of tissue surrounding or adjacent to breast tumor in order to detect pre-malignant histological and genetic changes that may be predictive of risk of breast cancer. In this study, histological and gene expression data of breast tissue were obtained from patients that underwent mastectomy at various stages of breast carcinoma. For each patient, samples were obtained from the tumor region and four surrounding zones (1cm, 2cm, 3 cm and 4 cm away from the grossly visible tumor boundary), where possible. We have also breast samples from healthy individuals undergoing breast reduction surgery (mammoplasty). Microarray gene expression profiles of tissue samples (tumor and surrounding tissue, and breast reduction mammoplasty) was performed. In addition pathologist reviewed all slides to provide a details histology of all samples (excluding breast mammoplasty)

Identification of novel biomarker candidates for the immunohistochemical diagnosis of cholangiocellular carcinoma - LC-MS Part

Project description:The aim of this study was the identification of biomarkers for the immunohistochemical diagnosis of cholangiocellular carcinoma (CCC). CCC is a primary cancer which arises from the epithelial cells of the bile ducts and is characterised by high mortality rates due to its late clinical presentation and limited treatment options. Tumorous tissue and adjacent non-tumorous liver tissue from eight CCC patients were analysed by two-dimensional differential in-gel electrophoresis (2D-DIGE) and by mass spectrometry-based label-free proteomics. After data analysis and statistical evaluation of the proteins which were found to be differentially regulated between the two experimental groups (fold change ≥ 1.5; p-value ≤ 0.05), 15 candidate proteins were chosen for verification by immunohistochemistry in an independent cohort of 14 patients. This confirmed the significant up-regulation of chloride intracellular channel protein 1 (CLIC1), gelsolin (GSN), pyruvate kinase isozymes M1/M2 (PKM2), serpin H1, 14-3-3 protein sigma (SFN), stress-induced phosphoprotein 1 (STIP1) and tax1-binding protein 3 (Tax1BP3) in tumorous cholangiocytes when compared to normal hepatocytes, whereas fatty acid-binding protein 1 (FABP1) and Betaine-homocysteine S-methyltransferase 1 (BHMT) were significantly down-regulated. Furthermore, STIP1 and SFN were able to differentiate between CCC tumour cells and non-tumorous cholangiocytes with a sensitivity of 100% and 86%, respectively and a specificity of 100% in both cases. We therefore conclude that these proteins should be considered as potential diagnostic biomarkers for CCC in an immunohistochemical application. Additional experiments addressing the validation of these novel biomarker candidates are being performed.

Project description:Colorectal carcinoma (CRC) is the third most common malignancy worldwide. In Egypt, the relative frequency of CRC is about 9-12% with high male predominance 3:1. The high mortality rates associated with CRC is reflective of several factors including: the lack of apparent symptoms in the early stages and the absence of cancer prevention strategies in developing countries. Insulin-like growth factor II m-RNA-binding protein 3 (IMP3) is a member of the IMP family which plays a key role in the transfer and stabilization of mRNA, cell growth, and migration during embryogenesis. IMP3 has gained considerable interest as a cancer-associated protein. Previous studies strongly suggest that IMP3 may represent a valuable prognostic marker in human cancer. Differential gene expression in oncostreams and no-oncostreams area in mouse model glioma

Project description:Our preliminary data demonstrate that malignant gliomas in both human GBM and mouse models display regular anatomical structures, ‘oncostreams’; we interpret these as a manifestation of GBM self-organization. Oncostreams’ presence correlates with tumor malignancy and parenchymal invasion. Molecular signaling pathways that regulate oncostreams’ organization and function, however, remain unknown. We will test the hypothesis that oncostreams’ present in mouse intracranial glioma model have a characteristic transcriptome that differs from surrounding tumor, and to identify critical genes that regulate oncostreams function.
