Project description:Donor pancreata were obtained from the Beta Cell Bank of the JDRF Centre for Beta Cell Therapy in Diabetes (Brussels, Belgium), from Pancreatic Islet Processing (ECIT center) of Diabetes Research Institute at the IRCCS San Raffaele Scientific Institute (Milan, Italy) and from the DRWF Human Islet Isolation Facility (Oxford, England). Full written consent for use of donor material for research was obtained according to Belgian, Italian and English laws. This project was approved by the Medical Ethical Committee of all institutions. Cells were cultured in 3D suspension culture for four days in Advanced RPMI supplemented with 5% FBS.

Project description:Reversing gene expression signatures in relapsed patient may restore chemosensitivity. We demonstrate that the histone deacetylase inhibitor vorinostat not only reprograms the aberrant gene expression profile of relapsed blasts but is synergistic when applied prior to chemotherapy in primary patient samples and leukemia cell lines Primary patient samples were collected from patients treated at the New York University Medical Center and from the Childrenâs Oncology Group (COG) cell bank,RNA was isolated from three primary patient samples and three B-lineage leukemia cell lines with or without treatment with vorinostat for 24 hours.

Inferior Temporal Gyrus in Human Ageing: a Quantitative Protein Profiling Work based on Chinese Human Brain Bank

Project description:The temporal lobe is the cerebral cortex with critical function. The superior and middle gyrus of temporal lobe have been well studied, however, present perceptions on inferior temporal gyrus remains limited. The understanding of age-related protein profile change in human inferior temporal gyrus has not yet been well established. This 3-plex TMT labeled proteomic study is performed based on the human brain bank at the Chinese Academy of Medical Sciences & Peking Union Medical College. Age distribution of the donors ranges from 22 to 90 years old, and were assigned to three age groups: 20-50, 50-70, and 70-90 years of death age. In this ageing cohort, no neurodegenerative disorders or major stroke events were identified via standard neuropathological classification. Proteomics and bioinformatics strategies were applied to identify the perturbations of protein expression and associated pathways. Among all the ITG samples, 3113 proteins were isolated, with 37 proteins upregulated and 21 proteins downregulated.

RNA profiling of matched pair samples of primary breast cancer vs. lung and pleura metastasis

Project description:Patient samples were recruited from a series of female patients with metastatic primary breast cancer (PBC) and biopsy-confirmed breast cancer lung or pleural metastasis (BCLPM) at the Department of Thoracic Surgery, Thoraxklinik, Univ. of Heidelberg, Germany. All patients were included who had received lung or pleural metastasis biopsies from 2003 to 2014 and had a prior history of PBC at the Department of Gynaecology and Obstetrics, Univ. Hospital. Formalin-fixed and paraffin-embedded tissue samples were provided by the Tissue Bank at the National Center for Tumor Diseases (NCT, Heidelberg, Germany) in accordance with the regulations of the Tissue Bank and approval of the Ethics Committee at the Medical Faculty of the Univ. of Heidelberg, approval no. S-716/2018.
