Sarcopenia diagnosed by computed tomography predicts postoperative complications in advanced epithelial ovarian cancer.

Project description:Background & aimsSurgery combined with chemotherapy remains the mainstay of treatment for advanced epithelial ovarian cancer. It is important to evaluate the occurrence of postoperative complications before operation and to prevent them. The purpose of this study is to investigate the role of sarcopenia diagnosed by CT scans in predicting postoperative complications in patients with ovarian cancer.MethodsPatients with PDS or IDS in the First Affiliated Hospital of Wenzhou Medical University from August 2017 to August 2022 were included. Sarcopenia was identified using CT scans at the T12 level. According to the Clavien-Dindo classification, postoperative complications are considered to have occurred in grades 2 and above. Logistic regression analysis was used to identify risk factors contributing to postoperative complications. P < 0.05 was considered statistically significant.ResultsA total of 139 patients were included. Sarcopenia was observed in 24(17.27%) patients with advanced epithelial ovarian cancer. Compared to the non-sarcopenia group, the sarcopenia group had a higher incidence of postoperative complications (62.50% VS 42.61%, p = 0.032). Multivariate logistic analysis confirmed sarcopenia (OR = 3.241, p = 0.026), age over 65 years (OR = 3.296, p = 0.005), and intraoperative bleeding (OR = 1.002, p < 0.001) as independent predictors of postoperative complications in ovarian cancer.Discussion & conclusionsSarcopenia diagnosed based on CT body composition analysis may serve as a potential predictor for postoperative complications. Further research is warranted to explore preventive strategies and interventions to improve outcomes in this population.

Results: Muscle transcriptomic profiles demonstrate a prominent signature of mitochondrial dysfunction in sarcopenic patients. Conclusions: Our study supports the fundamental role of mitochondrial dysfunction in driving pathological muscle and mobility decline in the elderly.

Chronic circadian misalignment accelerates sarcopenia progression in mice

Project description:In today's 24/7 society, circadian misalignment caused by environmental and lifestyle factors is associated with various adverse health consequences, underscoring the need to understand tissue-specific pathology. A potential association of environmental circadian misalignment with sarcopenia, or accelerated loss of skeletal muscle strength and mass, is poorly documented. Here, we exposed wild-type C57BL/6J male mice to a chronic (64 weeks) jet lag (CJL) paradigm. CJL caused significant reductions in muscle strength and weight relative to untreated mice. Transcriptomic and histological analysis showed activation of TWEAK/Fn14 signaling and reduced myofiber cross-sectional area, hallmark features of sarcopenia. Furthermore, CJL mice showed increased centrally nucleated fibers, myosin heavy chain co-expressing fibers, and myogenic gene expression, suggesting that insufficient muscle regeneration likely contributes to muscle atrophy. These findings demonstrate that circadian misalignment is an independent risk factor for sarcopenia, underscoring circadian rhythms as a key regulator and actionable target for sarcopenia prevention.

Project description:Sarcopenia, the age-related loss of skeletal muscle mass and strength, is a significant cause of morbidity in the elderly and is a major burden on health care systems. Unfortunately, the underlying molecular mechanisms in sarcopenia remain poorly understood. Herein, we utilized top-down proteomics to elucidate sarcopenia-related changes in the fast- and slow-twitch skeletal muscles of aging rats with a focus on the sarcomeric proteome, which includes both myofilament and Z-disc proteins--the proteins that constitute the contractile apparatuses.

Association between elevated expression of GDF15/GFRAL and sarcopenia risk

Project description:Glial cell line-derived neurotrophic factor family receptor α-like (GFRAL) is the exclusive receptor for growth differentiation factor 15 (GDF15) and plays distinct roles in various diseases. However, its impact on sarcopenia remains poorly understood. This study, comprising participants of White British and Chinese descent, demonstrated that elevated serum levels of GDF15 and its receptor GFRAL are associated with an increased risk of sarcopenia, particularly among individuals with low physical performance. While GDF15 exerts effects across the whole population, GFRAL appears to be more detrimental in women. Complementary single-cell RNA sequencing and immunohistochemical analyses in animal models further support the involvement of the GDF15–GFRAL axis in mediating skeletal muscle damage. These findings highlight the clinical and preventive relevance of the GDF15–GFRAL axis and underscore the need for future mechanistic studies to elucidate its role in sarcopenia.
