Temporal Response to seasonal and pandemic H1N1 infection in human DCs- Donor 1

Project description:An 8 hours timecourse was performed with human DCs infected either with A/California/7/2009 and A/Brevig Mission/1/1918 (pandemic) or A/New Caledonia/20/99 and A/Texas/36/91 seosonal. Human monocyte derived dendritic cells (DCs) were infected either with chicken egg grown A/California/7/2009, A/Brevig Mission/1/1918, A/New Caledonia/20/99or A/Texas/36/91. Samples were taken and fixed in RNA stabilizing regaent at 2, 2.7, 3.3, 4, 5, 6, 7 and 8 hours. Cells exposed to chicken egg alantoic fluid served as a control. Infections with A/California/7/2009, A/New Caledonia/20/99 or A/Texas/36/91 were carried out at a BSL2 enviroment. Infections with A/Brevig Mission/1/1918 were carried out in a BSL3 enviroment in parralel to the BSL2 infections with cells from the same donor and cell preparation.

Project description:The purpose of this experiment was to identify the genes with increased expression during sporulation in wild-type P. pastoris, and during sporulation in cells with Ndt80 deleted. Cells were grown unti log-phase in YPD or in sporulation media (0.5% sodium acetate, 1% potassium chloride, 1% glucose, 2% agar plates ) for 30 hours. Project description:We studied microRNA expression levels from serum of postmenopausal women with osteoporosis by investigating the anti-osteoporotic treatment denosumab for a period of 2 years in a longitudinal study. Serum RNA was extracted and subject to small RNA sequencing on an Illumina HiSeqV4 SR50 using 50bp, single end reads. After mapping against GRCh38.p12 provided by Ensembl and miRBase v22.1 differential expression analysis was undertaken with edgeRv3.28 using a quasi-likelihood negative binomial generalized log-linear model. We identified a panel of altered small non-coding RNAs between 3 different time points for all patients.

Peptide Ancestry Informative Markers in Uterine Neoplasms from Women of European, African and Asian Ancestry

Project description:Characterization of ancestry-linked peptide variants in disease-relevant patient tissues represents a foundational step to connect patient ancestry with molecular disease pathogenesis. Nonsynonymous single nucleotide polymorphisms (SNPs) encoding missense substitutions within tryptic peptides exhibiting high allele frequencies in European, African, and East Asian populations, termed peptide ancestry informative markers (pAIMs), were prioritized from 1000 genomes. In silico analysis shows that as few as 20 pAIMs can determine ancestry proportions similarly to >260K SNPs (R2=0.9905). Multiplexed proteomic analysis of >100 human endometrial cancer cell lines and uterine leiomyoma (ULM) tissues combined resulted in the quantitation of 62 pAIMs that correlate with self-described race and genotype-confirmed patient ancestry. Candidates include a D451E substitution in GC vitamin D-binding protein previously associated with altered vitamin D levels in African and European populations. These efforts describe a generalized set of markers for proteoancestry assessment that will further support studies investigating the impact of ancestry on the human proteome and how this relates to the pathogenesis of uterine neoplasms.
