Project description:The study includes methylC-capture sequencing (MCC-Seq) on 94 sperm DNA samples derived from both fertile and infertile individuals who were recruited from the Men’s Health Clinic at the Royal Victoria Hospital, Montreal, Quebec. All the data were generated with 100bp paired-end reads using the Illumina NovaSeq 6000 systems.

Project description:Male factor infertility is increasing and recognized as playing a key role in reproductive health and disease. The current primary diagnostic approach is to assess sperm quality associated with reduced sperm number and motility, which has been historically of limited success in separating fertile from infertile males. The current study was designed to develop a molecular analysis to identify male infertility using genome wide alterations in sperm DNA methylation. A signature of differential DNA methylation regions (DMRs) was identified to be associated with male infertility patients. A promising therapeutic treatment of male infertility is the use of follicle stimulating hormone (FSH) analogs which improved sperm numbers and motility in a sub-population of infertility patients. The current study also identified genome-wide DMRs that were associated with the patients that were responsive to FSH therapy versus those that were non-responsive.

Project description:Although piwi-interacting RNAs (piRNAs) play pivotal roles in spermatogenesis, little is known about piRNAs in the seminal plasma of infertile males. In this study, we systematically investigated the profiles of seminal plasma piRNAs in infertile males to identify piRNAs that are altered during infertility and evaluate their diagnostic value. Seminal plasma samples were obtained from 211 infertile patients (asthenozoospermia and azoospermia) and 91 fertile controls. High-throughput sequencing technology was employed to screen piRNA profiles in seminal plasma samples pooled from healthy controls and infertile patients. The results identified 61 markedly altered piRNAs in the infertile patient groups compared with the control group. Next, a quantitative RT-PCR assay was conducted in the training and validation sets to measure and confirm the concentrations of altered piRNAs. The results identified a panel of 5 piRNAs that were significantly decreased in the seminal plasma of infertile patients compared with healthy controls. The areas under the ROC curves for these piRNAs ranged from 0.796 to 0.996, suggesting that the diagnostic potential of these 5 piRNAs to distinguish asthenozoospermic and azoospermic individuals from healthy controls was high. In summary, this study identifies a panel of piRNAs that can accurately distinguish fertile from infertile males. This finding may provide pathophysiological clues that are involved in the development of infertility.

Project description:Alterations in the presence of sperm RNAs have been identified using microarrays in teratozoospermic (abnormal morphology) or other types of infertile patients. However, so far no studies had been reported on the sperm RNA content using microarrays in asthenozoospermic patients (low motility). We started the present project to with the goal to characterize the RNA expression in asthenozoospermic infertile patients as compared to normozoospermic fertile controls. We selected four normal fertile donors and four severe asthenozoospermic infertile patients. Equal amounts of RNA were extracted from the sperm samples, subjected to different quality controls and hybridized to the Affymetrix U133 Plus version 2 arrays.

Project description:The aim of this study was to achieve a higher resolution of the human sperm proteome for better understanding of the molecular causes of male infertility. For this purpose, we examined the proteomic profiles of sperm from 76 men for proteins with deregulated abundances. Infertile men had abnormal semen parameters and were involuntarily childless. Men in the cohort with normal fertility had normozoospermia and had fathered children without medical assistance. Mass spectrometry analysis led to an in-depth reconstruction of the human sperm proteome. The corresponding genes were mainly involved in cellular motility, response to stimuli, adhesion, and reproduction. We also observed increasing numbers of at least three-fold differentially abundant sperm proteins from oligozoospermia and oligoasthenozoospermia to oligoasthenoteratozoospermia, compared with normozoospermia. Primarily, flagellar assembly and sperm motility, sperm-egg interaction, and male gametogenesis were affected. This suggests that sperm of infertile men carry signatures of impaired sperm production and are functionally impaired. Not least, we present new male infertility markers in addition to established ones.

Project description:Array-based DNA methylation profiling in peripheral blood leukocytes of 30 infertile men with impaired spermatogenesis as compared to 10 fertile men using the Illumina Infinium HumanMethylation 450k Bead Chip reveald 471 CpG sites (287 genes) to be differentially methylated between both groups. These CpG loci were significantly enriched for the gene ontology functions MHC class II receptor activity and piRNA binding. The latter was associated with two methylation-sensitive SNPs in the genes PIWIL1 and PIWIL2, respectivly, which showed significant allele distribution skewing in the infertile cohort. 445/471 differentially methylated CpGs were associated with SNPs, but 26 (15 genes) were not genomically templated and included the ENO1, MTA2, BRSK2 and LBX2 genes previously associated with fertility and spermatogenesis. The study identifies surrogate DNA methylation markers for idiopathic infertility in peripheral blood and suggests allele-specific DNA methylation differences at regulatory sites of genes involved in piRNA regulation to be associated with disturbed spermatogenesis. Bisulfite converted DNA of peripheral blood leukocytes from 30 infertile men and 10 fertile men as controls were hybridized to the Illumina Infinium HumanMethylation 450k Bead Chip.

Project description:The global trend on (male)infertility is concerning and the unidentifiable causes in half of the cases demands a better understanding of the molecular, biochemical and metabolic mechanisms as well as the identification of external and internal factors operating behind it, that might help to explain this apparently unjustified infertility. This can only be achieved through a comprehensive analysis of the infertile men, in which external (lifestyle, occupational and environmental factors) and internal factors (psychological distress) should also be evaluated and considered, but also assessing sperm function beyond the routine seminal analysis. In this prospective cohort study, 79 sperm samples, from men who were male partners in couples seeking for infertility treatment, were collected at the Reproductive Medicine Unit (CHUC) from July 2018 to July 2022 and analyzed by SWATH-MS. Based on couples’ clinical data, seminal/hormonal analysis, and strict exclusion criteria, samples were categorized in the different groups, namely control (CTRL; 50 individuals), idiopathic infertile (ID; 19 patients) and unexplained infertile (UMI; 10 patients) men. In general, ID patients presented the worst sperm functional profile, while the UMI patients were observed to be similar to controls. These differences were also observed at the proteomics levels, which revealed 145 differentially expressed proteins (DEP) between the three groups, with more than 120 proteins being altered between ID and the other groups, and only 14 proteins were considered altered between CTRL and UMI.

Project description:Male factor infertility is involved in almost half of all infertile couples. Lack of the ejaculated sperm owing to testicular malfunction has been reported in 6-10% of infertile men, a condition named nonobstructive azoospermia (NOA). In this study, we investigated untargeted metabolomic profiling of the seminal plasma in NOA men using gas chromatography-mass spectrometry and advance chemometrics. In this regard, the seminal plasma fluids of 11 NOA men with TESE-negative, nine NOA men with TESE-positive and 10 fertile healthy men (as a control group) were collected. Quadratic discriminate analysis (QDA) technique was implemented on total ion chromatograms (TICs) for identification of discriminatory retention times. We developed multivariate classification models using the QDA technique. Our results revealed that the developed QDA models could predict the classes of samples using their TIC data. The receiver operating characteristic curves for these models were >0.88. After recognition of discriminatory retention time's asymmetric penalized least square, evolving factor analysis, correlation optimized warping and alternating least squares strategies were applied for preprocessing and deconvolution of the overlapped chromatographic peaks. We could identify 36 discriminatory metabolites. These metabolites may be considered discriminatory biomarkers for different groups in NOA.
