Project description:In the social amoebae (Dictyostelia) quorum sensing system mediates aggregation of single cells into multicellular aggregates by chemotactic movement towards gradients of diffusible molecules known as acrasins. The acrasin of P. violaceum is the unusual dipeptide N-propionyl-gamma-L-glutamyl-L-ornithine-delta-lactam-ethylester, known as glorin. Phylogenetic analysis has indicated that P. violaceum is more related to the most derived group 4 dictyostelids than to the ancient group 2 polysphondylids such as P. pallidum. Nevertheless it has been reported that P. pallidum cells respond to glorin in chemotaxis assays. This has led to the assumption that glorin-based communication may be the most ancient form of intercellular communication that Dictyostelia invented to organize early steps of multicellular development. In this study we show that glorin mediates rapid changes in gene expression at the transition from vegetative growth to aggregation, apparently without pronounced cross-talk with the cyclic AMP-based communication system that coordinates post-aggregation events in this species. We describe glorin-mediated changes in gene expression in the social amoeba Polysphondylium pallidum at the transition from unicellular growth to multicellular development. Comparison of gene expression in growing cells versus cells starving for 2 or 3 hours in the presence or absence of glorin.

Project description:Understanding how genomes encode complex cellular and organismal behaviors is an outstanding challenge of modern genetics. Unlike classical screening methods, analysis of the genetic variation that occurs naturally in wild populations can enable rapid, genome-scale trait mapping with a medium-throughput experimental design. Here we describe the results of the first genome-wide association analysis (GWAS) of a microbial eukaryote, using wild isolates of the filamentous fungus Neurospora crassa. We transcriptionally profiled each of 112 individuals collected in Louisiana and used RNA-seq data to genotype each strain at thousands of genetic loci genome-wide. We used these genotypes in a mapping analysis of microbial communication. In N. crassa, germinated asexual spores (germlings) sense the presence of other germlings, grow toward them in a coordinated fashion, and fuse. We evaluated germlings of each strain for their ability to chemically sense, chemotropically seek, and undergo cell fusion. GWAS identified one gene, NCU04379 (cse-1, encoding a homolog of neuronal calcium sensor), at which inheritance was strongly associated with the efficiency of germling communication. Deletion of cse-1 significantly lowered germling communication and fusion, and two genes encoding predicted binding partners of CSE-1 were also required for the communication trait. Additionally, mining our association results for signaling and secretion genes with a potential role in germling communication, we validated six more novel determinants of germling communication, including a secreted protease and two other genes whose deletion conferred a novel phenotype of increased communication and multi-germling fusion. These results establish protein secretion as a linchpin of germling communication in N. crassa and shed light on the regulation of communication molecules in this fungus. Our study demonstrates the power of population-genetic analyses for the rapid identification of genes contributing to complex traits in microbial species. 112 mycelial mRNA profiles of 16 hour wild type (WT) Neurospora crassa strains were generated by deep sequencing, using Illumina GAIIx.

Project description:Background: Mobile phones have become ubiquitous. There are concerns about possible health hazards posed by mobile phone-based radio frequency electromagnetic field (RF-EMF). Such exposure may be recognized as external stressor at molecular level which can be identified up by shot gun proteomics approach. Objective: To study molecular effects of mobile phone-based RF-EMF exposure using shot gun proteomics. Methods: 40 individuals (30 mobile phone users and 10 non-users) were recruited based on the inclusion and exclusion criteria. Mobile phone users had mobile phone usage of more than a year, an age group of 18-65 years with no major medical or surgical illness and normal results of hemogram, liver function tests (LFT), kidney function tests (KFT) and random/fasting sugars. Non-users had no mobile phone, Wi-Fi, and microwave usage. Both mobile phone users and non-users were non-alcoholic and non-smoker. RF-EMF exposure from mobile phone and miscellaneous sources was assessed using questionnaire. Calling-time duration per day was used as a measure of mobile phone exposure and the users were classified accordingly. Five biological replicates (1 set with both genders, 2 sets each of males and females) of proteomics experiments were designed. Human plasma samples were immunodepleted of high abundant proteins, concentrated, reduced, alkylated, digested, and labelled by i-TRAQ reagents for mass spectrometry analysis. Bioinformatics study was done on differentially expressed proteins for gene ontology (GO) classification, Protein-protein interactions (PPIs) and pathway enrichment. Results: The study group was quite young (median age 24 years). Pathway enrichment studies found Dopamine mediated receptor signaling, Adrenaline and Noradrenalin synthesis, Plasminogen activating cascade, Cholecystokinin receptor (CCKR) signaling, Gonadotropin-releasing hormone (GnRH) pathway and Vitamin-D signaling and its metabolism to be implicated. Conclusion: The involvement of the differentially expressed proteins in various signalling cascades have implications physiologically albeit large sample size studies incorporating non-users need to be conducted to draw out clear inferences.
