Temperature dependent sex determination in the turtle Trachemys scripta

Project description:Sex determination is the process by which and original bipotential gonad differentiate into either a testis or ovaries. While mammals and birds determine their sex solely by genetic clues (genetic sex determination, GSD), other vertebrates like the turtle Trachemys scripta are influenced by environmental factors, like temperature (environmental sex determination, ESD). In both cases an initially bipotential gonad develops into either testes or ovaries in response to GSD or ESD cues. In order to shed light into the differences and similarities between sex determination systems we performed single-cell RNA-seq on Trachemys scripta developing gonads during the sex determination window.

Gonadal DNA methylomes of half-smooth tongue sole Cynoglossus semilaevis

Project description:Environmental sex determination (ESD) occurs in divergent, phylogenetically unrelated taxa, and in some species co-occurs with genetic sex determination (GSD) mechanisms. Although epigenetic regulation in response to environmental effects has long been proposed to be associated with ESD, a systemic analysis on epigenetic regulation of ESD is still lacking. Using half-smooth tongue sole (Cynoglossus semilaevis) as a model – a marine fish which has both ZW chromosomal GSD and temperature-dependent ESD – we investigated the role of DNA methylation in transition from GSD to ESD by comparing gonadal DNA methylomes of parental females, parental pseudo-males, F1 females, F1 pseudo-males and normal males. To assess the gonadal DNA methylome patterns across different sexual types of tongue sole, we carried out BS-seq on bisulfite converted DNA extracted from adult gonads of parental females, parental pseudo-males, and F1 pseudo-males and females from a cross between a parental pseudo-male and a normal female. We also sampled normal male individuals as a control for the normal male DNA methylation pattern. For each of the five samples, two biological replicates were utilized, with each replicate being pooled by five fish. The phenotype and genotype of each selected fish was identified by the histological analysis and PCR validation using the W chromosome specific marker. DNA were isolated from five pooled gonads of the same replicate, then 5 ?g DNA was used to do the bisulfite conversion and BS-seq. The bisulfite conversion of sample DNA was carried out using a modified NH4HSO3-based protocol (Hayatsu et al. 2006). The paired-end library construction and sequencing were carried out using Illumina HiSeq 2000, according to the manufacturer’s instructions (Illumina). We also mixed 25 ng cl857 Sam7 Lambda DNA in each sample to use as conversion quality control for each library.

Mutation of medaka cyp27b1 promotes sex reversal of female to male

Project description:The mechanisms of sex determination in fish includes genetic sex determination (GSD) and environmental sex determination (ESD), and sex reversal is obtained in some fishes, which caused by the effect of gene expression level and environmental factors. In previous study, we constructed a sex reversal medaka mutant and identified the different expression level of cyp27b1 gene compared with wild-type. However, whether cyp27b1 affects the sex determination and gonad differentiation has not been reported. In this study, we constructed cyp27b1 heterozygote mutant by using CRISPR/Cas9 system in Japanese medaka. Phenotype analysis showed that sex reversal was occurred in the heterozygote mutant, in which the first and second sexual characteristics of male mutant (XX) was not significantly different from that of male wild-type (XY). In addition, knockout cyp27b1 gene leads to no effect on testis development. and the same phenotype was observed in the generation male mutants.

Exon-array profiling of peripheral blood cells in the first hours of acute coronary syndrome patients

Project description:Identifying novel candidate biomarker gene differentially expressed in the peripheral blood cells of patients with early stage acute myocardial infarction using microarray as a high throughput screening technology. In this study, we used exon array profiling to investigate biomarker gene analyzing 49 patients samples (six time points) and 6 control samples. Whole-blood cell drawn from an antecubital vein at six time points following STEMI diagnosis. The first time (T0) was on patient admission to the Emergency Unit prior to receiving any medication. The second time (T2) blood collection occurred after successful mechanical reperfusion and stenting of the infarct artery within 2 h after T0. Collections for the subsequent four time points were at 12 h (T12), 24 h (T24), 36 h (T36) and 48 h (T48). For the control group, peripheral blood was collected at one time point.

Metabolomics reveals an involvement of pantothenate for male production responding to the short-day stimulus in the water flea, Daphnia pulex

Project description:Under favorable conditions, the micro-crustacean Daphnia pulex produces female offspring by parthenogenesis, whereas in response to various unfavorable external cues, they produce male offspring to induce sexual reproduction (environmental sex determination: ESD). We recently established a suitable experimental system for ESD studies using D. pulex WTN6 strain, in which the sex of the offspring can be regulated simply by alterations in day-length; long-day and short-day conditions can induce female and male offspring respectively. Taking advantage of this system, we have already demonstrated that methyl farnesoate (MF) synthesis is necessary for male offspring production. Further, we identified ionotropic glutamate receptors as an upstream regulator of MF signaling by transcriptome analysis. Despite these findings, the molecular mechanisms associated with MF signaling have not yet been well elucidated in D. pulex. To more comprehensively understand the molecular mechanisms of ESD, we analyzed the whole metabolic profiles of mother daphniids reared under long-day (female-producing) and short-day (male-producing) conditions using mass spectrometry based metabolomics. We discovered that pantothenate (vitamin B5), a known precursor to coenzyme A, was significantly accumulated in response to the short-day condition. To confirm the role of pantothenate in D. pulex ESD, this metabolite was administered to mother daphniids (under long-day conditions) resulting in a significantly increased proportion of male offspring producing mothers. This study not only provides several candidates for further investigations into the molecular changes involved in male offspring production, but also novel insights into the metabolic mechanisms of the ESD system in D. pulex.

Impact of triphenyl phosphate on sperm quality reduction: evidence from human, mice and cells

Collectively, these data indicated that TPhP exposure could reduce male reproductive quality, which provided insights into the adverse effects of TPhP on human reproductive health and also provided a basis for the formulation of policy on TPhP usage by relevant government agencies.

Transcription profiling by array of mouse embyronic stem cells and ESD-EpiSCs

Project description:The comparison of mouse ESCs and ESD-EpiSCs was done using three biological replicates for each cell type. ESC samples (ESR1 p10, p20, ESR1/E p15) and ESD-EpiSC samples (EpiSC-7 p20, EpiSC-5 p22, and EpiSC-7 p25) were used for microarray analysis. Total RNA was extracted using Trizol and labeled and hybridized to Agilent Whole Mouse Genome Oligo 4X44K Microarrays (one-color platform) according to the manufacturers protocols.
