Small RNA-Seq of plasma-derived exosomes from Acute Respiratory Distress Syndrome patients and healthy subjects

Project description:Despite a significant progress in the treatment of Acute Respiratory Distress Syndrome (ARDS), our ability to identify early patients and predict outcome remains limited. In this study, we aimed to characterize small RNA content of plasma exosomes from ARDS patients in order to identify potential diagnostic biomarkers of the disease. For the first time, we profiled miRNA expression levels in plasma-derived exosomes from ARDS patients (n=8) compared to healthy subjects (n=10) by small RNA-seq. It allowed us to identify 12 exosomal miRNAs differentially expressed in ARDS context (padj<0.05).

Small RNA sequencing of cow's peripheral plasma exosomes

Project description:OBJECTIVE: To reveal the miRNA expression profile of placental exosomes by high-throughput sequencing and screen key biomarkers in different pregnancy period to provide new research ideas for the establishment and maintenance of pregnancy in cows and other ruminants.It can also provide reference for other pregnancy diseases. METHOD: Exosmal miRNAs were isolated from non-pregnant cows（n=3,Gestation Day(G.D.) 0）,early pregnant cows（n=3,G.D.60）,Middile pregnant cows（n=3,G.D.150）,Late pregnant cows（n=3,G.D.240）, small RNA profiles were generated by deep sequencing, in triplicate, using Illumina Hiseq SE50. RESULTS: During pregnancy, there was a high quantity of placental exosomes in the peripheral blood of cows, which selectively loaded some miRNAs. The abundance and species of miRNAs various from different stages of pregnancy, suggesting that exosomal miRNAs is involved in the regulation mechanism of pregnancy.

RNA-Seq analysis of LYN deficient HS-5 stroma cells cocultured with primary human CLL cells

Project description:LYN kinase is a tyrosine kinase, that regulates cellular homeostasis in a context-specific manner. Our group could show, that its expression in the leukemic microenvironment of chronic lymphocytic leukemia contributes to disease progression (Nguyen PH et al.; Cancer Cell; 2016). To analyze the effect of LYN kinase on the leukemia supportive phenotype of the bone marrow stromal cell line HS-5, we generated single cell clones of LYN deficient stroma cells. These cells were analyzed in a Multi-Omic approach, including ARNA-Seq of stromal cells after 72h of coculture with primary human chronic lymphocytic leukaemia (CLL) samples.
