ABSTRACT: Toxoplasmosis is a zoonotic disease caused by a ubiquitous protozoan parasite called Toxoplasma gondii, which can infect all mammal and bird species throughout the world. seroprevalence varies widely between countries. Studies have estimated that between 7-34% of people in the UK have been infected with T. gondii. The vast majority of these people will not have noticed any symptoms, however about 10% of people develop a mild to moderate self limiting flu-like illness. Following the acute active stage of the infection the parasite persists in the body in the form of cysts, particularly in heart and skeletal muscle and nervous system tissues, for many years, and usually for life. In immunocompetent persons these cysts do not pose a health risk. We will use RNA-seq to quantify the transcriptional response of macrophages to T gondii infection. Transcriptome sequencing of Strongyloides ratti

Project description:Strongyloides ratti is a common gastro-intestinal parasite of the rat. The adult parasites are female only, about 2mm long and live in the mucosa of the small intestine. These parasites produce eggs that pass out of the host in its faeces. In the environment infective larval stages develop either directly or after a facultative sexual free-living adult generation. Infective larvae infect hosts by skin penetration.S. ratti is the laboratory analogue of the parasite of humans, S. stercoralis. S. stercoralis is a wide-spread parasite of humans, occurring principally in the tropics and sub-tropics: some 100-200 million people are infected worldwide. Infection of immunosuppressed individuals can result in disseminated strongyloidiasis, in which worms occur throughout the body. This can be fatal unless anti-Strongyloides therapy is given. Other species of Strongyloides parasitise a wide range of vertebrates. As part of the Strongyloides ratti genome project we are profiling the transcriptome of the parasite across its life cycle using RNA-Seq.. This data is part of a pre-publication release. Transcriptomics of the immune response to Schistosomiasis

Project description:This data is part of a pre-publication release. For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/ Schistosomiasis is a severe debilitating illness that represents one of the most important parasitic diseases in tropical areas. It affects over 200 million people across 74 countries. The complete genome sequence of Schistosoma mansoni was published in 2009, but immune response against this parasite is not well characterized. This study is to investigate the overall immune response, antibody and TCR repertoire of immunologically nave mice, mice infected with S.mansoni and mice challenged with a secondary schistosome infection.RNA-seq data will be used to characterise the systemic immune response of mouse to Schistosoma infection, including both whole-transcriptome sequencing for a global picture of the immune response, and sequencing the lymphocyte antigen receptor genes to get a detailed picture of the adaptive immune response.

Project description:We have shown previously (Reid & Berriman, NAR, 2012) that by simultaneously examining host and parasite gene expression over the course of infection we can determine pairs of genes involved in host-parasite interaction. Here we are producing a high quality dataset which will specifically allow us to exploit this finding to identify genes involved in malaria host-parasite interaction. Transcriptional response of the mouse intestinal mucosa to infection by the parasitic nematode Trichuris muris

Project description:This data is part of a pre-publication release. For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/ The human-infective whipworm Trichuris trichiura is estimated to infect up to a billion people and is responsible for considerable morbidity, especially in children of developing countries. The closely related species T. muris is a naturally occurring nematode parasite of mice that serves as a remarkably tractable model system for dissecting immune responses and host-parasite relationships. Such studies are of relevance beyond parasitology as helminths have arguably had a significant impact on the evolution of the mammalian immune system. Both Trichuris species reside in the caecum and colon of the host where they burrow their front end for feeding into the intestinal mucosa, thereby breaching the mucus barrier and allowing access of the microflora directly to the epithelium. The interplay of intestinal helminths, the bacterial microflora and the host immune system is currently a research focus in various laboratories (Bancroft et al 2012). This study will study the transcriptional responses of the intestinal mucosa (caecum) from infected and uninfected mice.
