We extracted total mRNA from parental cells and resistant cell lines. We compared their expression by carried out Affymetrix Huex 1.0 ST expression array. Two paired parental/derived cell lines were screened as per the summary above. BRAF inhibitory resistant cells had gene expression compared to the parental cell lines.

Project description:BRAF inhibitors are highly effective therapies for patients with BRAF V600 mutated metastatic melanoma. Patients who receive BRAF inhibitors develop a variety of hyper-proliferative skin conditions, whose pathogenic basis is the paradoxical activation of the mitogen-activated protein kinase (MAPK) pathway in BRAF wild-type cells. Most of these hyper-proliferative skin changes improve when a MEK inhibitor is co-administered, as a MEK inhibitor blocks paradoxical MAPK activation. We tested whether we could take advantage of the mechanistic understanding of the skin hyper-proliferative side effects of BRAF inhibitors to accelerate skin wound healing by inducing paradoxical MAPK activation. Here we show that the BRAF inhibitor vemurafenib accelerates human keratinocyte proliferation and migration by increasing ERK phosphorylation and cell cycle progression. Topical treatment with vemurafenib in two wound-healing models in mice accelerated cutaneous wound healing and improved the tensile strength of healing wounds through paradoxical MAPK activation; addition of a MEK inhibitor reversed the benefit of vemurafenib-accelerated wound healing. The same dosing regimen of topical BRAF inhibitor did not increase the incidence of cutaneous squamous cell carcinomas in mice even after the application of a carcinogen. Therefore, topical BRAF inhibitors may have clinical applications in accelerating the healing of skin wounds. Project description:The Tasman Sea off southeast Australia exhibited its longest and most intense marine heatwave ever recorded in 2015/16. Here we report on several inter-related aspects of this event: observed characteristics, physical drivers, ecological impacts and the role of climate change. This marine heatwave lasted for 251 days reaching a maximum intensity of 2.9 °C above climatology. The anomalous warming is dominated by anomalous convergence of heat linked to the southward flowing East Australian Current. Ecosystem impacts range from new disease outbreaks in farmed shellfish, mortality of wild molluscs and out-of-range species observations. Global climate models indicate it is very likely to be that the occurrence of an extreme warming event of this duration or intensity in this region is respectively ≥330 times and ≥6.8 times as likely to be due to the influence of anthropogenic climate change. Climate projections indicate that event likelihoods will increase in the future, due to increasing anthropogenic influences.

Project description:The extreme El Niño (EN) events in 1997/98 and 1982/83, referred to as super EN, exerted remarkable global influence. A super EN was anticipated on the way in early 2014 but failed to materialize toward the end of 2014. Whilst the scientific community was still puzzling about the cause of the aborted EN event in 2014, the remnants of the decaying warming in late 2014 unexpectedly reignited since February 2015 and grew into a super EN by the end of 2015. Understanding the onset mechanism of the 2015 EN event and its differences from past super EN events is crucial for improving EN prediction in a changing climate. Our observational analyses and modeling studies demonstrate that the principal difference between the 2015 EN and the past super ENs lies in exceptionally strong and consecutive occurrence of westerly wind burst events that turned around unfavorable ocean thermocline conditions in tropical western Pacific in early 2015, reigniting rapidly the surface warming in the eastern Pacific. By August the sea surface temperature anomalies reached a critical amplitude similar to that of the past super ENs; positive atmosphere-ocean feedbacks further amplify this warm episode into a super EN by the end of 2015.
